: Objective: The present study projected to the synthesis of 3b-Hydroxy-lup- 20(29)-en-28-oic acid analogues in order to evaluate their possible biological activity. In addition, the structure–activity relationship is also investigated. Method: BA derivatives were prepared by conjugating 3b-Hydroxy-lup- 20(29)-en-28-oic acid with different amines through carbomyl linkage. Structures of synthesized compounds were elucidated by spectral data. Cytotoxic evaluation was done by CAM assay and MTT assay. Results: Among the synthesized compounds, Compound 7showed a pronounced cytotoxicity in antiangiogenic assay as well as compound ECV-304 cell line. Compound 10 also showed good cytotoxic activity. While compounds 6 and 8 showed moderate activity against A-549 and MCF-7 respectively. Conclusion: It is concluded that synthesized BA analogues are biologically active and developed into useful anticancer agents.Key words: Triterpenoids, Lupenoic acid, Carbomyl Linkage, Anticancer activity, MTT Assay.

Diarrhoea refers to an increase in the frequency, fand/or volume of faeces, greater than normal for an individual, resulting from an imbalan ce between the secretory and absorptive forces in the intestines. The World Health Organiza tion (WHO) estimates that 3‐5 billion cases occur each year (1billion in children less than 5 y ears of age), and that approximately 5 million deaths are due to diarrhoea annually (2.5 million i n children less than 5 years of age). The incidence of diarrhoeal diseases still remains high despite the efforts of many governments and international organisations to curb it. Many synthe tic chemicals like diphenoxylate, loperamide and antibiotics are available for the treatment of diarrhoea but they have some side effects and increasing resistance of pathogen to the common ant ibiotics . In recent years, special attention is being given on alternative safe natural bio-remedies to cure the infectious diseases becaus e of their less or no side effects, high efficacy, less cost. In this paper an attempt has been made to give an overview of certain plants with their phyto constituents and mechanism of action which have been studied for their antidiarrhoel activity.

Synthesis, characterization and pharmacological evaluation of some new 6(2-Chloro 3,5-substituted phenyl)-1,2,4-triazine derivatives

6(2-Chloro 3,5-substituted phenyl)-1,2,4-triazine derivatives were synthesized and studied for evaluation of anticonvulsant activity by maximal electroshock test and neurotoxicity by rotarod motor test. Some compounds were displayed protection against induced seizures at a dose level 100 mg/kg after 0.5 and 4.0 h and while some compounds were displayed neurotoxicity at highest administered dose (300 mg/kg) after 4.0 h. Screening data revealed that one compound was found most active in series with little CNS depressant effect without neurotoxicity when compared to standard drugs Phenytoin and Carbamazepine.

This study encapsulates a comprehensive study conducted to assess the dual antibacterial and antiinflammatory properties of a newly formulated ointment containing rosmarinic acid.The investigation employed a combination of experimental and computational methodologies, encompassing antibacterial activity assays and network pharmacology analyses, to provide a comprehensive understanding of the formulation's therapeutic potential.The antibacterial activity assays were employed to evaluate the ointment's efficacy in inhibiting bacterial growth.The results showcased its significant impact on bacterial proliferation, suggesting its viability as a potent antimicrobial agent.Concurrently, a network pharmacology study was conducted to unravel the intricate molecular interactions underlying the formulation's antiinflammatory effects.This computational analysis revealed the formulation's influence on key pathways associated with immune response modulation and inflammation attenuation.The convergence of empirical data from the antibacterial activity assays and computational insights from network pharmacology provided a holistic understanding of the formulation's mechanisms of action.This integrative approach highlighted its multifaceted therapeutic effects, suggesting potential applicability in addressing bacterial infections and inflammation-related conditions.By offering targeted and efficacious interventions, these findings hold promise for advancing pharmaceutical research and therapeutic strategies.In conclusion, this study bridges experimental and computational approaches to explore the antibacterial and antiinflammatory potential of a rosmarinic acid-enriched ointment formulation.The findings underscore its multifunctional attributes and pave the way for further exploration, potentially leading to the development of innovative treatment modalities for bacterial infections and inflammatory disorders.

An updated review on Bidens Pilosa L.

Herbal drugs from natural sources like plants are in used since the traditional system of medicine; they represent a precious reservoir of innovative bioactive molecules. Among them Bidens pilosa L is one of the important plant found in all tropical and subtropical region of the world. It is the herbaceous flowering plant, having white 'petals' around a intense bunch of orange florets, and it has been reported to possess effective pharmacological properties like Antibacterial activity, Anti-inflammatory and antiallergic activity, Antimalarial Activity, T helper cell modulator, Immunosuppressive antihyperglycemic, anti-hypertensive, antiulcerogenic, hepatoprotective, anti-leukemic, anticancer, antipyretic, anti-virus, anti-angiogenic, antirheumatic, antibiotic. The Bidens pilosa L. has various chemical constituents like polyacetylenes, Polyacetylenic glycosides, aurons, auron glycosides, p-coumeric acid derivatives, caffeoylquinic acid derivatives, pheophytins, diterpenes, tannins, phytosterols, ascorbic acid, carotene, essential oils, saponins, steroids and flavonoids and many others were recognized in this plant. In this review we provide an overview mainly on the pharmacognostic characteristics, conventional uses, phytochemistry and pharmacological actions of extracts or isolated compounds from Bidens pilosa L.

Triazines – A comprehensive review of their synthesis and diverse biological importance

Triazine is a heterocyclic aromatic ring structure, with three isomers distinguished by positions of their nitrogen atoms. 1,2,3-triazine derivatives like tubercidin, toyocamycin, sangivamycin, 2-azaadenosine and 2-aza-2-desamino-5,8-dideazafolic acid are the important active moieties in pharmaceutical field while 6-azacytosine, 6-azauracil, azaribine, tirapazamine, dihydromethyl furalazine, vardenafil, apazone, lamotrigine, ceftriaxone, pymetrozine, fervenulin (planomycin), reumycin and toxoflavin (panthothricin) are 1,2,4-triazine moieties used in clinical practices. The triazine ring structure is also found in naturally occurring antibiotics like fervenulin, reumycin and toxoflavin. 1,3,5-Triazine isomer or s-triazine is an oldest known organic compound, broadly used as a lead structure in ammeline, aceto-guanide, acetoguanamine, cyanuric acid and melamine. Some s-triazine containing drugs are hexamethylmelamine (altretamine), 2-amino-4-morpholino-s-triazine, hydroxymethyl-pentamethyl melamine, triethylenemelamine (tretamine), dioxadet, irsogladine, cycloguanil, almitrine, S9788 and DW1865. Triazines have a high significance in the field of pharmaceutical chemistry with wide-spectrum of pharmacological activities so useful for design and formation of novel drugs. Some triazine analogues are recently screened in clinical trials which may lead to potent type drugs with no side effects as presently available pharmacological agents. This review outlines the biological importance and synthesis of various types of triazine derivatives from various heterocyclic and drugs containing triazine moiety.

Safety of opicapone use in Parkinson’s disease: review of literature and real-world safety data

Parkinson's disease (PD) presents a significant global health concern with increasing prevalence and burden. This review article examines the safety data of drug Opicapone from literature and real-world sources of safety data, highlighting the need for vigilance over anti-parkinsonian drugs. For literature review, the PubMed database was mined which concluded that Opicapone, a selective COMT inhibitor for PD, demonstrated ADR like dyskinesia, nausea, vomiting, headache, and constipation where on the other hand real-world evidence showed no safety alerts or recalls initiated by regulatory authorities for Opicapone, but known risks like dyskinesia and hallucination were identified. However, clinical trial statistics revealed hypertension as the most probable ADR, alongside constipation, dizziness, dyskinesia, headache, somnolence, and hyperhidrosis. Whereas, unlisted events, such as somnolence, hyperhidrosis, pulmonary embolism, and abdominal pain, were also observed. Most ADRs belonged to the nervous system disorders and gastrointestinal disorders class which can be attributed to the pharmacological effect of Opicapone. It was concluded from this review that there is need for continuous safety vigilance for Opicapone use as despite the presence of well-structured safety guidance, new risks can emerge.
