Study of Bitespiramycin Distribution in Rats and Cerebrospinal Fluid of Patients by a Sensitive LC-MS/MS Method with Rapid Sample Preparation

Bitespiramycin, has been shown to have a therapeutic effect against respiratory tract inflammation, including a potential effect against COVID-19. A current clinical trial in China showed that bitespiramycin was an effective treatment for severe pneumonia and intracranial infection. However, there is lack of an analytical method to elucidate the distribution of bitespiramycin. In this study, a highly sensitive, rapid and reliable UPLC–MS/MS method was developed to comprehensively characterize the bitespiramycin distribution in various bio-samples, which is significantly improved upon the published work. A rapid sample preparation method was developed by using n-butanol as the solvent to extract bitespiramycin from different bio-samples. The extract was then directly analyzed by UPLC–MS/MS coupled with an alkaline-resistant column after centrifugation which avoids the time-consuming concentration process under nitrogen and redissolution. The method was employed to accurately quantify bitespiramycin and its metabolites in rat plasma, tissues, and human cerebrospinal fluid. Notably, the presence of bitespiramycin and its metabolites was identified for the first time in various rat organs including brain, testis, bladder and prostate as well as in human cerebrospinal fluid. This newly developed approach shows great promise for drug distribution assays including other antibiotics and can help elucidate the ADME of bitespiramycin.

Highly Efficient Catalyzed by Imidazolium-Based Dual-Sulfonic Acid Functionalized Ionic Liquids for Liquid Phase Beckmann Rearrangement: Experiments and Cosmo-Rs Calculations

A production technique of ε-Caprolactam (CPL), including high yield, mild reaction process, must be developed on the environmentally friendly chemical industry. This technology is highly desired to design and synthesize high−performance catalysts for liquid phase Beckmann rearrangement of cyclohexanone oxime (CHO) to CPL. In this work, 3-methyl-1-(propyl-4-sulfonyl) imidazolium methanesulfonate ([PHSO3MIM] [MSA]) with highly efficient and excellent yield is synthesized successfully. In the presence of zinc chloride (ZnCl2), it exhibits the high selectivity (94%) of CPL at 90 ℃ and the reaction time is as short as 1 h. Interestingly, Fourier-transform infrared (FT-IR) investigations show that that the functional Brønsted−Lewis acidic types of ionic liquids (ILs) are formed by the uniformly distributed ZnCl2 and [PHSO3MIM] [MSA]. In addition, the hydrogen bond (H-bond) is formed between CHO and ILs. After ten reaction cycles, no significant structure changes are observed in the recovered [PHSO3MIM] [MSA]·[ZnCl2]. On the basis of the COSMO-RS model, a method is developed to predict the solubility of ILs for the first time, which indicates that [PHSO3MIM] [MSA] is considered to be one promising candidate for the liquid phase Beckmann rearrangement of CHO into CPL. Finally, a theoretical model of the H-bond interactions between ILs and CHO is further confirmed to support the advance of reaction mechanism. Therefore, this work provides a feasible way for the CPL production technique in the liquid phase Beckmann rearrangement reaction.

Cation/Ca2+ Exchanger 1 (MdCCX1), a Plasma Membrane-Localized Na+ Transporter, Enhances Plant Salt Tolerance by Inhibiting Excessive Accumulation of Na+ and Reactive Oxygen Species

High salinity causes severe damage to plant growth and significantly reduces crop yields. The CCX family proteins can facilitate the transport of multiple ions to prevent toxicity. CCX proteins play an important role in regulating plant salt tolerance, but no detailed studies on CCX proteins in apples have been reported. Here, the CCX family gene MdCCX1 was cloned from apple (Malus domestica). It is constitutively expressed in various apple tissues and is significantly induced by salt stress. As a plasma membrane-localized protein, MdCCX1-overexpression could complement the Na+-sensitive phenotype of yeast mutants and reduce the Na+ content in yeast cells under NaCl treatment, suggesting that MdCCX1 could be a plasma membrane-localized Na+ transporter. To identify the function of MdCCX1 in salt response, we transformed this gene into Arabidopsis, apple calli, and apple plants. Overexpression of MdCCX1 significantly improved the salt tolerance of these transgenic materials. The significantly reduced Na+ content under NaCl treatment indicated that MdCCX1 overexpression could enhance plant salt tolerance by inhibiting the excessive accumulation of Na+. Besides, MdCCX1 overexpression could also enhance plant salt tolerance by promoting ROS scavenging. These findings provide new insight and rich resources for future studies of CCX proteins in plant species.

Feasibility and Transferability of Transfer Learning: A Mathematical Framework

Transfer learning is an emerging and popular paradigm for utilizing existing knowledge from previous learning tasks to improve the performance of new ones. Despite its numerous empirical successes, theoretical analysis for transfer learning is limited. In this paper we build for the first time, to the best of our knowledge, a mathematical framework for the general procedure of transfer learning. Our unique reformulation of transfer learning as an optimization problem allows for the first time, analysis of its feasibility. Additionally, we propose a novel concept of transfer risk to evaluate transferability of transfer learning. Our numerical studies using the Office-31 dataset demonstrate the potential and benefits of incorporating transfer risk in the evaluation of transfer learning performance.

Crop Leaf Phenotypic Parameter Measurement Based on the RKM-D Point Cloud Method

Crop leaf length, perimeter, and area serve as vital phenotypic indicators of crop growth status, the measurement of which is important for crop monitoring and yield estimation. However, processing a leaf point cloud is often challenging due to cluttered, fluctuating, and uncertain points, which culminate in inaccurate measurements of leaf phenotypic parameters. To tackle this issue, the RKM-D point cloud method for measuring leaf phenotypic parameters is proposed, which is based on the fusion of improved Random Sample Consensus with a ground point removal (R) algorithm, the K-means clustering (K) algorithm, the Moving Least Squares (M) method, and the Euclidean distance (D) algorithm. Pepper leaves were obtained from three growth periods on the 14th, 28th, and 42nd days as experimental subjects, and a stereo camera was employed to capture point clouds. The experimental results reveal that the RKM-D point cloud method delivers high precision in measuring leaf phenotypic parameters. (i) For leaf length, the coefficient of determination (R2) surpasses 0.81, the mean absolute error (MAE) is less than 3.50 mm, the mean relative error (MRE) is less than 5.93%, and the root mean square error (RMSE) is less than 3.73 mm. (ii) For leaf perimeter, the R2 surpasses 0.82, the MAE is less than 7.30 mm, the MRE is less than 4.50%, and the RMSE is less than 8.37 mm. (iii) For leaf area, the R2 surpasses 0.97, the MAE is less than 64.66 mm2, the MRE is less than 4.96%, and the RMSE is less than 73.06 mm2. The results show that the proposed RKM-D point cloud method offers a robust solution for the precise measurement of crop leaf phenotypic parameters.

Corrosion Inhibition and Adsorption Process of 3-Amino-5-Mercapto-1,2,4-Triazole on Aluminium Alloy: Experimental and Theoretical Studies

The corrosion inhibition effect of 3-amino-5-mercapto-1,2,4-triazole (AMT) on AA2024 aluminium alloy in 3.5 wt.% NaCl solution was investigated, and the corrosion inhibition mechanism was revealed. The influence of AMT concentration on the corrosion inhibition performance was evaluated by potentiodynamic polarization curve and electrochemical impedance spectroscopy (EIS). Surface analysis and surface-enhanced Raman scattering (SERS) spectra were used to study the adsorption process and corrosion inhibition mechanism of AMT on the alloy surface. Polarization curve and EIS results showed that when the AMT concentration was 1.5 g/L, the corrosion current density ( i corr ) was the lowest and the resistance of adsorption film ( R f ) was the largest, illustrating the highest corrosion inhibition efficiency. Moreover, the adsorption kinetics process of AMT was revealed by SERS measurement, and a positive correlation between the SERS intensity and R f values of AMT after different immersion time was achieved. It indicated that the efficient adsorption of corrosion inhibitors significantly enhanced the corrosion inhibition performance. Density functional theory (DFT) and molecular dynamics simulations were used to give further insight into the adsorption and inhibition mechanism of AMT on the aluminium alloy surface.

Effects of variable maternal temperature on offspring development and reproduction of Rhopalosiphum padi, a serious global pest of wheat

1. Global warming is occurring at an unprecedented rate. Information about how variable temperature affects insect life‐history traits is still scarce. 2. The current study first evaluated the effects of two variable‐temperature treatments [high‐temperature‐variation (HT) treatments and mild‐temperature‐variation (MT) treatments] on the life‐history traits of a maternal generation (F 0 ) of Rhopalosiphum padi , a serious global pest, using a constant normal‐temperature (NT) treatment as a control. Following this, the life‐history traits of the offspring generation (F 1 ) under the NT, MT and HT scenarios were analysed. 3. The total developmental duration of the aphid F 0 generation was significantly shortened by MT treatments, while it was significantly increased by HT treatments. Adult longevity and the fecundity of F 0 were significantly decreased by HT treatments compared with those in the MT and NT treatments, whereas no significant difference was found between the latter two treatments. The HT treatments applied to the F 0 generation significantly prolonged the total developmental duration of the F 1 generation. The offspring adult longevity was not affected by the mothers' temperature experience. The offspring fecundity was significantly increased when the F 0 generation experienced MT treatments. The intrinsic rate of increase ( r m ) was significantly decreased when the F 0 generation experienced an HT scenario. However, the MT scenario experienced by the F 0 generation did not significantly affect the r m of their offspring. 4. The results will provide new insights into the effects of variable maternal temperature on the individual development and population dynamics of offspring under a global warming scenario.
