Erythropoietic Activity of Untreated and Deproteinized Normal Human Plasma.

Significant erythropoietic stimulating activity was found in 27 out of 42 samples of untreated normal human plasma assayed in the transfused polycythemic mouse. Deproteinization by boiling of plasma resulted in a marked reduction in the fraction of active samples. Both whole untreated normal plasma and deproteinized extracts were essentially inactive in hypophysectomized and starved rats. The possibility that these results are due to low levels of erythropoietin in normal plasma is discussed.

Effect of Low Pressure on the Blood Picture of Necturus Maculosus

This problem was undertaken for three reasons. (1) There are no well controlled experiments, so far as I know, in which an aquatic form has been subjected to a constant low pressure and examined for changes in its blood picture. Necturus was chosen as the experimental animal because of the readily available source of blood in the afferent branchial arteries of the external gills and because the normal blood picture has been thoroughly worked out by Dawson. (2) Experiments such as those described below supply a method for determining the nature of the red cell ancestors in Necturus by causing them to leave the erythrocytopoietic loci and enter the circulating blood. (3) It is interesting to compare the intensity and duration of the stimulus required to send out erythroid cells into the blood stream of an aquatic form with the intensity and duration necessary to evoke an analogous response in mammals (i. e., increase the reticulocyte and total red cell count). Groups of Necturi were placed in jars of water and subjected to a constant pressure of 330 mm. Hg. in a specially devised low pressure chamber for varying periods up to 9 weeks of continuous exposure. (For the construction of the apparatus, see Dubin.) The animals were removed once a day for about an hour, and the water in which they were kept was replaced by fresh water. All animals, including the controls, were fed live earthworms twice weekly. In order to minimize the effects of hemorrhage, the small quantity of blood necessary for total counts and 2 smears was drawn on 2 occasions only, once, before the animals were placed in the tank, and later, when they were examined to determine the effects of the reduced pressure.

Reply Creatine supplementation in chronic heart failure

Dr. M.L. Field has in a Letter to the Editor pointed out several interesting theoretical aspects on creatinine loading of cardiomyoytes concluding the lack of efficacy of creatine supplementation in treating the failing myocardium. The Letter discusses the possibly important role of the free creatine store as ATP acceptor with simultaneous ADP release at the inner mitochondrial membrane. An increased creatine concentration would amplify the local ADP concentration and stimulate respiration [l]. The creatine concentration in the human diseased heart is 1218 mM [2,3], but in the normal contractile cycle 60-70% of the creatine is phosphorylated [4]. The free creatine available for mitochondrial creatine kinase will thus correspond to approximately 5 mM, which is below the K, value given by Saks et al. [5]. This would indicate a positive role for creatine supplementation as stimulator of respiration in heart tissue in cardiac failure. This will of course not necessarily give an increase of the ejection fraction, but the measurement seemed justified since the hypothesis has never been tested. Furthermore, depressed myocardial total creatine levels were reported from explanted hearts [3] that probably had a more deranged myocardial function than present in our patient group. Perhaps a recruitment of a considerably larger and sicker patient population would be necessary to achieve efficacy.

Reticuloendothelial system (RES) hyperfunction and erythropoietin (Ep) production in the regenerating liver

Abstract Hepatic cells in rats were evaluated after subtotal hepatectomy using scintillation scanning with technetium sulfur colloid (TSC), autoradiography, and microstereology techniques. The ability of the liver to accumulate TSC increased during the course of the regeneration as did the labeling of Kupffer and parenchymal cells with tritiated thymidine ( 3 H‐tdR). Kupffer to parenchymal cell number ratios and Kupffer cell relative areas were also elevated, attaining peak values at 72 hours post‐hepatectomy. This period corresponds to the time of peak erythropoietin (Ep) production in rats with regenerating livers after nephrectomy and exposure to hypoxia. These findings suggest that the Kupffer cell may function as a cellular site of Ep formation.

Quantitative Effects of Corticosterone on Rat Bone Marrow

A method has been described for the determination of the total numbers of bone marrow cells in the rat femur. Administration of corticosterone to adrenalectomized rats results in significant increases in the numbers of nucleated erythroid cells and decreases in the numbers of neutrophils and eosinophils within femoral marrow.

Renal Mechanisms Underlying Actions of Androgen and Hypoxia on Erythropoiesis.

The appearance of increased quantities of ESF in the sera of rats following androgen administration or exposure to hy-poxia has been related to increased amounts or activity of the renal erythropoietic factor (REF). Plethora induced in rats by hyper-transfusion results in a marked decrease in REF activity in response to androgen or hypoxia. This is proposed as the mechanism by which plethora inhibits the production of ESF.

Chromosomal evidence for the transplantability of the shay chloroleukemia

Abstract Chloroma cells, which were shown to contain the X and Y chromosomes, were used to induce Shay chloroleukemia (SCL) in female recipients. Karyotype analyses of leukemic myeloblasts which appeared in the peripheral blood of the female recipients during blast crisis demonstrated invariably the presence of the X and Y chromosomes. These results directly demonstrate that transmission of SCL is the result of donor cell colonization and therefore SCL can be considered a true transplantable leukemia. Reports by other workers of viral association with this dissease are discussed in light of our results.

Inhibition of erythroleukemia in mice by induction of hemolytic anemia prior to infection with Rauscher leukemia virus.

The hematological changes induced by Rauscher leukemia virus under normal, erythropoietically stimulated, and erythropoietically depressed conditions were studied quantitatively. It was found that suppression of erythropoiesis induced by exhypoxic polycythemia, and maintained by transfusion-induced plethora, retarded the disease process over a 4-month period, whereas stimulation of erythropoiesis by phenylhydrazine-induced hemolytic anemia resulted in a greatly decreased incidence of Rauscher leukemia virus disease. The measurement of gs3 antigen by fluorescent antibody definitely established that, while most erythroblasts in phenylhydrazine-treated mice were infected, normal maturation to erythrocytes occurred. In mice not pretreated with the drug prior to virus infection, reduced numbers of infected cells were seen, although maturation inhibition and consequent terminal anemia and hepatosplenomegaly were prominent.
