Spectrum of Multiple Congenital Anomalies-Hypotonia-Seizures Syndrome 3 (MCAHS3) Due to Phosphatidylinositol Glycan Biosynthesis Class T (PIGT) Gene Mutations: A Narrative Review

Multiple congenital anomalies-hypotonia-seizures syndrome 3 (MCAHS3) results from mutations in the phosphatidylinositol glycan biosynthesis class T (PIGT) gene leading to defects in glycosylphosphatidylinositol transamidase complex (GPI-TA) synthesis. Glycosylphosphatidylinositol serves as an anchor to more than 150 mammalian proteins for attachment on cell surfaces, enabling specific functional properties. Mutations in the PIGT gene result in disruption of this extremely important post-translational protein modification, yielding dysfunctional proteins leading to MCAHS3. An exhaustive literature search was conducted across various electronic databases to reveal only 41 reported cases of MCAHS3 worldwide, emphasizing the rarity of this condition. Multiple congenital anomalies-hypotonia-seizures syndrome 3 has been reported as secondary to 18 different known PIGT variants to date, manifesting as a varying spectrum of craniofacial dysmorphism, developmental delay with epilepsy, cardiac and renal malformations, and unique features in biochemical testing and neuroimaging. This review aims to highlight the constellation of clinical symptoms, diagnostic modalities, and management challenges associated with MCAHS3 cases. It would help determine optimal diagnostic and treatment strategies for newly identified cases and facilitate new research on this rare condition.

Advances in innovative seed potato production systems in India

India is the second largest producer of potatoes in the world. Seed is the single most important input in potato cultivation. High seed rate (2.5–3.0 tons/ha), low rate of multiplication, progressive viral degeneration, storage, and transportation are major issues of potato seed production in the country. Potato seed alone accounts for 40%-50% of the total potato production cost, and huge quantities of potentially edible food is put back into the soil as potato seed. The delayed penetration of new improved potato/seed varieties into farmers’ fields due to the slow multiplication rate and frequent seed replacement because of degeneration are associated issues. To circumvent these issues, continuous efforts are being made by potato researchers to develop innovative technologies for quick multiplication of initial healthy breeder’s seed of the released varieties in sufficient quantities to meet the demand in our country. A paradigm shift in potato seed production methods has taken place globally since the early 1900s. Major potato producers of the world have shifted from conventional to hi-tech seed production systems to improve the seed quality and enhance seed multiplication rate. New innovations can overcome many of the problems associated with potato seed production, particularly in tropical and sub-tropical countries. Recent advances in potato seed production systems in India and challenges ahead for seed production are described here.

In Silico Analysis of Tumor Necrosis Factor (TNF) in Homo sapiens

Tumor Necrosis Factor (TNF) encodes a pleiotropic proinflammatory cytokine that plays a major role in regulating immune cells. The cytokine involved the perception of a wide spectrum of biological processes, including cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It has also been implicated in various diseases such as autoimmune disorders, insulin resistance, cancer, etc. Considering the in-depth significance of TNF, the present study was designed for their in silico analysis in Homo sapiens. This study is undertaken to find the functional and structural basis of TNF in H. sapiens, by using various bioinformatics tools. In silico evaluation of these proteins includes physicochemical parameters, prediction of secondary structures, and validation of different models. These proteins could be directly or indirectly stabilized by the amino acid interactions and hydrogen-bond networks. It elucidated the functional deviations of protein to some extent and laid the foundation for docking and molecular modeling for future prospects.

A repository of web-based bioinformatics resources developed in India

Abstract IndiaBioDb ( https://webs.iiitd.edu.in/raghava/indiabiodb/ ) is a manually curated comprehensive repository of bioinformatics resources developed and maintained by Indian researchers. This repository maintains information about 543 freely accessible functional resources that include around 258 biological databases. Each entry provides a complete detail about a resource that includes the name of resources, web link, detail of publication, information about the corresponding author, name of institute, type of resource. A user-friendly searching module has been integrated, which allows users to search our repository on any field. In order to retrieve categorized information, we integrate the browsing facility in this repository. This database can be utilized for extracting the useful information regarding the present scenario of bioinformatics inclusive of all research labs funded by government and private bodies of India. In addition to web interface, we also developed mobile to facilitate the scientific community.

HumCFS: A database of fragile sites in human chromosomes

Abstract Genomic instability is the hallmark of cancer and several other pathologies, such as mental retardation; preferentially occur at specific loci in genome known as chromosomal fragile sites. HumCFS ( http://webs.iiitd.edu.in/raghava/humcfs/ ) is a manually curated database provides comprehensive information on 118 experimentally characterized fragile sites present in human chromosomes. HumCFS comprises of 19068 entries with wide range of information such as nucleotide sequence of fragile sites, their length, coordinates on the chromosome, cytoband, their inducers and possibility of fragile site occurrence i.e. either rare or common etc. Each fragile region gene is further annotated to disease database DisGenNET, to understand its disease association. Protein coding genes are identified by annotating each fragile site to UCSC genome browser (GRCh38/hg38). To know the extent of miRNA lying in fragile site region, miRNA from miRBase has been mapped. Comprehensively, HumCFS encompasses mapping of 5010 genes with 19068 transcripts, 1104 miRNA and 3737 disease-associated genes on fragile sites. In order to facilitate users, we integrate standard web-based tools for easy data retrieval and analysis.
