Effect of different tissues in Pyrus L. for genome DNA extract on RAPD.

RAPD technique was applied on study of germplasm genetic diversity.In order to provide RAPD information to application of Pyrus germplasmic resources,Yali pear and Betulaefolia were tested.Genome DNA of the pears was extracted from different tissues or different phases of the same tissue,such as new leaves,old leaves and phloem by CTAB method,and checked by RAPD amplification.The results showed that OD260/OD280 of the DNA both from leaves and phloem were among 1.6-2.0,and all of them could be amplified successfully by RAPD and with the same amplification effect.
