Objective:To further raise the knowledge of MR manifestation of hemangioblastomas.Methods:The MR appearance of 22 cases of hemangioblastomas proved by surgery and pathology were analyzed retrospectively.Results:The location of hemangioblastomas 14 cases was in hemisphere cerebellum,4 cases in vermis cerebelli,2 cases in cervical cord,1 case in medulla oblongata,1 case in hemisphere cerebellum and cervical cord,14 cases with cyst and tubercle type,2 cases with cyst type,6 cases with solid type.The tumor tubercle and tumor body of solid type were highly enhanced.Blood flow void phenomena could be seen in 6 cases with MR.There was slight and medium edema around the tumor.Conclusion:MR supplemented show important for diagnosis and differential diagnosis of hemangioblastomas.
Objective:To study the relationship between SCT findings and the expression of N ras gene?C myc gene in hepatocellular carcinoma(HCC).Methods:SCT scan was performed in 36 patients with pathologically proved hepatocellular carcinoma,and the expression of N ras?C myc gene in tumor specimens was estimated with immunohistochemical SABC method.Results:C myc protein positive rate was higher in the group whose diameter is smaller than 5cm and equal to 5cm,and larger than 3cm than that whose diameter is smaller than 3cm and equal to 3cm,and lower in the group cirrhosis of liver(P0.05).Conclusion:This study shows the SCT findings of HCC is related with the expression of the gene,and the SCT imaging of HCC can indicate the expression of C myc gene to some extent.
In order to explore the regulation mechanism of fibroin heavy chain gene in silkworm (Bombyx mori),the recombinant AcMNPV was used as the transfer vector and green fluorescent protein (EGFP) gene was used as the reporter gene to investigate the expression profiles of the reporter gene in silkworm tissues of the 5th instar larvae driven by differently truncated promoters of fibroin heavy chain gene (FibH) (-2 127 /+23,-925 /+23 and-238 /+23). The results showed that there was ectopic expression of EGFP in fat body and hemocyte which was driven by three FibH promoters of various lengths. Green fluorescence could be observed directly on silkworm cuticle under fluorescence microscope. The three promoters of various lengths could initiate transcription of EGFP efficiently in posterior silk gland and produced green fluorescent light. Promoters of 0.9KH and 0.2KH could initiate transcription of EGFP in middle silk gland and yielded green fluorescent light. That of 2.1KH could initiate transcription of EGFP but no green fluerescent light was detected. All three promoters could also drive ectopic expression of EGFP in Sf9 cultured cells. The above results revealed that the promoter of fibroin heavy chain gene does not have strict tissue specificity.
Objective:To study the value of MRI in diagnosis and differential diagnosis of spinal metastatic tumor.Methods:The MRI appearances of 34 cases of spinal metastatic tumor were retrospectively analyzed.Results:①of 34 cases,single vertebra was involied in 6 cases,and multiple vertebrae were involved in 28 cases;②The lesions were all hypointense on T 1WI.On T 2WI,26 cases appeared hyperintense,7 isointense and one hypointense;③The invasion of vertebra could be divided into 3 types:osteolytic,osteoblastic and mixed type;④In 7 cases a paravertebral mass was observed;⑤There was narrowing of the spinal canal in 13 cases;⑥The jump over sign occurred in 20 cases;⑦The intervertebral disks were normal in all 34 cases.Conclusion:The spinal metastatic tumor has characteristic features on MRI.Careful and comprehensive analysis is helpful in diagnosis and differential diagnosis.
Objective:To study the influnence of elemene or heat shock treatment upon the expression of membrane HSP70 and HSPs genes in HepG 2 cells.Methods:HSP70 expression was observed by immunofluorescence and FCM techniques.HSP70 gene transcription was blocked by ActD.Genechip was used to study the expression of HSPs genes and the genes associated with the control of HSPs transcription.Results:The treatment of either elemene(50 μg/ml,1 h) or heat shock(42℃,1 h)could increase the membrane expression of HSP70,ActD had a synergistic action with both treatments.HSP70EP(Enhancer Protein) gene was up regulated and HSPA2 gene was down regulated in both treatments.HSF1 gene was up regulated in heat shock treatment but down regulated in elemene treatment.The genes closely correlated with tumor immune,such as HSP70,HSP72,HSP75,HSP90 and gp96 were not changed.Conclusion:Comparing with heat shock treatment,elemene can cause a higher percent of HSP70 membrane expression in HepG 2 cells and that may be caused by its altering the distribution of the already existing HSP70 and/or accelerating the HSP70 mRNA translation.Both treatments can change the expression of genes associated with the transcription control factors of HSPs and decrease HSPA2 gene expression.
Objective To explore the possibility of detecting Grp78 expression levels in the hydrothorax detached cells in the differential diagnosis in chest tumors.Methods The expression levels of Grp78 in 36 cases of suspended chest tumor patients were detected by western blot and the hydrothorax detached cells analyses were also detected by HE staining.Results In the 21 cases of patients that Grp78 is overexpressed in the hydrothorax detached cells,19 cases of patients were identified as lung cancer by retrospective analysis of clinicopathological data.In the 15 cases of patients that Grp78 was radically expressed,none of which was diagnosed as cancer.In the 12 cases of patients diagnosed as lung cancer by the hydrothorax detached cells analysis by HE staining,4 cases were excluded from the diagnosis of cancer based on clinicopathological analysis.The sensitivity of HE staining is 47% compared with 81% of western blot.The specificity of HE staining and western blot is 85% and 85.7% respectively.Conclusions Detection of Grp78 expression levels in the hydrothorax detached cells is a useful target in the differential diagnosis of chest tumor.
