Zinc-α2-glycoprotein (ZAG) is decreased in neurons of epilepsy patients and models. Either insulin or overexpressing ZAG suppresses seizure and epileptiform discharges. ZAG is known to influence insulin sensitivity of tissues, but whether insulin regulates ZAG is unknown. This study investigated the effect and mechanism of insulin on ZAG expression and epileptiform discharge-induced oxidative stress. Primary cultured cortical neurons were treated with insulin, AXL1717 (inhibitor of insulin-like growth factor-1 receptor), or BMS-754807 (inhibitor of both insulin receptor and insulin-like growth factor-1 receptor). Mg2+-free epileptiform discharge model was also made. Levels of ZAG and AZGP1 mRNAs in neurons were measured. Oxidative stress in Mg2+-free-treated treated neurons underwent AZGP1 knock-down, AZGP1 overexpression, or insulin treatment was determined. Insulin treatment increased ZAG expression in neurons; this insulin-induced ZAG increase was abolished by either AXL1717 or BMS-754807. Either insulin treatment or ZAG overexpression suppressed epileptiform discharge-induced oxidative stress in neurons. Knock-down of ZAG abolished the antioxidative stress effect of insulin. Insulin-induced ZAG increase in neurons was mainly related to the activation of insulin-like growth factor-1 receptors. Insulin presented its antioxidative stress effect in neuronal epileptiform discharge models by increasing ZAG.
Resting tremor in Parkinson's disease (PD) is associated with the activity in the basal ganglia and cerebello-thalamo-cortical circuits/network. However, most insights stem from functional MRI research, and structural studies, which can provide basis for and constrain functional activity, remains limited. We investigated the structural change in PD patients with resting tremor (PD-WR) from a network perspective. 42 early-stage PD-WR, 27 PD patients without resting tremor (PD-NR), and 56 healthy controls (HC) were included. PD-WR showed lower cortical thickness in several motor-related lobules. Compared to HC, significant atrophy was found in right lobule VIIA (t = -3.076, p = 0.016, Cohen's d = 0.627), left lobule VI (t = -3.323, p = 0.007, Cohen's d = 0.678), and right lobule VI (t = -3.052, p = 0.017, Cohen's d = 0.623) in PD-WR. Compared to PD-NR, left lobule V also had a significant reduction (t = -2.958, p = 0.023, d = -0.657). PD-WR had higher fractional anisotropy in cerebello-cortical connection compared to HC (t = 3.209, p = 0.009, d = 0.926), with reduced radial (t = -2.561, p = 0.046, d = 0.739) and mean (t = 2.614, p = 0.046, d = 0.871) diffusivity compared to PD-NR. At the network level, better hierarchy (rho = 0.598, p = 0.004), small-worldness (rho = 0.621, p = 0.003), and increased nodal involvement of the thalamus (rho = 0.718, p = 0.031) and motor cortex (rho = 0.660, p = 0.055) were positively correlated with tremor amplitude. Our study supports the alternation of the cerebello-thalamo-cortical circuit in PD-WR. However, further research with other forms of PD, a wide range of disease stage and larger sample size is needed.
Objective To analyze the importance of collaborative public management in health management from theoretical level,to provide useful reference for the deepening medical reform. Result Collaborative public management has become a new strategy of local government in the management of public affairs in the new period. In contrast,China's management of public health cooperation is not smooth,efficiency is not high,the overall advantages of cooperative health resources is to be excavated,and the relevant aspects of cooperation needs to be strengthened. Conclusion Under the condition of the relative shortage of health resources in our country and the health service demand of urban and rural residents increasing rapidly,a new mechanism for networked collaborative public management should be established in the relevant departments within the governments,departments and health systems,to maximize the existing health resources,and to better protect the health of urban and rural residents.

Mesenchymal stem cell (MSC) transplantation is a potential treatment for inflammatory bowel disease. The PI3Kγ pathway, which is affected by drugs, is involved in inflammation. Considering the possible interaction between MSCs and PI3Kγ-associated drugs, we evaluated the efficacy and mechanism of MSCs in the treatment of colitis in PI3Kγ-/- mice. Trinitro-benzene-sulfonic acid enema was used to create a colitis model, and MSCs were transplanted through the caudal vein to treat colitis in wild-type and PI3Kγ-/- mice. We sequenced microbial 16S rRNA genes in the colonic mucosa of PI3Kγ-/- and wild-type mice and quantified the levels of colonic IgA, IL-2, IL-10, IL-17A, occludin, and serum IgA. Transplantation of MSCs led to more serious decreased weight of trinitro-benzene-sulfonic acid-administered PI3Kγ-/- mice compared to wild-type mice. The disease activity index, pathological scoring, number of taxa in the colon, Berger–Parker index, I-index, proportion of Proteobacteria , and IgA level in the blood were higher in PI3Kγ-/- mice than in wild-type mice after MSC transplantation. Occludin and IL-10 levels in the colon tissues were decreased before and after transplantation of MSCs into PI3Kγ-/- mice, whereas these proteins were increased in wild-type mice. IL-17 level was decreased in both wild-type and PI3Kγ-/- mice, with knockout mice showing a greater decrease. Therefore, transplantation of MSCs in PI3Kγ-/- mice led to increased numbers of exogenous pathogenic microorganisms and colitis that was difficult to relieve.
Mincer's human capital earnings function has been widely used in empirical studies for estimating the rate of return to education. However,OLS estimates of Mincer's function potentially suffer from omitted variables,measurement error,sample selectivity bias, heterogeneity and so on. Many econometric approaches are developed correspondingly to reduce the above-mentioned problems. This paper aims to conduct a thorough review of the results associated with those corrective approaches in order to provide an accurate estimate of China's rate of return to education.
