Physiological and transcriptomic analysis of OsLHCB3 knockdown lines in rice

Abstract The photosystem II (PSII) outer antenna OsLHCB3 protein plays critical roles in binding PSII chlorophyll, absorbing sunlight and transfering the excitation energy for photosynthesis. Here, OsLHCB3 knockdown mutants were produced using the RNAi system. Phenotypic analyses showed that OsLHCB3 knockdown led to pale green leaves and lower chlorophyll contents at both tillering and heading stages. In addition, mutant lines exhibited decreased non-photochemical quenching (NPQ) capacity and net photosynthetic rate (Pn) by downregulating the expression of PSII-related genes. Moreover, RNA-seq experiments were performed at both tillering and heading stages. The diferentially expressed genes (DEGs) mainly involved in chlorophyll binding, response to abscisic acid, photosystem II, response to chitin, and DNA-binding transcription factor. Besides, our transcriptomic and physiological data indicated that OsLHCB3 was essential for binding chlorophyll, but not for the metabolism of chlorophyll in rice. OsLHCB3 RNAi knockdown plants affected the expression of PS II-related genes, but not PS I-related genes. Overall, these results suggest that OsLHCB3 also plays vital roles in regulating photosynthesis and antenna proteins in rice as well as responses to environment stresses.

Mutant KRAS and CK2 Cooperatively Stimulate SLC16A3 Activity to Drive Intrahepatic Cholangiocarcinoma Progression

Abstract Intrahepatic cholangiocarcinoma (iCCA) is a lethal malignancy affecting the liver and biliary system. Enhanced understanding of the pathogenic mechanisms underlying iCCA tumorigenesis and the discovery of appropriate therapeutic targets are imperative to improve patient outcomes. Here, we investigated the functions and regulations of solute carrier family 16 member 3 (SLC16A3), which has been reported to be a biomarker of poor prognosis in iCCA. High SLC16A3 expression was enriched in KRAS-mutated iCCA tumors, and mutant KRAS elevated SLC16A3 expression via the PI3K/AKT/mTORC1/HIF1α pathway. SLC16A3 not only enhanced glycolysis but also induced epigenetic reprogramming to regulate iCCA progression. Phosphorylation of SLC16A3 at S436 (p-S436) was vital for its oncogenic function and was linked to iCCA progression. Casein kinase 2 (CK2) directly phosphorylated SLC16A3 at S436, and CK2 inhibition with CX-4945 (silmitasertib) reduced the growth of KRAS-mutated iCCA tumor xenografts and patient-derived organoids. Together, this study provides valuable insights into the diverse functions of SLC16A3 in iCCA and comprehensively elucidates the upstream regulatory mechanisms, providing potential therapeutic strategies for iCCA patients with KRAS mutations.

Low Temperature Storage and in vitro Pollen Germination of Selected Spring Wheat Accessions

The study was conducted to evaluate in vitro pollen germination of 50 spring wheat (Triticum aestivum L.) accessions at three storage temperatures (23 °C, 5 °C, and -20 °C). Germination of the mature pollen was measured right after harvest (0 time), and 24, 48, and 72 h after storage (HAS). Differences in fresh pollen germination between accessions were significant and ranged from 21.15% to 1.09%. Pollen germination was the lowest at 24 h at room temperature. The pollens of 24 wheat accessions (AAC Scotia, AW636, AW725, AW780, AW804, AW822, AW823, Brookfield, BRS Pardela, ECSW05, ECSW38, ECSW48, ECSW49, ECSW69, Hoffman, Kleos, Major, Nass, 12NQW-237, 12NQW-413, 12NQW-414, 12NQW-436, 12NQW-754, and 12BW0374) were viable after 24 h when they were stored at 5 °C. Sub-zero temperature did not prolong pollen viability.
