Effects of perfluorododecanoic acid on testicular function in mice

Abstract Perfluorodecanoic acid (PFDoA) is a widely distributed environmental pollutant that can affect the functions of many organs. However, systematic evaluations of the effects of PFDoA on testicular functions are lacking. The aim of this study was to investigate the effects of PFDoA on mouse testicular functions, including spermatogenesis, testosterone synthesis, and stem Leydig cells (SLCs) in the interstitial tissue of the testis. PFDoA (0, 2, 5, 10 mg/kg/d) was administered via gavage to 2-month-old mice for 4 weeks. Serum hormone levels and sperm quality were assayed. Furthermore, to investigate the mechanisms by which PFDoA affects testosterone synthesis and spermatogenesis in vivo, the expression of StAR and P450scc in testicular tissue was measured by immunofluorescence staining and quantitative real-time PCR. In addition, the levels of SLC markers, including nestin and CD51, were studied. PFDoA decreased the luteinizing hormone concentration and sperm quality. Although the difference was not statistically significant, mean testosterone levels showed a downward trend. The expression of StAR, P450scc, CD51, and nestin was also suppressed in the PFDoA-treated groups compared with the control group. Our study suggested that PFDoA exposure can decrease testosterone biosynthesis, and even reduce the number of SLCs. These results indicated that PFDoA suppressed the main functions of testis, and further researches are required to identify strategies for preventing or reducing the effect of PFDoA on testicular function.

Mosaic graft with fetal cartilage for the treatment of avascular necrosis of the femoral head

Objective:To explore the simple and effective method for avascular necrosis of the femoral head(ANFH)in order to protect hip joint and improve clinical symptom.Methods:Eighty-five patients(91 hips)of ANFH(62 males and 23 females,ranging in age from 14 to 59 years,with an average of 39.8 years)were treated with mosaic graft with fresh fetal cartilage.The synovium of hip joint were excised,the femoral marrow was decompressed and the lateral circumflex femoral blood vessel were implanted.Results:All the patients were followed up from 1.5 to 10 years.Clinical symptom obtained improvement and hip joint function obtained good recovery.According to hip joint score of Harris,45 cases reached excellent results,32 good,4 fair.The excellent-and -good rate was 90.59%(77/85).Conclusion:Mosaic graft with fresh fetal cartilage for ANFH in the stage Ⅱ,Ⅲ of Ficat is suitable,especially in middle-age and youth.

Assess of the Value of Semen Examination Items in the Diagnosis of Male Infertility

Background: Although semen analysis is a basic method to evaluate male fertility, there was a certain deviation between the semen examination and the actual situation in clinical application. Objective: To evaluate the application value of semen examination items in the diagnosis of male infertility. Method: Males recruited from the Third Affiliated Hospital of Sun Yat-sen University from June 2017 to December 2018 were divided into three groups: normal fertility group, physical examination group (premarital physical examination), and male infertility group. Each group received the following examination such as semen analysis (including semen volume, sperm concentration, progressive motility, total motility, round cell concentration), acrosin activity, and sperm morphology. All the semen analysis items mentioned above were operated according to the WHO laboratory manual for the examination and procession of human semen (Fifth edition) and related documents. Results: The normal fertility group and physical examination group showed significantly higher levels in sperm concentration, progressive motility, total motility, and sperm morphology than those in the male infertility group (P P < 0.001. The percentage of patients having all qualified items in each group was 28.13%, 6.67%, and 6.51% separately. Conclusion: The diagnosis efficiency was higher in sperm concentration, progressive motility, total motility, and sperm morphology than in other semen examination items. A more objective and efficient method of semen examination should be explored to facilitate the diagnoses of male infertility in the future.

Combining Network Pharmacology with Experimental Validation to Elucidate the Mechanism of Salvianolic Acid B in Treating Diabetic Peripheral Neuropathy

Background. Salvianolic acid B (Sal B) is a bioactive component of Radix Salviae, which has antiinflammation and antiapoptotic activity in diabetic complications. However, the molecular mechanism of action of Sal B on diabetic peripheral neuropathy (DPN) is unknown. This study was designed to identify a mechanism for Sal B in the treatment of DPN by using a pharmacology network, molecular docking, and in vitro experiments. Methods. Sal B and DPN-related targets from Gene Cards and OMIM platforms were retrieved and screened. Then, an analysis of possible targets with STRING and Cytoscape software was conducted. KEGG signaling pathways were determined using the R software. Subsequently, the binding capacity of Sal B to target proteins was analyzed by molecular docking and in vitro experiments. Results. A total of 501 targets related to Sal B and 4662 targets related to DPN were identified. Among these targets, 108 intersection targets were shared by Sal B and DPN. After topological and cluster analysis, 11 critical targets were identified, including p38MAPK. KEGG analysis revealed that the AGE-RAGE signaling pathway likely plays an important role in Sal B action on DPN. The p38MAPK protein is a key target in the AGE-RAGE signaling pathway. Molecular docking results suggested that Sal B and p38MAPK have excellent binding affinity (<−5 kcal/mol). The in vitro experiments revealed that Sal B downregulates the expressions of p-P38MAPK, inflammatory cytokines, and apoptosis targets, which are upregulated by hyperglycemia. Conclusion. Sal B may alter DPN by inhibiting inflammation and apoptosis activated by p38MAPK.

Human CD133-Positive Hematopoietic Progenitor Cells Enhance the Malignancy of Breast Cancer Cells

Abstract Background: Human CD133 + hematopoietic progenitor cells (HPCs) are a specific subset of cells and can regulate the malignancy of tumors. However, how CD133 + HPCs affect the malignancy of human breast cancer has not been clarified. Methods: The CD133 + HPCs were isolated and purified from human umbilical cord blood (UCB) .We used in vitro culture of the MCF-7 and MDA-MB-231 cell line, and MCF-7 and MDA-MB-231 cells into nude mice in order to tested whether CD133 + HPCs affected the apoptosis, proliferation, invasion and EMT of breast cancer cells. Results: Co-culture with CD133 + HPCs, but not UCB CD133- cells, promoted the proliferation of human breast cancer MCF-7 or MDA-MB-231 cells, accompanied by reducing their spontaneous apoptosis in vitro and co-administration with CD133 + HPCs significantly enhanced the growth of implanted breast cancer in vivo. Furthermore, co-culture with CD133 + HPCs, enhanced the invasion of breast cancer cells, N-cadherin and Vimentin expression, but reduced E-cadherin expression in Breast cancer cells. Conclusions: The data indicated that CD133 + HPCs enhanced the malignancy of breast cancer cells by inhibiting their spontaneous apoptosis and promoting the process of epithelial mesenchymal transition. These findings may provide new insights in the role of human CD133 + HPCs in the pathogenic process of breast cancer. Therefore, CD133 + HPCs may be a new therapeutic target for inhibiting the progression of breast cancer.

[Expression of 17 beta-hydroxysteroid dehydrogenase type 1 in the kidney of rats: the capacity of the kidney for synthesizing sex hormones].

To investigate the expression of 17 beta-hydroxysteroid dehydrogenase type 1 (17β-HSD1) in the kidney of rats and explore the capacity of the kidney for synthesizing sex hormones.The expressions of 17-HSD1 and sex hormones were detected by Western blotting and radioimmunoassay in rat renal cells in primary cultured for 24 and 48 h in the presence or absence of follicle-stimulating hormone (FSH) and luteinizing hormone (LH).After cell culture for 24 h, the primary rat renal cells expressed a low level of 17β-HSD1 (0.1843±0.076), which increased to 1.6651±0.044 (P<0.01) in response to co-stimulation by FSH and LH. Low levels of estradiol, progesterone and testosterone were also detected in rat renal cells (3.30±3.78, 62.60±12.33, and 22.12±3.36, respectively), and co-stimulation of FSH and LH significantly increased their levels to 8.50±2.64, 117.80±9.79, and 45.04±4.39, respectively (P<0.05). The levels of these hormones showed no significant differences between cells cultured for 24 h and 48 h (P>0.05).The rat renal cells express 17β-HSD1 and are capable of stably secreting sex hormones in response to co-stimulation with FSH and LH, suggesting the capacity of the rat kidneys for synthesizing sex hormones. These findings enrich the understanding of the endocrine function of the kidney.

d-Aspartate in Low-Protein Diets Improves the Pork Quality by Regulating Energy and Lipid Metabolism via the Gut Microbes

d-Aspartate is critical in maintaining hormone secretion and reproductive development in mammals. This study investigated the mechanism of different d-aspartate levels (0, 0.005, 0.05, and 0.5% d-aspartate) in low-protein diets on growth performance and meat quality by mediating the gut microbiota alteration in pigs. We found that adding 0.005% d-aspartate to a low-protein diet could dramatically improve the growth performance during the weaned and growing periods. Dietary d-aspartate with different levels markedly increased the back fat, and 0.5% d-aspartate significantly increased the redness in 24 h and reduced the shear force of the longissimus dorsi (LD) muscle. Moreover, d-aspartate treatments decreased the mRNA expression of MyHC II a and MyHC IIx in the LD muscle. The protein expression of MyH1, MyH7, TFAM, FOXO1, CAR, UCP2, and p-AMPK was upregulated by 0.005% d-aspartate. Additionally, the abundance of Alistipes, Akkermansia, and the [Eubacterium]_coprostanoligenes_group in the intestinal chyme of pigs was significantly decreased by d-aspartate treatments at the genus level, which was also accompanied by a significant decrease in acetate content. These differential microorganisms were significantly correlated with meat quality characteristics. These results indicated that d-aspartate in low-protein diets could improve the growth performance and meat quality in pigs by regulating energy and lipid metabolism via the alteration of gut microbiota.
