Study on factors influencing the repeatability of HLA-DRB genotyping with PCR-SSCP

PCR-SSCP (PCR-single strand conformation polymorphism)technique was applied in the HLA-DRB typing and factors influencing the repeatability of experiments were studied. The results showed that the major factors which affect HLA-DRB genotyping included the temperature of electrophoresis, pretreatment of samples, the gel focusing time and the silverstaining procedure, ect. Strict control over the experimental conditions could guarantee the repeatability of the typing of HLA-DRB with PCR-SSCP.
