A Novel Anticancer Stem Cell Compound Derived from Pleuromutilin Induced Necroptosis of Melanoma Cells

Necroptosis has been recently confirmed as a non-apoptotic form of programmed cell death. Discovery of novel chemical entities, capable of inducing necroptosis of cancer cells, is likely to act as an alternative strategy for dealing with drug resistance clinically. In this study, the identification of a novel Pleuromutilin derivative (compound 38) is presented, capable of significantly increasing the cellular level of ROS and inducing melanoma cancer cell death via necroptosis. Furthermore, compound 38 noticeably ablated various cancer stem cells and inhibited the growth of melanoma cancer cells both in vitro and in vivo. Moreover, 38 exhibited low toxicity in animal models and excellent PK properties, which is currently being verified as a potential anticancer drug candidate.

The chemokine CXCL13 in lung cancers associated with environmental polycyclic aromatic hydrocarbons pollution

More than 90% of lung cancers are caused by cigarette smoke and air pollution, with polycyclic aromatic hydrocarbons (PAHs) as key carcinogens. In Xuanwei City of Yunnan Province, the lung cancer incidence is among the highest in China, attributed to smoky coal combustion-generated PAH pollution. Here, we screened for abnormal inflammatory factors in non-small cell lung cancers (NSCLCs) from Xuanwei and control regions (CR) where smoky coal was not used, and found that a chemokine CXCL13 was overexpressed in 63/70 (90%) of Xuanwei NSCLCs and 44/71 (62%) of smoker and 27/60 (45%) of non-smoker CR patients. CXCL13 overexpression was associated with the region Xuanwei and cigarette smoke. The key carcinogen benzo(a)pyrene (BaP) induced CXCL13 production in lung epithelial cells and in mice prior to development of detectable lung cancer. Deficiency in Cxcl13 or its receptor, Cxcr5, significantly attenuated BaP-induced lung cancer in mice, demonstrating CXCL13’s critical role in PAH-induced lung carcinogenesis.

Factors controlling the CO intercalation of h-BN overlayers on Ru(0001)

The space between a two-dimensional (2D) material overlayer and a metal surface can be regarded as a nanoreactor, in which molecule adsorption and surface reaction may occur. In this work, we present CO intercalation under a hexagonal boron nitride (h-BN) overlayer on Ru(0001) at room temperature, observed using X-ray photoelectron spectroscopy, ultraviolet photoelectron spectroscopy, and scanning tunneling microscopy. Critical factors influencing the interfacial process have been investigated, including CO partial pressure, h-BN coverage, and oxygen pre-adsorption on the Ru surface. It has been identified that CO adsorption on the bare Ru surface region plays an important role in CO intercalation. Comparative studies of CO intercalation at h-BN/Ru(0001) and graphene/Ru(0001) interfaces indicate that CO starts to intercalate h-BN overlayers more easily than graphene. Temperature-programmed CO desorption experiments from h-BN/CO/Ru(0001) and graphene/CO/Ru(0001) surfaces reveal a similar confinement effect of the 2D cover on CO adsorption, which results in a more abrupt and quick CO desorption in comparison with the CO/Ru(0001) surface.

Two-Dimensional Lanthanide Metal–Organic Framework Heterostructures for Noninvasively Photoresponsive High-Security Photonic Barcodes

Stimuli-responsive micro/nanoscale photonic barcodes show great capacity for encryption and anticounterfeiting technologies due to multiple authentications, yet their application is commonly restricted by invasive stimuli. Herein, we report noninvasive light-stimulated high-security photonic barcodes based on spatially assembled photoresponsive two-dimensional (2D) 1,3,5-benzenetribenzoate (BTB)@Ln-MOF host-guest heterostructures. The photoluminescence (PL) spectra information on BTB@Ln-MOF heterostructures could be precisely controlled by the different wavelengths of ultraviolet (UV) light trigger. By using the PL properties and 2D heterostructures as cryptographic primitives, spatially resolved smart photonic barcodes based on both spectral and graphical coding are realized in BTB@Ln-MOF host-guest materials. These results will pave an avenue for the development of smart stimuli-responsive photonic barcodes for anticounterfeiting applications.

Graphene as a surfactant for metal growth on solid surfaces: Fe on graphene/SiC(0001)

X-ray photoelectron spectroscopic and scanning tunneling microscopic results demonstrate that annealing of Fe/carbon-rich 6H-SiC(0001) surface between 650 and 750 °C leads to Fe intercalation under the surface carbon layer. Accompanied with the metal intercalation, the carbon nanomesh surface was transformed into a graphene surface. Moreover, the formed graphene layers always float out to the topmost surface even after deposition of more than 10 monolayer Fe, acting as a surfactant. Using graphene as the surfactant may not only promote the 2D growth but also can improve the film performance considering that graphene is stable and robust.
