Abstract The PLCG1 gene, which encodes the phospholipase C γ1 isoform, is located within the commonly deleted region of the long arm of chromosome 20 (del(20q)) observed in myelodysplastic syndromes (MDS). Phospholipase C is involved in diverse physiological and pathological cellular processes through inositide signaling. We hypothesized that reduced PLCG1 expression because of haploinsufficiency by del(20q) plays a role in the molecular pathogenesis of MDS. Therefore, we analyzed PLCG1 expression in bone marrow mononuclear cells at diagnosis in 116 MDS patients with or without del(20q) by quantitative RT‐PCR to evaluate its clinical significance. The expression level of PLCG1 was significantly lower not only in MDS patients with del(20q) but also in those without del(20q) compared to that of the controls, which suggests that reduced PLCG1 expression is a common molecular event in MDS. Patients in the lowest quartile (Q4) group for PLCG1 expression had lower overall survival (OS) compared to that of other patients (Q1‐Q3) (log‐rank test, P = .0004) with estimated median OS times of 22 in the Q4 group and 106 months in the Q1‐3 group. Univariate and multivariate analysis indicated reduced PLCG1 expression (Q4) was associated with lower OS (hazard ratio 2.58, 95% CI 1.35‐4.84, P = .0049), which suggests that reduced PLCG1 expression is an independent prognostic factor for OS. In addition, patients were well‐stratified for OS by combining PLCG1 expression level (Q4 vs Q1‐3) and bone marrow blast percentage (5% or more vs less than 5%). Thus, the level of PLCG1 expression at time of diagnosis is a prognostic biomarker for MDS.
Human follicular fluid (HFF), which is composed by essential proteins required for the follicle development, provides an important microenvironment for oocyte maturation. Recently, overweight status has been considered as a detrimental impact factor on oocyte maturation, but whether HFF proteome could provide protein markers for assessing overweight-based oocyte maturation deficiency is still unknown.To reveal the HFF-based molecular characteristics associated with abnormal oocyte maturation, an iTRAQ-based comparative proteomic analysis was performed to investigate different HFF protein expression profiles from normal weight women and overweight status women.Two hundred HFF proteins were quantified in our data, of which 43% have not been overlapped by two previous publications. Compared with the HFF proteins of normal weight women, 22 up-regulated HFF proteins and 21 down-regulated HFF proteins were found in the overweight status women. PANTHER database showed these altered HFF proteins participated in development, metabolism, immunity, and coagulation, and STRING database demonstrated their complicated interaction networks. The confidence of proteomic outcome was verified by Western blot analysis of WAP four-disulfide core domain protein 2 (WFDC2), lactotransferrin (LTF), prostate-specific antigen (KLK3), fibronectin (FN1), and glyceraldehyde 3-phosphate dehydrogenase (GAPDH). Further, ELISA assay indicated WFDC2 might be a potentially useful candidate HFF marker for the diagnosis of oocyte maturation arrest caused by overweight status.Our work provided a new complementary high-confidence HFF dataset involved in oocyte maturation, and these altered HFF proteins might have clinical relevance and diagnostic and prognostic value for abnormal oocyte maturation in overweight status women.
OBJECTIVE To analyze the morbidity,pathogens,risk factors and prevention management measures.METHODS The data of the infected patients,reasons,bacteria isolates and antibiotic susceptibity tests were retrospectively analyzed.RESULTS A total of 277 patients(11.32%) among the 2248 patients during 3 years were infected in the hospital.The frequency of hospital infecton was 13.44%.Respiratary tract infection(57.14%) was the most common part,31.00% in upper respiratory tract infection and 26.14% in lower respiratory tract.G-bacilli(78.56%) were the predominant bacteria.G+ cocci(47.06%) were the predominant bacteria isolated from meningitis patients.CONCLUSIONS The hospital infection rate in neurosurgery department is higher than other departments.The risk factors of hospital infection are invasive procedure,antibiotic usage and hypoimmunity.
Objective
To evaluate the feasibility and efficacy of transcatheter arterial embolization with tissue adhesive for the treatment of acute gastrointestinal arterial hemorrhage.
Methods
Endovascular embolization performed in 24 patients (15 male and 9 female, aged 28–87, mean age, 41 years) with angiographic appearance of contrast extravasion, pseudoaneurysm, enlarged and tortuous vessels was retrospectively analyzed. Upper gastrointestinal hemorrhage was found in 14 patients and lower gastrointestinal hemorrhage in 10 patients. Transcatheter superselective embolization was performed with the mixture of N-butyl cyanocrylate (NBCA) and iodized oil. Among them, 4 patients who were not cured by embolization with coils and embolic particles were transferred to our hospital, receiving transcatheter arterial embolization with tissue adhesive. We chose tissue adhesive as the embolic agents of first choice in the remaining 20 patients. The technical and clinical success rate, rebleeding rate and complications were summarized.
Results
The technical success rate was 100.0% (24/24). The bleeding was controlled in 21 patients who had no rebleeding during follow-up period and the clinical success rate was 87.5%(21/24). Rebleeding occurred in two patients, one was managed with emergency surgery and the other underwent the second embolization therapy. Mild abdominal pain occurred in 20 patients with spontaneous symptom alleviation within 24 hours after embolization. One patient died of disseminated inravascular coagulation at 20 hours after the procedure. Of 13 cases with coagulation dysfunction, embolization was successful in 12 patients (92.3% ) without rebleeding. Neither ectopic embolism nor severe complications such as intestinal necrosis was observed.
Conclusion
Transcatheter superselective embolization with the mixture of NBCA and iodized oil is a safe and effective treatment for acute gastrointestinal arterial hemorrhage.

The incidence of cutaneous squamous cell carcinoma (cSCC) has been increasing in recent years. Meanwhile, microRNAs (miRNAs) have been found to play vital roles in various cancers, including cSCC. This study aimed to investigate the expression of microRNA-573 (miR-573) in cSCC, its relationship with long non-coding RNA PICSAR and analyze its biological role. The relationship between PICSAR and miR-573 was confirmed by dual-luciferase reporter assay and Pearson’s correlation coefficient analysis. The levels of PICSAR and miR-573 were measured using quantitative Real-Time PCR. Cell Counting Kit-8 assay was used to evaluate the cSCC cell proliferation ability. The migration and invasion abilities of cSCC cells were evaluated by Transwell assay. PICSAR expression was increased and miR-573 was decreased in tumor tissues and cSCC cell lines. PICSAR and miR-573 can bind directly, and miR-573 expression was downregulated by PICSAR in cSCC. Overexpression of miR-573 significantly inhibited the proliferation, migration and invasion abilities of A431 and SCC13 cells. Additionally, miR-573 overexpression reversed the promotion effects of PICSAR overexpression on cSCC cell proliferation, migration and invasion abilities. In conclusion, our findings indicated that miR-573 expression was decreased in tumor tissues and cSCC cells and was downregulated by PICSAR in cSCC. Additionally, miR-573 overexpression inhibited cSCC cell proliferation, migration and invasion, and reversed the promotion effects of PICSAR overexpression on cSCC cell biological functions. Thus, miR-573 might function as a tumor suppressor and might be involved in the regulatory effects of PICSAR on tumorigenesis in cSCC.
This paper reviews and summarizes, in an objective way, the status of the Japanese language education at Tsinghua University during the last 30 over years. It also gives a narration of the present condition and future prospect of this language education. The paper also describes the cause of establishing the Japanese language specialty at Tsinghua University and the developments and changes of faculties, teaching conditions and courses offered as well as achievements in teaching.
The cephalochordate amphioxus is the closest living relative to the vertebrates, with a simple chordate body plan, and a genome lacking the large-scale gene duplications, most closely approximates to that of the last common ancestor of all vertebrates. It possesses an important evolutional status and has long been regarded as the model animal for insight into the evolutionary biology and developmental biology. Here we provide a detailed review of the discovery history of amphioxus body plan, category, distribution region, living habit and their embryonic development.
