Rapid purification of high quality DNA from large whole blood samples using a new rapid protocol

The efficiency and quality of DNA from blood samples is one of the important issues for genetic studies on human diseases. In this study a rapid purification method (PUREGENE EP) is described and compared with the standard protocol to purify DNA in parallel from replicate 3 mL whole blood samples from 16 individuals. A significant timesaving (30 % reduction) was observed (62.0 min vs. 92.5 min for the same 16 samples) with no significant difference in yield (24.86 ± 10.21 μg/mL vs. 23.51 ± 12.10 μg/mL). In addition, the quality of the DNA was similar for both protocols either according to the A 260 /A 280 ratio (1.83±0.02 vs. 1.82±0.03) or the agarose gel electrophoresis. In summary, the PUREGENE EP protocol provides effective and efficient purification of high quality DNA while reducing the time required for purification.
