OBSERVATIONS ON THE INTERACTIONS OF PROTAMINE WITH MOUSE WHOLE BLOOD COMPONENTS

This experiment studied the interactions of protamine sulfate with mouse whole blood components. BALB/c/Tex mice were inoculated i.p. with varying concentrations of protamine sulfate. At selected time intervals, blood samples were obtained and examined by electrophoretic, immunological and morphological methods. Detectable changes were evident when 1 mg of protamine was used. At one and four hours after inoculation, serum protein electrophoretic results were abnormal but returned to normal by 24 hours. Prothrombin times were abnormal until 48 hours postinoculation. The latter results correlated with the blood protamine level. After inoculation with protamine, an absolute increase in circulating leukocytes also occurred with a relative percent increase in neutrophils and a decrease in lymphocytes. These values were normal by 24 hours postinoculation. Changes in red cell lactate dehydrogenase isoenzyme patterns were observed. The significance of this observation is discussed.

Cytologic assessment of tumor cell kinetics: Applications of monoclonal antibody Ki‐67 to fine‐needle aspiration smears

Rates of tumor-cell proliferation often provide prognostic information about a given neoplasm. Previously available methods for accessing cell kinetics are time consuming and expensive, and often require special equipment or radioactive reagents. Monoclonal antibody Ki-67 binds a nuclear antigen expressed in proliferating but not in resting cells. We studied Ki-67 immunostaining of fine-needle aspiration smears from 40 benign and malignant masses. Labeling indices ranged from 0 (thyroid follicular adenoma) to 75 percent (pulmonary oat-cell carcinoma). Frozen section immunostaining (11 cases) and flow cytometric assessment of cell proliferation (8 cases) were in good agreement with Ki-67 labeling indices on smear material. We suggest that this method provides a rapid, inexpensive, and dependable means of assessing tumor-cell kinetics in cytologic preparations.

Ornithosis: Immunization of Turkeys with Miyagawanella of Low Virulence

INTRODUCTION The clinical signs of ornithosis in turkeys have been related directly to the toxin-producing ability of Miyagawanella ornithosis (12). From natural outbreaks of this disease, causative microorganisms with various toxin-producing capacities have been isolated (12,9). Miyagawanella with a high index usually produce a higher incidence of mortality in infected turkeys (6). Isolates with a low toxic index isolated from infected turkeys have produced little mortality or clinical signs of disease (7). Cases of ornithosis have been reported in persons associated with turkeys infected with both types of microorganisms (6,10). At present, control of the disease in turkeys has been based upon prophylactic use of broad-spectrum antibiotics (3). Because control of this disease by immunological rather than prophylactic use of antibiotics might be desirable, this investigation was conducted to study the efficacy of Miyagawanella ornithosis isolates with low virulence used as immunizing agents against this disease.

The fluorescent antibody technique, since its first application to the study of microorganisms (2), has been successfully applied to the identification of microorganisms in clinical specimens (4,9,12,13). With respect to the causative agent of ornithosis in turkeys, one investigator (5) applied this method to the identification of this agent in direct smears but reported the method gave poor results. Nevertheless, because of its potential value in diagnostic and epidemiological studies of this disease, application of the fluorescent antibody technique to the detection of this infectious agent was re-examined.

Use of a liquid nitrogen container for convenient transport of frozen mammary carcinoma tissue for hormone-receptor assays is described. The container can be used in dry or wet mode and is appropriate for all types of transport, including air. Experience shows a significantly higher incidence of positive tests on specimens transported by this means than on specimens transported in dry ice.

Chronic myelogenous leukemia in Hodgkin's disease: Immunofluorescence of cells

A patient with Hodgkin's disease is described who developed chronic myelogenous leukemia 8 1/2 years after the start of radiation therapy. This therapy was fractional and was administered to most of the patient's skeleton. The radiation was an important factor in the development of leukemia. The presence of positive reactions to the antileukemic fluorescent antibodies may be consistent with the hypothesis that viruses are concerned with human leukemia.
