Can Urban Forest Settings Evoke Positive Emotion? Evidence on Facial Expressions and Detection of Driving Factors

There is increasing interest in experiences of urban forests because relevant studies have revealed that forest settings can promote mental well-being. The mental response to a forest experience can be evaluated by facial expressions, but relevant knowledge is limited at large geographical scales. In this study, a dataset of 2824 photos, detailing the evaluated age (toddler, youth, middle-age, and senior citizen) and gender of urban forest visitors, was collected from Sina Weibo (a social media application similar to Twitter in China) between 1–7 October 2018, in 12 randomly chosen cities in China. Happy and sad expressions were rated as scores by FireFACE software V1.0, and the positive response index (PRI) was calculated by subtracting sad scores from happy scores. Regional environmental factors were collected to detect driving forces using regression analyses. Happy scores were higher in forests than in urban settings, while sad scores for toddlers were lower in forests than in promenades and squares. Females showed more positive emotional expressions than males. Increases in happy scores were driven by the increase of daily minimum temperature; while PRI declined with increases in latitude. Overall, an urban forest experience can evoke positive emotions, which is likely due to comfortable feelings in warm temperatures.

Clinical significance of serum Th1/Th2 cytokines in patients with endometriosis

Endometriosis (EMS) is a challenging gynecological disorder prevalent in reproductive-aged women, and Th1/Th2 cytokines are implicated in EMS progression. This study probed the serum levels and clinical values of Th1/Th2 cytokines in EMS patients. Firstly, the clinic characteristics of EMS and control patients were recorded. The levels of interferon (IFN)-γ, interleukin (IL)-2, IL-4, and IL-10 in the serum of EMS and control patients were identified, respectively. The correlations between Th1 and Th2 cytokines and the diagnostic values of these cytokines in EMS were analyzed. We observed that EMS patients had obvious differences from the controls in dysmenorrhea, dyspareunia, pelvic pain, nulliparous, and CA125 levels. Serum IFN-γ and IL-2 were lower while IL-4 and IL-10 were higher in EMS patients. Serum IFN-γ, IL-4 were negatively correlated with serum IL-2, and IL-10 in EMS patients. Th1/Th2 cytokines may help the diagnosis of EMS. Serum IFN-γ and IL-2 were independent protective factors for EMS while dysmenorrhea, dyspareunia, nulliparous, and serum IL-4 and IL-10 were independent risk factors for EMS. Collectively, serum Th1/Th2 cytokine levels helped the diagnosis of EMS, with IFN-γ and IL-2 serving as independent protective factors whilst IL-4 and IL-10 serving as independent risk factors.

The Mechanisms of Resistin-Like Molecule-β-Mediated Airway Inflammation in Chronic Obstructive Pulmonary Disease via Autophagy

Background: The role of irreversible airway inflammatory damage in chronic obstructive pulmonary disease (COPD) progression is evident. Autophagy is an essential process in the cellular material metabolic cycle, and a family of resistant vegetative molecules may be involved in the COPD autophagic process. In this study, we investigated the mechanism of resistin-like molecule β (RELMβ) in COPD smoking-induced autophagy. Methods: Firstly, the expression differences of RELMβ and autophagy markers between COPD and control groups were analyzed in the Gene Expression Omnibus (GEO) datasets and clinical specimens. Secondly, in vitro and in vivo experiments were conducted using immunoblotting, immunofluorescence, immunohistochemistry, and other methods to investigate the mechanism by which RELMβ promotes airway inflammation through autophagy in a cigarette smoke extract-induced 16HBE cell inflammation model and a cigarette smoke-induced COPD-like mouse model. In addition, immunoprecipitation was used to analyze the binding of RELMβ to the membrane protein TLR4. Results: The expression of RELMβ and autophagy genes p62 and LC3B in lung tissue of COPD patients was significantly increased. RELMβ can mediate the activation of autophagy in 16HBE cells, and through autophagy, it increases the expression of inflammatory cytokines in a cigarette smoke extract-induced 16HBE cell inflammation model. RELMβ promotes cigarette smoke-induced COPD-like mouse airway inflammation through autophagy, and RELMβ can mediate signal transduction through the cell membrane receptor TLR4. Conclusion: The RELMβ binds to TLR4 to encourage signal transduction and that RELMβ can promote inflammation in smoky COPD lungs through autophagy. Testing and its clinical significance of plasma neutrophil gelatinase-associated lipocalin in patients with acute pancreatitis

Objective To quantify the concentration of peripheral blood plasma neutrophil gelatinase-associated lipocalin (NGAL) in patients with acute pancreatitis (AP) and to explore its value in assessment of the severity of AP.Methods From June 2011 to March 2012,83 patients with AP were selected,among those 43 cases were mild acute pancreatitis (MAP) and 40 were severe acute pancreatitis (SAP).The control group included 30 healthy individuals.The peripheral blood of patients with AP and healthy controls was collected,and plasma was isolated after centrifuged.The concentration of NGAL in plasma was detected by enzyme-linked immunosorbent assay (ELISA).The t-test was performed for comparison between groups.The correlation between the concentration of NGAL in plasma and clinical parameters of AP was analyzed by Spearman rank order correlation analysis.The diagnosis value of the concentration of NGAL in SAP was evaluated by receiver operating characteristic (ROC) curve and area under the curve (AUC).Results The concentration of plasma NGAL in AP group ((10.30± 5.97)nmol/L) was higher than that in healthy control group ((1.94±1.35) nmol/L) and the difference was statistically significant (t=11.924,P＜0.01).The concentration of plasma NGAL in SAP group ((14.61 ±5.28) nmol/L) was higher than that in MAP group ((6.27±-3.09) nmol/L) and healthy control group,the differences was statistically significant (t=8.677 and 14.539,both P＜0.01).The concentration of plasma NGAL of AP patients was positively correlated with acute physiology and chronic health evaluation-Ⅱ score,Ranson score,bedside index for severity in acute pancreatitis score,computed tomography (CT) severity index,C-reactive protein,white blood cells and the days of hospitalization (r=0.651,0.556,0.514,0.620,0.320,0.458 and 0.346,all P＜0.05).The area under the ROC curve of plasma NGAL concentration in diagnosis of SAP was 0.926 (95％CI:0,870-0.983).The cutoff value of plasma NGAL level in diagnosis of SAP was 8.44 nmol/L.The sensitivity and specificity was 87.5 ％ and 88.9％,respectively.Conclusions Plasma NGAL level is correlated with the severity of patients with AP.NGAL may be one of the markers for the early diagnosis of SAP. Identification of the key genes in chronic obstructive pulmonary disease by weighted gene co-expression network analysis

Chronic obstructive pulmonary disease (COPD) is prevalent mainly in older adults, especially those who are smokers. It appears to be regulated by multiple genes, but there is some degree of familial clustering. The evidence to date suggests that COPD-associated biomarkers are largely inadequate for disease diagnosis, so we conducted a comprehensive search for more specific genetic markers.We used 3 datasets from the Gene Expression Omnibus (GEO) database. By investigating the biological information [i.e., Gene Ontology, Kyoto Encyclopedia of Genes and Genomes and weighted gene co-expression network analysis (WGCNA)], we filtered out 8 differentially expressed genes (DEGs) and validated the transcript levels of those hub genes in 16HBE cell lines, THP-1 cell lines and lung tissue of COPD patients.The 8 hub genes comprised amyloid precursor protein (APP), fibronectin 1, insulin-like growth factor 1 (IGF1), β-actin, capping actin protein of muscle Z-line subunit alpha 2, secreted phosphoprotein 1 (SPP1), catalase (CAT), and colony stimulating factor 2 (CSF2) were selected from among the DEGs. Cigarette smoke extract-stimulated 16HBE cells were found to highly express SPP1, CSF2, and IGF1. In addition, IGF1 levels were increased and IGF1 and APP levels were decreased in CSE-stimulated THP-1 cells. SPP1 and FN1 showed increased expression levels in lung tissue of COPD patients, but the opposite held for APP and CAT.We identified 8 hub genes of COPD based on GO, KEGG and WGCNA, which have provided insights into the pathophysiological mechanisms of COPD.

Research progress of phenolic compounds regulating IL‐6 to exert antitumor effects

Cytokine therapy, which activates the host immune system, has become an important and novel therapeutic approach to treat various cancers. Recent studies have shown that IL‐6 is an important cytokine that regulates the homeostasis in vivo. However, excessive IL‐6 plays a pathological role in a variety of acute and chronic inflammatory diseases, especially in cancer. IL‐6 can transmit signals through JAK/STAT, RAS /MAPK, PI3K/ Akt, NF‐κB, and other pathways to promote cancer progression. Phenolic compounds can effectively regulate the level of IL‐6 in tumor cells and improve the tumor microenvironment. This article focuses on the phenolic compounds through the regulation of IL‐6, participate in the prevention of cancer, inhibit the proliferation of cancer cells, reduce angiogenesis, improve therapeutic efficacy, and reduce side effects and other aspects. This will help to further advance research on cytokine therapy to reduce the burden of cancer and improve patient prognosis. However, current studies are mostly limited to animal and cellular experiments, and high‐quality clinical studies are needed to further determine their antitumor efficacy in humans.

Background: Elevated estradiol (E 2 ) levels are an inevitable outcome of the controlled ovulation hyperstimulation.However, the effect of this change on pregnancy is still uncertain.Our study aimed to analyze the impact of increased serum E 2 at the day of human chorionic gonadotropin (hCG) administration on the clinical outcomes of women with fresh embryo transfer (ET) cycles.Methods: This study included 3,009 fresh ET cycles from October 2015 to September 2021.Based on the stage of embryos transferred, these cycles were categorized into the cleavage group and blastocyst group.Both groups were then divided into four sets according to E 2 levels when hCG was administered: set 1 (E 2 ≤ 2,000 pg/ml), set 2 (E 2 = 2,001-3,000 pg/ml), set 3 (E 2 = 3,001-4,000 pg/ml), and set 4 (E 2 > 4,000 pg/ml).The primary outcome was the clinical pregnancy rate (CPR).Binary logistics regression analysis was established to explore the association between CPR and E2 levels.Specifically, the threshold effect of serum E2 on CPR was revealed using the two-piecewise linear regression analyses.Results: The multivariate regression model in the cleavage group showed that patients' CPR in set 4 was 1.59 times higher than those in reference set 1, but the statistical difference was insignificant (P = 0.294).As for the blastocyst group, patients in set 4 had a lower CPR with adjusted ORs of 0.43 (P = 0.039) compared to patients in set 1. The inflection point for the blastocyst group was 39.7 pg/dl according to the results of the two-piecewise linear regression model.When E 2 levels were over the point, the CPR decreased by 17% with every 1 pg/dl increases in serum E 2 (adjusted OR = 0.83, 95% CI [0.72-0.96],P = 0.012).Conclusions: Elevated E 2 levels (>39.7 pg/dl) on hCG trigger day were associated with decreased CPR in patients with fresh blastocyst ET.However, it had no similar effect on the CPR of patients with fresh cleavage-stage ET.

The relationship between parental parenting stress and children 's anger-aggression and anxiety-withdrawal: the mediating effect of parenting efficacy

This study aimed to explore the relationships between parenting stress and children 's anger-aggression and anxiety-withdrawal, and further investigate the underlying moderated mediation mechanism. Methods: Parenting Stress Scale, Parenting Efficacy Scale, and Children 's Social Ability and Behavior Rating Scale were used to conduct a questionnaire survey on 525 parents of 2-5-year-old children in five kindergartens in Hunan, Jiangxi and Hainan. Results: (1) Father / mother parenting stress positively related to children 's anger-aggression ( rfather = 0.27, P < 0.01 ) ; rmother = 0.52, P < 0.01 ) and anxiety-withdrawal ( rfather = 0.31, P < 0.01 ) ; rmother = 0.50, P < 0.01 ). (2) Parental parenting efficacy mediated the relationships between parenting stress and children 's anger-aggression ( βfather = 0.05, 95 % CI [ 0.023, 0.086 ] ; βmother = 0.05, 95 % CI [ 0.032, 0.085 ] ) and anxiety-withdrawal ( βfather = 0.05, 95 % CI [ 0.025, 0.090 ] ) ; βmother= 0.07, 95 % CI [ 0.046, 0.103 ] ). Conclusion: Parenting stress is related with increased children 's anger-aggression and anxiety-withdrawal through decreasing parenting efficacy, which provides a new insight to alleviate the negative outcome of parenting stress.

Limonin ameliorates acute pancreatitis by suppressing JAK2/STAT3 signaling pathway

Acute pancreatitis (AP) is one of the most common acute abdomen of digestive system and has the characteristics of dangerous condition and rapid development. Limonin has been confirmed to hold anti-inflammatory and antioxidant effects in various diseases. However, its potential beneficial effect on AP and the concrete mechanisms have never been revealed. Here, two mouse models were used to investigate the protective effects of limonin on AP, the caerulein-induced mild acute pancreatitis (MAP) model and L-arginine-induced severe AP (SAP) model. Firstly, it was found that limonin administration attenuated lipase and serum amylase levels and ameliorated the histopathological manifestations of pancreatic tissue in a dose-dependent manner. Additionally, the amelioration of AP by limonin was associated with reduced levels of inflammation initiators (IL-6, IL-1β, CCL2, and TNF-α). Mechanistically, we found that limonin suppressed the Janus Activating Kinase 2 (JAK2)/Signal Transducer and Activator of Transcription 3 (STAT3) signaling pathway, as evident by the decreased levels of JAK2 and p-STAT3. And activation of JAK2 using JAK2 activator rescued the protective effects of limonin on AP. Thus, our results demonstrate that limonin can ameliorate AP in two mice models via suppressing JAK2/STAT3 signaling pathway.
