The invention belongs to an enzyme-linked immunosorbent assay method. The method comprises the following steps: bonding glucose oxidase and a monoclonal antibody to form an enzyme-labeled antibody compound; after specifically bonding a detected antigen and the enzyme-labeled antibody compound, adding an enzyme reaction substrate, namely glucose, and enabling the enzyme reaction substrate to react with the glucose oxidase to generate nascent-state active oxygen; enabling the nascent-state active oxygen and a color developing agent to be subjected to color developing reaction and amplifying a signal under the catalytic effect of a catalytic amount of horseradish peroxidase, so as to detect an antigen to be detected. The immunosorbent assay method provided by the invention is a sandwich-process enzyme-linked immunosorbent assay method and is a simple, convenient and rapid immunosorbent assay method capable of realizing high-throughput detection; a detection result is accurate and reliable, has significant meanings in the aspects of production cost, transportation and storage properties, safety in utilization, environment friendliness and the like, and has a very high application value.
