Monocentric, retrospective, cross-sectional study. To investigate the age-related normative values of sagittal parameters and establish their relationships in asymptomatic community-dwelling adults aged over 50. Degenerative changes in the spine with aging alter the sagittal balance, causing spine pathologies. However, there was a lack of comprehensive study on age-related normative values of sagittal alignment and their relationship during degenerative evolution. Among a total of 1,370 volunteers, 701 asymptomatic volunteers were recruited. Participants underwent a lateral X-ray, and sagittal parameters were assessed using a customized computer application. Age showed a stronger correlation with upper arc of thoracic kyphosis (Upper_TK) (r=0.39) compared to sagittal vertical axis (SVA) (r=0.28) and T1_slope (r=0.29). Lumbar lordosis (LL) did not exhibit a significant decrease with age. The geometrical relationship revealed a significant correlation between TK and LL (r=0.52), and LL and pelvic incidence (PI) (r=0.63), as per the equations: TK=Upper_TK+Lower arc of TK (Lower_TK), LL=Upper arc of LL (Upper_arc)+sacral slope (SS), Lower_TK≈Upper_arc, and PI=SS+pelvic tilt (PT). However, after removing intermediate control variables of Lower_TK (Upper_arc) and SS, the correlation between TK, LL, and PI was statistically weak, due to the little to no correlation between Lower_TK (Upper_arc), SS, and PT. Upper_arc (Lower_TK) was a relatively stable parameter with an age-independent value of 15.93±5.61° in elderly adults. For adults aged 60, Upper_TK significantly increased with age, while individuals over 75 exhibited a significant increase in PT and a decrease in SS and LL. Our study provided a normative age-related value of spinal sagittal parameters. To assess the spinal sagittal alignment, it was suggested that LL and TK be divided into two tangential arches based on the location of the lordotic or kyphotic apex. This study offered insights for spine surgeons in preparing for surgical corrections.

Antioxidative mechanisms and anticolitic potential of Desmodium styracifolium (Osb.) Merr. in DSS-induced colitic mice

This study aimed to determine the anticolitic effect of Desmodium styracifolium (DS) extract in ulcerative colitis (UC) mice and its possible antioxidative mechanisms. Phenolic antioxidants and free radical scavenging activities of the herb extract were determined. LCMS was used to identify phenolic compounds in the herbal extract. Ulcerative colitis in C57BL/6 SPF mice was induced with dextran sodium sulfate (DSS). The mice were treated with two doses of the DS extract during the 7-day experiment. Colon length, spleen weight, histopathological changes in colonic tissues, and disease activity index (DAI) of the mice were measured. The bioactive antioxidants in DS extract were flavonoids. The DS extract-treated UC mice had a lower DAI than the untreated UC mice. Also, the herbal treatment had downregulated expressions of proinflammatory cytokine genes in the colonic tissues of the mice compared to the control. Therefore, it is a potent anti-inflammatory agent for reducing UC symptoms.

Effect of preoperative dynamic cervical sagittal alignment on the loss of cervical lordosis after laminoplasty

Abstract Purpose Cervical laminoplasty (CLP) is a developed surgical procedure for the treatment of cervical spondylotic myelopathy (CSM), but only a few of those studies focus on preoperative dynamic cervical sagittal alignment and the study of different degrees of loss of cervical lordosis (LCL) is lacking. This study aimed to analyze patients who underwent CLP to investigate the effect of cervical extension and flexion function on different degrees of LCL. Methods In this retrospective case–control study, we analyzed 79 patients who underwent CLP for CSM between January 2019 and December 2020. We measured the cervical sagittal alignment parameters on lateral radiographs (neutral, flexion, and extension positions) and used Japanese Orthopedic Association (JOA) score to assess clinical outcomes. We defined the extension ratio (EXR) as 100 × Ext ROM (cervical range of extension)/ROM (cervical range of motion). We observed the relationships between collected variables (demographic and radiological variables) and LCL. Patients were classified into the following three groups according to the LCL: stability group: (LCL ≤ 5°); mild loss group (5° < LCL ≤ 10°); and severe loss group (LCL > 10°). We compared the differences of collected variables (demographic, surgical and radiological variables) among the three groups. Results Seventy-nine patients were enrolled (mean age 62.92 years; 51 men, 28 women) in the study. Among the three groups, cervical Ext ROM was the best in the stability group ( p < 0.01). Compared with the stability group, range of flexion (Flex ROM) was significantly higher ( p < 0.05) and EXR was significantly lower ( p < 0.01) in the severe loss group. Compared with the severe loss group, JOA recovery rates were better ( p < 0.01) in the stability group. Receiver-operating characteristic curve (ROC) analysis to predict LCL > 10° (area under the curve = 0.808, p < 0.001). The cutoff value for EXR was 16.80%, with sensitivity and specificity of 72.5% and 82.4%, respectively. Conclusion CLP should be carefully considered for patients with a preoperative low Ext ROM and high Flex ROM, as a significant kyphotic change is likely to develop after surgery. EXR is a useful and simple index to predict significant kyphotic changes.

Cervical sagittal alignment changes following anterior cervical discectomy and fusion, laminectomy with fusion, and laminoplasty for multisegmental cervical spondylotic myelopathy

Cervical sagittal alignment changes (CSACs) influence outcomes and health-related quality-of-life. Anterior cervical discectomy and fusion (ACDF), laminectomy with fusion (LCF), and laminoplasty (LP) are common treatments for multisegmental cervical spondylotic myelopathy; however, these approaches need to be compared.Our study included 167 patients who underwent ACDF, LCF, or LP. Patients were divided into four groups according to C2-C7 Cobb angle (CL): kyphosis (CL < 0°), straight (0° ≤ CL < 10°), lordosis (10° ≤ CL < 20°), and extreme lordosis (20° ≤ CL) groups. CSACs consist of two parts. CSAC from the preoperative period to the postoperative period is surgical correction change (SCC). CSAC from the postoperative period to the final follow-up period is postoperative lordosis preserving (PLP). Outcomes were evaluated using the Japanese Orthopaedic Association score and the neck disability index.ACDF, LCF, and LP had equivalent outcomes. ACDF had greater SCC than LCF and LP. During follow-up, lordosis decreased in the ACDF and LCF groups but increased in the LP group. For straight alignment, ACDF had greater CSAC and greater SCC than the LCF and LP groups but similar PLP. For lordosis alignment, ACDF and LP had positive PLP, and LCF had negative PLP. For extreme lordosis, ACDF, LP, and LCF had negative PLP; however, cervical lordosis in the LP group was relatively stable during follow-up.ACDF, LCF, and LP have different CSAC, SCC, and PLP according to a four-type cervical sagittal alignment classification. Preoperative cervical alignment is an important consideration in deciding the type of surgical treatment in CSM.

A novel metal-organic framework (MOF) with two-dimensional (2D) crystal structure was developed using Cu(NO3)2·3H2O and 2,2',5,5'-tetramethoxy-[1,1'-biphenyl]-4,4'-dicarboxylic acid. Further, its structure was characterized using infrared spectroscopy, thermogravimetry, X-ray diffraction, and X-ray crystallography. The activated Cu-MOF was used to catalyze the dehydrogenative oxidation of alcohol and N-arylation of azole compounds. Furthermore, it could be easily recovered and reused.

Bronchial epithelial transcriptomics and experimental validation reveal asthma severity-related neutrophilc signatures and potential treatments

Abstract Airway epithelial transcriptome analysis of asthma patients with different severity was used to disentangle the immune infiltration mechanisms affecting asthma exacerbation, which may be advantageous to asthma treatment. Here we introduce various bioinformatics methods and develop two models: an OVA/CFA-induced neutrophil asthma mouse model and an LPS-induced human bronchial epithelial cell damage model. Our objective is to investigate the molecular mechanisms, potential targets, and therapeutic strategies associated with asthma severity. Multiple bioinformatics methods identify meaningful differences in the degree of neutrophil infiltration in asthma patients with different severity. Then, PTPRC, TLR2, MMP9, FCGR3B, TYROBP, CXCR1, S100A12, FPR1, CCR1 and CXCR2 are identified as the hub genes. Furthermore, the mRNA expression of 10 hub genes is determined in vivo and in vitro models. Reperixin is identified as a pivotal drug targeting CXCR1, CXCR2 and MMP9. We further test the potential efficiency of Reperixin in 16HBE cells, and conclude that Reperixin can attenuate LPS-induced cellular damage and inhibit the expression of them. In this study, we successfully identify and validate several neutrophilic signatures and targets associated with asthma severity. Notably, Reperixin displays the ability to target CXCR1, CXCR2, and MMP9, suggesting its potential therapeutic value for managing deteriorating asthma.

Transcriptomic reveals the ferroptosis features of host response in a mouse model of Zika virus infection

Zika virus (ZIKV) is a neurotropic flavivirus. The outbreak of ZIKV in 2016 created a global health emergency. However, the underlying pathogenic mechanisms remain elusive. We investigated the host response features of in vivo replication in a mouse model of ZIKV infection, by performing a series of transcriptomic and bioinformatic analyses of ZIKV and mock-infected brain tissue. Tissue damage, inflammatory cells infiltration and high viral replication were observed in the brain tissue of ZIKV infected mice. RNA-Seq of the brain indicated the activation of ferroptosis pathways. Enrichment analysis of ferroptosis regulators revealed their involvement in pathways such as mineral absorption, fatty acid biosynthesis, fatty acid degradation, PPAR signaling pathway, peroxidase, and adipokinesine signalling pathway. We then identified 12 interacted hub ferroptosis regulators (CYBB, HMOX1, CP, SAT1, TF, SLC39A14, FTL, LPCAT3, FTH1, SLC3A2, TP53, and SLC40A1) that were related to the differential expression of CD8+ T cells, microglia and monocytes. CYBB, HMOX1, SALT, and SLAC40A1 were selected as potential biomarkers of ZIKV infection. Finally, we validated our results using RT-qPCR and outside available datasets. For the first time, we proposed a possible mechanism of ferroptosis in brain tissue infected by ZIKV in mice and identified the four key ferroptosis regulators.
