Application of grading technique in flat-shaped tea in Guizhou.

The straight rate,flat-shape rate,de-pubescence rate and broken tip rate of flat-shaped Xinqueshe tea samples with three different gradations were determined to study the effect of grading technique in the processing on quality of flat-shaped tea.The results showed that the straight rate,flat-shaped rate,de-pubescence rate and broken tip rate of flat-shaped tea using grading technique were 84.0%,76.0%,80.3% and 0.6% respectively and its flat-shaped rate,uniformity and external quality were better than non-graded tea samples significantly.

Rainbow trout (Oncorhynchus mykiss) is a cold-water species, and often affected by high temperatures.To understand hsp30 mRNA expression trend and molecular mechanism responding to high temperature stress, real-time qPCR was used to quantify hsp30 mRNA expression level in different tissues (heart, liver, gill, brain, spleen and head kidney) in control (18°C) and heat-stressed (25°C for 2, 4, 8 and 12 h) groups.Results showed that hsp30 mRNA in all tissues had already synthesized at 18°C.hsp30 mRNA expression in all tissues was significantly up-regulated and reached the peak after heat stress at 25°C for 2-4 h (P<0.05), then significantly down-regulated at 8 h (P<0.05), and finally returned to control level at 12 h and HSR (P>0.05).The highest hsp30 mRNA expression levels were recorded in heart and liver after 2-4 h.However, the most significant changes of hsp30 mRNA after 2-4 h were observed in gill by 15668.243-and11452.125-foldcompared with 18°C.These results indicated that hsp30 mRNA expression in rainbow trout was induced by heat stress and responded in a time-and tissue-specific manner.Hsp30 can provide some protection in heat stress, but it is only within a certain range.

Retracted: Bevacizumab synergises with the BCL 2 inhibitor venetoclax to effectively treat B‐cell non‐Hodgkin's lymphoma

Abstract Objectives Our present study has shown a potential role for VEGF‐A‐mediated autocrine signalling to promote survival and proliferation of SU‐DHL‐6 cells, but the cells could not undergo apoptosis but rather decrease proliferation after bevacizumab treatment. Therefore, we would like to further study the antitumour efficacy of venetoclax (BCL2 inhibitor) in combination with bevacizumab in B‐cell NHL. Methods The human cytokine antibody array, RT‐qPCR, Western blot, ELISA, apoptosis assay and xenografted mouse model et al were used. Results We described a unique phenomenon that SU‐DHL‐6 cells showed cell density‐dependent survival and growth. Then, we suggested the expression of VEGF‐A was positively correlated with the cell density using a human cytokine antibody array and indicated an important role of VEGF‐A in the survival and proliferation of SU‐DHL‐6 cells. Additionally, xenografted SU‐DHL‐6 cells formed tumours in mice that grew in response to VEGF stimulation. GEO data set also suggested that high VEGF‐A expression reflected poor prognosis. The combination therapy with bevacizumab and navitoclax could significantly induce of cell death in vitro and reduce the tumour size and weight with well tolerated in vivo. Conclusions Our findings propose a novel combined strategy in which bevacizumab synergises with the BCL2 inhibitor venetoclax that is effective against B‐cell NHL.

Objective To improve the level of aged patients' rational use of drug.Methods Taking multiple steps to supervise aged patients' use of drug.The measures included establishing and perfecting the organization,rules and regulations;taking various methods to give nursing staff chance in education and training;in order to improve the rationality of medication operation,strengthening the management in correct dealing with medical order,choosing the best medication,the best dosage form and the best time,normalizing the medication operation,avoiding the incompatibility of medicine,reinforcing health education in rational use of drug,increasing the chance of communication among doctors,nurses and pharmacists,and cooperating to monitor the drug adverse reaction and so on.Results Security and compliance of medication of aged patients has enhanced.The rate of drug treatment mistakes decreased.Nursing staff's level of knowledge regarding medication rationality has improved.Conclusion Multiple-step management strategy could increase the level of aged patients' rational use of drug.

Insights into neurotransmitter release from the structure of Munc13-1 C1C2BMUN

Munc13-1 acts as a master regulator of neurotransmitter release, mediating docking-priming of synaptic vesicles and diverse presynaptic plasticity processes.It is unclear how the functions of the multiple domains of Munc13-1 are coordinated.The crystal structure of a Munc13-1 fragment including its C1, C2B and MUN domains (C1C2BMUN) reveals a 195 Å-long multihelical structure with the C1 and C2B domains packed at one end.The similar orientations of the respective diacyglyceroland Ca2+-binding sites of the C1 and C2B domains suggest that the two domains cooperate in plasma-membrane binding and that activation of Munc13-1 by Ca2+ and diacylglycerol during short-term presynaptic plasticity are closely interrelated.Electrophysiological experiments in mouse neurons support the functional importance of the domain interfaces observed in C1C2BMUN.The structure imposes key constraints for models of neurotransmitter release and suggests that Munc13-1 bridges the vesicle and plasma membranes from the periphery of the membrane-membrane interface.Details of the hurdles faced during the challenging structure determination will also be provided, which included high crystal non-isomorphism, severe data anisotropy and low resolution.

The Epidemic of Human Immunodeficiency Virus, Hepatitis C Virus, and Syphilis Infection, and the Correlates of Sexually Transmitted Infections among Men Who Have Sex with Men in Zhenjiang, Jiangsu, China

A cross sectional survey was performed to investigate the epidemic of human immunodeficiency virus (HIV), hepatitis C virus (HCV), syphilis, and HIV-1 subtype infections among men who have sex with men (MSM) and to evaluate factors that were correlated with sexually transmitted infections (STIs) among 657 MSM in Zhenjiang, Jiangsu, during 2012-2013. Data on socio-demographic parameters and risky behaviors were collected, and blood samples were obtained. In this survey, the prevalence of STIs due to HIV (5.3%), HCV (0.6%), and syphilis (19.2%) were measured. Of 35 HIV-infected MSM, 1 (0.2%) and 17 (2.6%) were co-infected with HCV and syphilis. Among 33 samples with available HIV-1 genomic fragment sequences, 6.1% B, 72.7% CRF01_AE, and 21.2% CRF07_BC were identified. Multivariate logistic regression analysis showed that age > 30 years (odds ratio [OR] 2.002; 95% confidence interval [CI], 1.144-3.502) and having had STI symptoms in the past year (OR, 11.673; 95% CI, 3.092-44.078) were positively correlated with having an STI, while consistent use of a condom when having sex with male partners (OR, 0.621; 95% CI, 0.408-0.945) was significantly associated with a low STI rate. The worsening epidemic of HIV, HCV, and syphilis infections, and the prevalence of complex subtypes of HIV-1 and other risk factors suggest that effective intervention strategies should be strengthened for MSM in Zhenjiang, Jiangsu.

Effects of genistein on proliferation,apoptosis and invasiveness in methotrexate-resistant human choriocarcinoma JAR/MTX cells and it's mechanism

AIM:To study the effects of genistein on JAR/MTX cell proliferation,apoptosis and invasion and it's mechanism in vitro.METHODS: MTT assay,Annexin-Ⅴ and propidium iodide label analysis and invasion assay were used to determine the effects of genistein on proliferation,apoptosis and invasiveness in JAR/MTX methotrexate-resistant human choriocarcinoma cells.RT-PCR was used to estimate the relative mRNA amounts of estrogen receptor(ER),MTA3 and snail in the cells.Western blotting and gelatin zymography assay were used to estimate the relative protein amounts of MMP-2,MMP-9 and E-cadherin in the cells.RESULTS: After treatment of genistein,the proliferation and invasiveness of JAR/MTX cells were decreased significantly in a dose-dependent manner.10 μmol/L genistein induced apoptosis,whereas 25,50,100 μmol/L genistein induced apoptosis and necrosis significantly.Genistein led to an increase in ERβ,MTA3 mRNA and E-cadherin protein expression,and decreases in the amounts for snail mRNA and MMP-2 and MMP-9 protein expression of JAR/MTX cells.CONCLUSIONS: Genistein inhibits the cell proliferation by inducing cell apoptosis and necrosis.Genistein also may inhibit JAR/MTX cell invasion in part through the upregulation of E-cadherin and downregulation of MMP-2 and MMP-9.The signal transduction pathway of invasion suppression induced by genistein in JAR/MTX cells may be as follows: MTA3→snail→ E-cadherin.
