Selecting Cell Lines for SNP Human Identification Assay Development

SNP technology for human identification application is still under development. One challenge for the SNP assay development is to have unlimited amount of DNA with consistent genotypes which represent different population. Using DNA extracted from individuals is not a good choice because of limited amount and changing genotypes. DNA extracted from human cell lines of different populations can provide unlimited amount of DNA with consistent genotypes. But cell line chromosome often contains aneuploidy and other chromosome alteration, such as deletion and duplication, after long time passages in tissue culture. These chromosome changes will affect SNP assay results. So the cell lines need to be carefully selected for each SNP marker. In this study we will provide experiment data and set up criteria for selecting the cell line for SNP applications.
