Abstract Ethylene response factors (ERF) are members of the APETALA2/ERF transcription factor family, and they play an important role in plant growth, development, and response to various environmental stresses. In the present study, an ERF transcription factor HcERF4 was isolated and characterized from kenaf. The protein encoded by the HcERF4 has 233 amino acid residues with a theoretical isoelectric point of 8.89 and a predicted molecular weight of 25.53 kDa. HcERF4 had an over than 86.97% identity to HsERF4 (XP_039019980.1), and shared a closest phylogenetic relationship with Hibiscus syriacus . Subcellular localization analysis shows that HcERF4 is located in the nucleus. Transactivation assays in yeast demonstrated that HcERF4 functions as a transcriptional activator. The expression of HcERF4 was enriched in leaf and root, and can be induced by salt or drought treatments in kenaf. The VIGS-silenced HcERF4 plant showed significantly reduced plant height, stem diameter, fresh weight, and relative water content (RWC) compared with wild type plants under salt or drought stress condition; In addition, the contents of MDA, O 2 − , H 2 O 2 , and free proline is significantly increased, and the activities of SOD and CAT are significantly reduced. The DAB/NBT staining results showed that the H 2 O 2 and O 2 − contents in HcERF4 -silenced plants were consistent with the determination. Based on these results, it is proposed that HcERF4 plays an important role in regulating salt and drought stress in kenaf.

To understand the current status and clinical relevance of early (<24 weeks) glucose challenge test (GCT) in pregnant women with risk factors of gestational diabetes mellitus (GDM) in China.Data from the survey of incidence of GDM in China were re-analyzed. The incidence of abnormal glucose metabolisms and the rate of early GCT in all women were calculated according to different numbers of risk factors. Sixteen risk factors were included in the survey. However, 4 independent risk factors were considered separately in this re-analysis. The ADA criteria for GDM diagnosis were applied.A total of 16 286 pregnant women were included in this analysis and 64.3% (10 468) presented with at least one risk factor. The incidence of GDM became elevated with the increasing number of risk factors (P < 0.001). Early GCT was performed in 1687 (16.1%) pregnant women and the early detected GDMs only accounted for 11.9% of all GDMs among those with at least one risk factor. Among those who had early GCT, the GDM diagnosis rate increased with the number of risk factors (P < 0.001). Previous analysis in this survey identified 4 independent risk factors for GDM among 16 risk factors: BMI > or = 24, age over 30 years old, family history of DM and southerners. Similar analysis was performed according to the above 4 risk factors and similar results were found as those found for 16 risk factors. No significant difference was found in the GDM and GIGT incidence between the two analyses in those with at least one risk factor.Early GCT is necessary for pregnant women with risk factors of GDM, but the screening rate in China is low. GCT should be repeated for those women with risk factors of GDM and normal GCT at early screening.
Loss of imprinting (LOI) of insulin-like growth factor II gene (IGF2) is anepigenetic abnormality associated with human diseases. However, little is known about thecharacteristics of IGF2 imprinting in newborn cord blood cells. METHODS: A total of 923cord blood samples from term singletons and related clinical data were collected; IGF2imprinting status in 273 specimens were successfully analyzed using RT-PCR andrestriction fragment length polymorphism. RESULTS: LOI of IGF2 was detected in 20.9%of informative samples. The mean birth weights (BW) in the LOI and the normal imprintinggroups were 3462.7 ± 460.2 g and 3363.7 ± 427.7 g, respectively. The abdominal perimetersin the LOI group tended to be larger than that in the normal imprinting group. Pregnancycomplications, delivery modes, newborn diseases, occurrences of malignant tumors ingrandparents, and other maternal factors were not associated with LOI of IGF2. 22.2% ofthe infants with IGF2 LOI also showed LOI in their father’s lymphocytes while 21.4% intheir mother’s lymphocytes. CONCLUSIONS: About 20% of Han Chinese newbornsindicated LOI of IGF2 in their cord blood lymphocytes that may represent the epigeneticcharacteristics in this ethnic group. While IGF2 LOI tends to be weakly inherited between parents and offspring, abnormal imprinting seems to be statistically unrelated with phenotypes of newborns, although it might have an association with later phenotypes of infants.
Objective: To observe morphological characteristics and in-vitro maturation ratio of all metaphase I arrested oocytes in IVF-ET cycles.Methods: In two IVF-ET cycles,maturation culture in vitro was performed and we evaluated morphological characteristics of all oocytes arrested in metaphase I.Results: Metaphase I arrested oocytes presented with uniform cytoplasm,compact zone and no space between oocyte and zone.Incompact association was observed between granule cell and zone without radiated structures.There was no markedly alterations in morphological characteristics after 24,48,72 hours of maturation culture,respectively.All oocytes were arrested in MetaphaseⅠ and no polar growed.Conclusion: All metaphase I arrested oocytes possess of uniqe morphological characteristics and fail to mature following current culture assay in IVF-ET cycles.Natl J Androl,2005,11(11):838-840
