A new technique for immunochromatographic serodiagnosis of tuberculosis that is based on the interaction of specific anti-mycobacterial immunoglobulins from blood serum with label—antigen conjugate and subsequent binding of the formed complex with the Staphylococcus aureus protein A immobilized on a test-strip has been suggested. Unlike the conventional technique in which all the immunoglobulins in a sample react with the label—immunoglobulin-binding protein conjugate, and then the labeled specific antibodies interact with an immobilized antigen, this technique prevents binding of the label to immunoglobulins that are not specific to Mycobacterium tuberculosis. The suggested assay was implemented using the recombinant 38 kDa (Rv0934) protein of M. tuberculosis as an antigen and colloidal gold as a label. It was experimentally shown that the proposed technique allows to increase the percentage of detection for seropositive serum with low concentrations of the specific antibodies to the M. tuberculosis cause.

[Improving the efficacy of tuberculosis immunodiagnosis using anti-human IgG monoclonal antibodies].

Monoclonal antibodies (MAb) F5 to human IgG were used for creating immunoperoxidase conjugate. MAb dissociation constant was 10(-9)M-1 and the number of binding sites 1. Enzyme immunoassay (EIA) and immunoblotting showed that MAb F5 specifically recognize conformation epitope on intact human IgG molecule but not on other human immunoglobulins or denatured IgG. The resultant peroxidase conjugate with MAb F5 was used for EIA titration of antibacterial antibodies in sera from 30 patients with pulmonary tuberculosis, 28 patients with nonspecific pulmonary diseases (bronchitis and/or asthma, pneumonia), and 12 donors. For comparison similar studies were carried out with reference commercial immunoperoxidase conjugate to human IgG(H + L) manufactured at N. F. Gamaleya Institute of Epidemiology and Microbiology. Mycobacterium tuberculosis monoantigen (mol. weight 15-18 kD), affinity isolated by antibacterial MAb S4C1G4 (alpha S4C1G4), and PPD (Batch RT 45, Stattens Seruminstitut, Denmark) were used. Sensitivity and specificity of serum antibacterial antibodies were compared. The specificity of conjugate based on MAb F5 with monoantigen alpha S4C1G4 was 78.21%, sensitivity 94.50%, while those of conjugate to human IgG(H + L) were 53.30 and 76.89%, respectively (p < 0.001). For PPD the specificity and sensitivity were 56.75 and 72.33%, respectively (conjugate with MAb F5) versus 47.67 and 62.38% for conjugate against IgG(H + L), p < 0.001. Similar values were obtained in assessment of the concentrations of antibodies to alpha S4C1G4 for MAb F5 conjugate: specificity and sensitivity 47.16 and 71.56%, respectively, versus 23.67 and 95.16% for PPD (p < 0.05). No statistically significant differences between the experimental groups were detected with IgG(H + L) conjugate. We believe that specific MAb-based conjugate to human IgG will improve the efficacy of EIA as a method for the diagnosis of pulmonary tuberculosis.

Protease-resistant protein and lipid mycobacterial antigens are present in biological fluids of pulmonary tuberculosis patients. Anti-tuberculosis monoclonal antibodies (MAb) reacting to proteins participating in the molybdopterin synthesis have been obtained. With the help of two MAb recognizing different epitopes on one antigen with molecular mass of 32, 50 and 75 kilodalton. With the help of which the two-site sandwich of IFA type capable of detecting the mycobacterial antigen in antigenic preparations of х M. tuberculosis and in urine samples of tuberculosis patients. However the specificity of this test is fairly low since when testing urine samples this antigen was found in the half of healthy donors. It is due to the fact that in human urogenital tract some opportunistic pathogens are present which belong to normal bacterial population with which the tested MAb have the cross-reaction.

ESTIMATION OF SPECIFICS OF ANTITUBERCULOUS HOMOGENEOUS ANTIBODIES IN THE SANDWICH IMMUNOENZYMOMETRIC ANALYSIS

В двусайтовом иммуноферментном анализе с антимикобактериальными моноклональными антителами (МАТ) и кроличьей противотуберкулезной сывороткой (АС) проанализирована специфичность распознаваемых МАТ антигенов микобактерий. Исследованы 14 МАТ против различных антигенов M.tuberculosis и 12 нетуберкулезных микобактерий. Наибольший интерес вызвал антиген против которого были направлены МАТ 1B7 (конформационный эпитоп?). Авторы считают, что исследование микобактериальных антигенов в подобном «гетеросэндвиче» МАТ-АГ-АС позволяет быстро проверить специфичность как самих МАТ, так и нативных АГ цельных клеток микобактерий с которыми они реагируют.

DIRECT AND COMPLEMENT-MEDIATED EFFECTS OF ANTIMYCOBACTERIAL MONOCLONAL ANTIBODIES ON MYCOBACTERIUM TUBERCULOSIS GROWTH IN VITRO

Впервые представлены экспериментальные данные по поиску противотуберкулезных моноклональных антител (МАТ), действующих на рост живых вирулентных микобактерий в присутствие комплемента и без него in vitro. В цитотоксических тестах исследована панель из 32 МАТ. Среди них не выявлены антитела, которые бы ингибировали репликацию возбудителя in vitro. Добавление комплемента к МАТ также не приводило к статистически значимым результатам воздействия на микобактерии. Авторы предполагают, что специфичности антител, способные ингибировать рост микобактерий in vitro редки или вовсе отсутствуют в организме хозяина.

MONOCLONAL ANTIBODIES TO Mycobacterium avium IN THE RESEARCH INTO PULMONARY MYCOBACTERIOSIS

We used hybridoma technology to obtain 15 monoclonal antibodies (mAbs) to antigens of whole-cell suspension of M. avium. The isotypes and specificity of mAbs to mycobacterial and bacterial antigens were characterized. The chemical nature of the antigens/antigenic determinant recognized by the mAbs under treatment with proteinase K and metaperiodate Na and using extracted with ethanol and benzene M. avium suspension was investigated. All mAbs were purified by affinity chromatography and used in heterologous indirect two-site ELISA to determine diagnostic properties in the study of IgG antibodies in sera of patients with pulmonary tuberculosis, pulmonary mycobacteriosis or healthy donors. The presented method makes it possible to individually identify and take into account non-specific “natural” IgG antibodies of each serum under study (patent for invention No. 2794855). mAbs 1B11 showed diagnostic specificity in the detection of mycobacteriosis − 62.47% and sensitivity – 74.78%, 3E12 (anti-54kDa) − 88.89% and 52.63%, and 1H7G6 − 66.81% and 83.80%, respectively.
