Expression and significance of Tiam1,Fascin-1 and HSPB1 in human colorectal cancer

2008
Objective To investigate the expression of T lymphoma invasion and metastasis gene 1(Tiam1),Fascin-1 and heat shock 27kDa protein 1(HSPB1) in human colorectal cancer(CRC) and their significance. Methods A tissue microarray(TMA) containing 100 informative cases of CRC was constructed.Immunohistochemistry SP method was used to detect the expression of Tiam1,Fascin-1 and HSPB1 in TMA of CRC respectively. Results Ninety-six percents of dots on the TMA were observable morphologically with clear background and vivid contrast after immunohistochemistry. The positive rates of Tiam1,Fascin-1 and HSPB1 were 74%,51% and 68% respectively in TMA of colorectal cancer.Expression of Tiam1,Fascin-1 and HSPB1 protein in CRC with metastasis was higher than those in CRC without metastasis. This finding indicated that Tiam1,Fascin-1 and HSPB1 were positively correlated with metastasis of CRC.We next analyzed the correlation among Tiam1,Fascin-1 and HSPB1.Tiam1 expression was positively correlated with Fascin-1(r=0.678,P0.01) and HSPB1(r=0.650,P0.01) in colorectal cancer. Conclusion Tiam1,Fascin-1 and HSPB1 are related to metastasis of CRC.Overexpression of Fascin-1 and HSPB1 may relate to Tiam1 regulation.
