[Characteristics of the condylar movement traces during lateral excursion of the unilateral posterior buccal crossbite].

To investigate the characteristics of the condylar movement traces during lateral excursion of the unilateral posterior buccal crossbite and to compare the difference with the individual ideal occlusion.Twenty-six subjects with normal occlusion and 26 subjects with unilateral posterior buccal crossbite were tested by CADIAX for the condylar movement. The lateral excursion traces were recorded. The data were analyzed using SPSS 10.0 software package, group t test,paired t test or rank sum test were used to compare the difference between the two groups.The condylar traces of the experiment subjects were rugged and asymmetrical. The sagittal,vertical and space shift of the non-buccal crossbite side were larger than those of the buccal crossbite side and the normal subjects, but the horizontal shift was significantly smaller(P<0.05). The sagittal condylar inclination of the non-buccal crossbite side was significantly larger than that of the buccal crossbite side and the normal subjects, but the transversal condylar inclination was significantly smaller(P<0.05).The bilateral condylar movement traces during lateral excursion of the unilateral posterior buccal crossbite are asymmetrical.

Dihydroartemisinin eliminates senescent cells by promoting autophagy‐dependent ferroptosis via AMPK/mTOR signaling pathway

Abstract Cellular senescence is an irreversible cell‐cycle arrest in response to a variety of cellular stresses, which contribute to the pathogenesis of a variety of age‐related degenerative diseases. However, effective antisenescence strategies are still lacking. Drugs that selectively target senescent cells represent an intriguing therapeutic strategy to delay aging and age‐related diseases. Thus, we thought to investigate the effects of dihydroartemisinin (DHA) on senescent cells and elucidated its mechanisms underlying aging. Stress‐induced premature senescence (SIPS) model was built in NIH3T3 cells using H 2 O 2 and evaluated by β‐galactosidase staining. Cells were exposed to DHA and subjected to cellular activity assays including viability, ferroptosis, and autophagy. The number of microtubule‐associated protein light‐chain 3 puncta was detected by immunofluorescence staining. The iron content was assessed by spectrophotometer and intracellular reactive oxygen species (ROS) was measured by fluorescent probe dichlorodihydrofluorescein diacetate. We found that DHA triggered senescent cell death via ferroptosis. DHA accelerated ferritin degradation via promoting autophagy, increasing the iron contents, promoting ROS accumulation, thus leading to ferroptotic cell death in SIPS cells. In addition, autophagy inhibitor BafA1 preconditioning inhibited ferroptosis induced by DHA. Moreover, Atg5 silencing and autophagy inhibitor BafA1 preconditioning inhibited ferroptosis induced by DHA. We also revealed that the expression of p‐AMP‐activated protein kinase (AMPK) and p‐mammalian target of rapamycin (mTOR) in senescent cells was downregulated. These results suggested that DHA may be a promising drug candidate for clearing senescent cells by inducing autophagy‐dependent ferroptosis via AMPK/mTOR signaling pathway.

[Preliminary study of cranio-maxillofacial hard tissue structure feature of young mandibular retrusion orthognathic patients with delaire cephalometric analysis].

To guide the orthodontic diagnosis, treatment planning and prognosis by analyzing the craniofacial morphology and reimbursement mechanism of young patients with mandibular retrusion in Chongqing territory by Delaire cephalometric analysis.Both conventional cephalometric analysis and Delaire cephalometric analysis were conducted on the basis of the lateral cephalograms from 56 young mandibular retrusion patients and 40 teenagers with normal occlusion.Decreases in angle SNB, Co-Go, Co-Pog, U1-L1 and increases in angle ANB, angle SN-MP, L1-MP were seen in mandibular retrusion group by conventional cephalometric analysis. Decreases in angle 1, angle 5, (Cp-Oi)/C2 and increases in angle 2, (M-Cp)/C2, angle 3, angle 4 were seen in mandibular retrusion group by Delaire cephalometric analysis. Me-F1 and Me-Met were -8.70 mm +/- 2.48 mm and 5.74 mm +/- 2.58 mm respectively.Delaire cephalometric analysis could evaluate cranio-maxillofacial architectural features of mandibular retrusion patients more visually and quantitatively, which suggests that mandibular retrusion is usually caused by the retrusion of mandibular position and the hypodevelopment of mandibule.

[Researches on biomechanics of micro-implant-bone interface and optimum design of micro implant's neck].

To compare and analyze the stress distribution at the micro-implant-bone interface based on the different micro-implant-bone conditioned under orthodontic load, and to optimize the design of micro implant's neck.An adult skull with all tooth was scanned by spiral CT, and the data were imported into computer for three-dimensional reconstruction with software Mimics 9.0. The three dimensional finite element models of three micro-implant-bone interfaces(initial stability, full osseointegration and fibrous integration) were analyzed by finite element analysis software ABAQUS6.5. The primary stress distributions of different micro-implant-bone conditions were evaluated when 2N force was loaded. Then the diameter less than 1.5 mm of the micro implant's neck was added with 0.2 mm, to compare the stress distribution of the modified micro-implant-bone interface with traditional type.The stress mostly concentrated on the neck of micro implant and the full osseointegration interface in all models showed the lowest strain level. Compared with the traditional type, the increasing diameter neck of the micro implant obviously decreased the stress level in all the three conditions.The micro-implant-bone interface and the diameter of micro implant's neck both are the important influence factors to the stress distribution of micro implant.

Autophagy inhibition via Becn1 downregulation improves the mesenchymal stem cells antifibrotic potential in experimental liver fibrosis

Abstract Liver fibrosis (LF) is the result of a vicious cycle between inflammation‐induced chronic hepatocyte injury and persistent activation of hepatic stellate cells (HSCs). Mesenchymal stem cell (MSC)‐based therapy may represent a potential remedy for treatment of LF. However, the fate of transplanted MSCs in LF remains largely unknown. In the present study, the fate and antifibrotic effect of MSCs were explored in a LF model induced by CCl 4 in mouse. Additionally, MSCs were stimulated in vitro with LF‐associated factors, tumor necrosis factor‐α (TNF‐α), interferon‐γ (IFN‐γ), and transforming growth factor‐β1 (TGF‐β1), to mimic the LF microenvironment. We unveiled that MSCs exhibited autophagy in response to the LF microenvironment through Becn1 upregulation both in vivo and in vitro. However, autophagy suppression induced by Becn1 knockdown in MSCs resulted in enhanced antifibrotic effects on LF. The improved antifibrotic potential of MSCs may be attributable to their inhibitory effects on T lymphocyte infiltration, HSCs proliferation, as well as production of TNF‐α, IFN‐γ, and TGF‐β1, which may be partially mediated by elevated paracrine secretion of PTGS2/PGE 2 . Thus, autophagy manipulation in MSCs may be a novel strategy to promote their antifibrotic efficacy.

[Mandibular condensability and length contraction: an animal experimental study].

To investigate the condensability of mandibular length.In six goats were used in the study. Corticotomy at right mandibular angles was performed via extral-oral accession. Special devices were applied to shorten the mandible by 0.5 mm per three days respectively.1. Mandibular angles of the six goats were shorten by 0.8 cm to 1.3 cm respectively in 48 days to 78 days; 2. In spite of the lower ascending ramus moving forward and angles being blunt, the occlusion scarcely varied because of contralateral bite-lock; 3. X-ray demonstrated that, at first, bone density in contracted areas declined, and then increased gradually to almost normal density; 4. Under microscope there were three tissues layers from central to lateral within the bone gap: fiber layer, cartilage layer and bone layer, and fiber layer gradually transform into cartilage layer with the fixed time. At the end of fixation they all transform into bone tissue.Contraction osteogenesis is actually a process of compression, absorption and rebuilding. It is feasible that using contraction osteogenesis to shorten the mandible via cortcotomy.

Aims: To investigate the association between the methylenetetrahydrofolate dehydrogenase 1 (MTHFD1) polymorphism rs 2236225 (c.1958G>A) and susceptibility to non-syndromic cleft of the lip and/or palate (NSCL/P).Methods: An extensive literature review has been conducted using PubMed, Web of Science, Cochrane Library, Google Scholar, the China National Knowledge Infrastructure (CNKI), and Wanfang Database for eligible researches. The terms for searching were “cleft lip OR cleft palate OR CLP OR CL/P OR oral facial cleft OR OFC” AND “methylenetetrahydrofolate dehydrogenase (NADP+ dependent) 1 OR methenyltetrahydrofolate cyclohydrolase formyltetrahydrofolate synthetase OR MTHFD1 OR MTHFD”. Two independent researchers screened, evaluated and extracted the data of included studies. The pooled odds ratios (OR) with 95% confidence intervals (95% CI) were calculated by random effects model under five gene models. Subgroup, sensitivity analysis and publication bias were also assessed.Results: Ten case-control studies have been included in the systematic review and eight studies have been considered for the meta-analysis. Overall, the MTHFD1 polymorphism rs2236225 and the risk of NSCL/P showed pooled OR (95% CI) of 1.02 (0.86-1.21) under allelic model. A higher degree of heterogeneity was observed in Asian countries (I2 = 75.6%) compared to non-Asian countries (I2 = 48.9%). Similar consequence appeared in the subgroup of children (I2 = 78.6%) compared with that of mothers (I2 = 0.0%). There was no significant difference in the publication bias by the Begg’s funnel plot (P = 0.711) and Egger’s regression test (P = 0.746).Conclusion: Our assessment suggested there was no significant association between the MTHFD1 polymorphism rs 2236225 (c.1958G>A) and the susceptibility to NSCL/P. Further investigations using a large sample size and a more advanced technique should be adopted to reach a more precise conclusion in the future.
