Multi‐ATOM: Ultrahigh‐throughput single‐cell quantitative phase imaging with subcellular resolution

A growing body of evidence has substantiated the significance of quantitative phase imaging (QPI) in enabling cost-effective and label-free cellular assays, which provides useful insights into understanding the biophysical properties of cells and their roles in cellular functions. However, available QPI modalities are limited by the loss of imaging resolution at high throughput and thus run short of sufficient statistical power at the single-cell precision to define cell identities in a large and heterogeneous population of cells-hindering their utility in mainstream biomedicine and biology. Here we present a new QPI modality, coined multiplexed asymmetric-detection time-stretch optical microscopy (multi-ATOM) that captures and processes quantitative label-free single-cell images at ultrahigh throughput without compromising subcellular resolution. We show that multi-ATOM, based upon ultrafast phase-gradient encoding, outperforms state-of-the-art QPI in permitting robust phase retrieval at a QPI throughput of >10 000 cell/sec, bypassing the need for interferometry which inevitably compromises QPI quality under ultrafast operation. We employ multi-ATOM for large-scale, label-free, multivariate, cell-type classification (e.g. breast cancer subtypes, and leukemic cells vs peripheral blood mononuclear cells) at high accuracy (>94%). Our results suggest that multi-ATOM could empower new strategies in large-scale biophysical single-cell analysis with applications in biology and enriching disease diagnostics.

Cost-effective approaches for high-resolution bioimaging by time-stretched confocal microscopy at 1μm

Optical imaging based on time-stretch process has recently been proven as a powerful tool for delivering ultra-high frame rate (< 1MHz) which is not achievable by the conventional image sensors. Together with the capability of optical image amplification for overcoming the trade-off between detection sensitivity and speed, this new imaging modality is particularly valuable in high-throughput biomedical diagnostic practice, e.g. imaging flow cytometry. The ultra-high frame rate in time-stretch imaging is attained by two key enabling elements: dispersive fiber providing the time-stretch process via group-velocity-dispersion (GVD), and electronic digitizer. It is well-known that many biophotonic applications favor the spectral window of ~1μm. However, reasonably high GVD (< 0.1 ns/nm) in this range can only be achieved by using specialty single-mode fiber (SMF) at 1μm. Moreover, the ultrafast detection has to rely on the state-of- the-art digitizer with significantly wide-bandwidth and high sampling rate (e.g. <10 GHz, <40 GS/s). These stringent requirements imply the prohibitively high-cost of the system and hinder its practical use in biomedical diagnostics. We here demonstrate two cost-effective approaches for realizing time-stretch confocal microscopy at 1μm: (i) using the standard telecommunication SMF (e.g. SMF28) to act as a few-mode fiber (FMF) at 1μm for the time-stretch process, and (ii) implementing the pixel super-resolution (SR) algorithm to restore the high-resolution (HR) image when using a lower-bandwidth digitizer. By using a FMF (with a GVD of ~ 0.15ns/nm) and a modified pixel-SR algorithm, we can achieve time-stretch confocal microscopy at 1μm with cellular resolution (~ 3μm) at a frame rate 1 MHz.

Arbitrary two-dimensional spectrally encoded pattern generation—a new strategy for high-speed patterned illumination imaging

Patterned illumination is now a proven technique in optical metrology and imaging, and is widely employed in both industrial and biological applications. However, existing techniques are unable to meet the pressing need for higher imaging rates. To address this challenge, we propose and demonstrate two-dimensional (2D) mechanical-scan-free arbitrary patterned illumination by 2D spectral encoding. The illumination pattern is flexibly generated at high speed by spectral shaping together with wavelength-to-2D space mapping. Performance optimization of this new patterned illumination scheme (e.g., pattern distortion and resolution) is generally an ill-defined problem involving multiple interrelated parameters. We demonstrate proof-of-principle experiments based on a multiobjective optimization routine using a genetic algorithm to validate our optimization model as well as to show the feasibility of patterned illumination by use of spectral interferometry. Adopting the wisdom of high-speed arbitrary waveform generation routinely practiced in telecommunications as well as ultrafast wavelength-sweep mechanisms, the proposed method could have an impact on advanced imaging modalities, particularly when speed is of a critical concern.

Peran Dukungan Sosial Dan Resiliensi Terhadap Kualitas Kehidupan Orang Tua Empty Nest

Kondisi empty nest syndrome seringkali dikaitkan dengan rasa kehilangan yang dialami orang tua saat terpisah dari anak-anaknya, hal tersebut berdampak pada kehidupan orang tua dengan empty nest syndrome. Empty nest syndrome merupakan fenomena pada orang tua yang mengalami transisi perpisahan dengan anak-anaknya. Tinggi atau rendahnya tingkat empty nest syndrome dapat dipengaruhi oleh resiliensi dan dukungan sosial yang dimiliki orang tua. Tujuan penelitian adalah untuk mengetahui peran resiliensi dan dukungan sosial terhadap kualitas kehidupan orang tua empty nest. Penelitian ini dilakukan pada partisipan yang memenuhi karakteristik orang tua empty nest dengan rentang usia 45-65 tahun, dengan jumlah 166 partisipan yang berdomisili dari berbagai wilayah di Indonesia. Pengambilan data dilakukan dengan teknik purposive sampling dengan mengisi form kuesioner yang disebarkan melalui platform online selama tiga bulan. Terdapat tiga alat ukur yang digunakan, yaitu Multidimensional Scale of Perceived Social Support, Resilience Scale, dan alat ukur Kualitas Kehidupan. Analisis data menggunakan korelasi Pearson Correlation pada variabel resiliensi dan dukungan sosial dengan kualitas kehidupan menunjukkan bahwa hanya dukungan sosial yang memiliki korelasi signifikan dengan kualitas kehidupan, dengan nilai signifikan (p< .001). Uji regresi linier berganda menunjukkan bahwa dukungan sosial memiliki peran terhadap kualitas kehidupan (F(163) = 12.031; p < .001). Sumbangan efektif faktor dukungan sosial terhadap kualitas kehidupan orang tua empty nest syndrome sebesar 12.9%.

Optical time-stretch confocal microscopy at 1 μm

We demonstrate optical time-stretch confocal microscopy in the 1 μm spectral window for high-speed and high-resolution cellular imaging. In contrast to the prior demonstrations of time-stretch imaging, which all operated in the telecommunication band, the present work extends the utility of this imaging modality to a wavelength regime (~1 μm), which is well known to be the optimal diagnostic window in biophotonics. This imaging technique enables us to image the nasopharyngeal epithelial cells with cellular resolution (<2 μm), at a line scan rate of 10 MHz, and with a field of view as wide as ~0.44 mm × 0.1 mm. We also theoretically and experimentally characterized the system performance. As the low-loss dispersive fibers for the time-stretch process as well as other essential optical components for enhancing the imaging sensitivity are commonly available at 1 μm, time-stretch confocal microscopy in this wavelength range could usher in realizing high-speed cell imaging with an unprecedented throughput.
