The Effect of Sodium Orthovanadate on the Expressionof Caspase-3 in Esophageal Cancer Cell Line EC109

Objective : To study the effects of sodium orthovanadate on the activation of Caspase-3 in esophageal cancer cell line EC109.Methods: EC109 esophageal cancer cells grown in cell culture were treated with sodium orthovanadate for 1 hour at the dose of 0,0.1,1.0,5.0,10.0,50.0,100.0,200.0 μmol/L respectively.Whole cell extracts were harvested and subjected to Western blotting technique to examine the activation of Caspase-3.Results: There are no statistically significant changes for the activation of Caspase-3 after sodium orthovanadate treatment at the dose of 0.1,1.0,5.0,10.0 μmol/L.However,the protein level of Caspase-3 markedly increases after sodium orthovanadate treatment at the dose of 50.0,100.0,200.0 μmol/L.Conclusion: Sodium orthovanadate at high dose may trigger the activation of caspase-3 in EC109 cells.
