Aromatase Gene Polymorphism in Sheep Breeds of India

A study was conducted to find the single nucleotide polymorphism in the aromatase gene in nine Indian sheep breeds. Polymorphism was recorded in the Indian sheep breeds which have both the alleles A/B in the populations. The overall frequencies of alleles A and B in the breeds and crossbreds were 0.48 and 0.52, respectively. AB genotype predominated in all the breeds, whereas AA and BB genotype was absent in Magra, Patanwadi and Dumba breeds.

Effect of gene polymorphism of growth hormone on growth traits in Malpura and Avikalin sheep

A study was done to find association of haplotypes with body weight at different ages in Avikalin and Malpura breeds of sheep. There was difference in body weights for different haplotype group. However, the effect of the genotype was not significant on body weight at different ages in both the breeds. This may be due to highly unbalanced genotype distribution in the population. Further studies are required for the assessment of other loci and genes for their association with growth traits.

Major Histocompatibility Complex-DQA Variants Analysis: Rich Repertoire with Selection Advantages and Disadvantages

Insulin dependent diabetes mellitus, an autoimmune disorder in human being, is associated with MHC class-II, which reacts with auto peptide released by MHC class-II. HLA-DQ alleles (DQA1 and DQB1) are revealed to be linked with type 1 diabetes in humans. The DQ alleles passed from parents to the offspring predisposes them with risk of type I diabetes. In present study, DQ variants (DQA1 and DQA2) were amplified and sequenced with product size 826 bp (DQA1) and 865 bp (DQA2) from sheep’s blood. Sequences were insilico characterized for percent identity, phylogeny, and protein property. Phylogeny analysis suggests a rich repertoire of DQA alleles in sheep. Ramachandran plot analysis had shown the residues are occurring under the core region of both the DQ loci with 90.8% and 92.1% of residues lie in the favored region in DQA1 and DQA2 loci, respectively.

The objective of the study is impotance of casein protein genetic polymorphism of goat milk. Goat milk is highly digestible and has medicinal properties in human being. Goat milk and its value added dairy products have higher demand in many contries of world. Due to evolution process, much genetic variability has been developed in casein protein genes. This is originatd due to point mutation; addition/deletion and many other factors lead to huge genetic variation in goat casein genes. Because of this goat milk, chemical and biological properties have been affected. Goat milk casein contents decide the cheese making properties as well as nutritional and medicinal values. Goat milk is good alternative source of nutrition in human when people have cow milk problems. Goat milk has humerous inactive bioactive peptides in their casein protein. These have important place for human health and nutrition. In the future, this is great possibility for development of medicinal value functional dairy products from functional bioactive peptides.

Sequence analysis and identification of new variations in the coding sequence of melatonin receptor gene (MTNR1A) of Indian Chokla sheep breed

Melatonin receptor 1A gene is the prime receptor mediating the effect of melatonin at the neuroendocrine level for control of seasonal reproduction in sheep. The aims of this study were to examine the polymorphism pattern of coding sequence of MTNR1A gene in Chokla sheep, a breed of Indian arid tract and to identify new variations in relation to its aseasonal status. Genomic DNAs of 101 Chokla sheep were collected and an 824 bp coding sequence of Exon II was amplified. RFLP was performed with enzyme RsaI and MnlI to assess the presence of polymorphism at position C606T and G612A, respectively. Genotyping revealed significantly higher frequency of M and R alleles than m and r alleles. RR and MM were found to be dominantly present in the group of studied population. Cloning and sequencing of Exon II followed by mutation/polymorphism analysis revealed ten mutations of which three were non-synonymous mutations (G706A, C893A, G931C). G706A leads to substitution of valine by isoleucine Val125I (U14109) in the fifth transmembrane domain. C893A leads to substitution of alanine by aspartic acid in the third extracellular loop. G931C mutation brings about substitution of amino acid alanine by proline in the seventh transmembrane helix, can affect the conformational stability of the molecule. Polyphen-2 analysis revealed that the polymorphism at position 931 is potentially damaging while the mutations at positions 706 and 893 were benign. It is concluded that G931C mutation of MTNR 1A gene, may explain, in part, the importance of melatonin structure integrity in influencing seasonality in sheep.

Availability of energy in the ruminant livestocks depends on the types of food and rumen fermentation process efficiency. Forage/fodder digestions in ruminant livestocks are nearly half of what have been taken in feeding interval, because the green or dry forage/fodder digestion is not much efficient due to poor rumen fermentation process in mostly ruminants. Rumen microorganism has wide genus and species level bio-diversity which depend on the availability of forage/fodder. Rumen microorganisms are living in anaerobic environment condition and most of them are unculturable in the laboratory. Very few rumen microorganisms have been isolated and cultivated in the laboratory. This review highlights the diversity and important function of the rumen micro biota. Rumen micro biota availability and activity is affected by diet composition in ruminant livestocks and wild animals. In the future, there is lot of scope for the improvement of rumen fermentation. Through modification of rumen micro-biota population activity can increase livestock’s productivity by conserving the natural resources.

Proteasomal inhibition sensitizes cervical cancer cells to mitomycin C-induced bystander effect: the role of tumor microenvironment

Abstract Inaccessibility of drugs to poorly vascularized strata of tumor is one of the limiting factors in cancer therapy. With the advent of bystander effect (BE), it is possible to perpetuate the cellular damage from drug-exposed cells to the unexposed ones. However, the role of infiltrating tumor-associated macrophages (TAMs), an integral part of the tumor microenvironment, in further intensifying BE remains obscure. In the present study, we evaluated the effect of mitomycin C (MMC), a chemotherapeutic drug, to induce BE in cervical carcinoma. By using cervical cancer cells and differentiated macrophages, we demonstrate that MMC induces the expression of FasL via upregulation of PPAR γ in both cell types (effector cells) in vitro , but it failed to induce bystander killing in cervical cancer cells. This effect was primarily owing to the proteasomal degradation of death receptors in the cervical cancer cells. Pre-treatment of cervical cancer cells with MG132, a proteasomal inhibitor, facilitates MMC-mediated bystander killing in co-culture and condition medium transfer experiments. In NOD/SCID mice bearing xenografted HeLa tumors administered with the combination of MMC and MG132, tumor progression was significantly reduced in comparison with those treated with either agent alone. FasL expression was increased in TAMs, and the enhanced level of Fas was observed in these tumor sections, thereby causing increased apoptosis. These findings suggest that restoration of death receptor-mediated apoptotic pathway in tumor cells with concomitant activation of TAMs could effectively restrict tumor growth.
