Systemic lupus erythematosus (SLE) is a multisystem autoimmune disorder mainly mediated by lymphocytes and autoantibodies, which have a pervasive negative impact on the majority of organs.1 2 It deserves more attention to further explore the pathogenesis because of the unclear complex pathogenesis and the limited clinical efficacy of SLE treatment. Recently, a cross-sectional discovery cohort in the USA conducted by Azzouz et al , published in Annals of Rheumatic Diseases, suggested that specific gut commensal strains, especially Ruminococcus gnavus , may contribute to the disease activity and autoantibody production in SLE patients,3 which put forward a novel concept for the immune pathogenesis of SLE. Their study was well performed and analysed the abundance of intestinal flora and sera profiled for antibacterial and autoantibody responses between SLE patients with the different disease activity index scores and matched healthy controls. Cloning and expression of IL-15 mutants in E.coli and its identification

Objective: To construct two mutants in order to study IL 15 structure function relationship, Methods: The mutant genes were cloned into pBV220 and were expressed in E.coli. The recombinant IL 15 was purified and renaturated and its mutants were tested for bioactivity by CTLL 2 proliferation assay. Result: The results showed that N terminus deleted mutant lost its bioactivity, but C terminus deleted mutant kept this bioactivity. [

Temporal persistence and long-term effect on noninvasive prenatal testing by residual fetal cell-free DNA of demised co-twin after selective fetal reduction in dichorionic diamniotic twin pregnancies

Objectives: To determine the temporal persistence of residual cell-free DNA (cfDNA) of deceased co-twin in maternal circulation after selective fetal reduction and evaluate its long-lasting effect on noninvasive prenatal testing (NIPT) results. Design: Prospective observational study Setting: The Third Affiliated Hospital of Guangzhou Medical University Population: Dichorionic diamniotic twins (n=5) underwent selective fetal reduction of a co-twin with trisomy. Methods: With consent, maternal blood was collected immediately before reduction and periodically after reduction until birth. CfDNA of each maternal blood sample was sequenced for NIPT and analyzed for fetal trisomies and fetal fractions. Main Outcome Measured: Detectable T-scores for trisomy identification and three types of fetal fractions including the total fetal fraction, the fetal fraction of the deceased co-twin, and the fetal fraction of the surviving co-twin.

CT analysis on misdiagnosis of 15 patients with pulmonary periphery inflammatory lesions as cancers

Objective:To analyze the reasons of misdiagnosis in 15 patients with pulmonary periphery spherical lesions and to summarize their CT characteristics of differential diagnosis with pulmonary periphery cancers.Methods:The patients in this study included 9 men and 6 women,with ages ranging from 50 to 78 years old.All patients were scanned by chest plain CT,enhance CT and high resolution CT.The CT films were reviewed retrospectively by two senior radiologists for analyzing the misdiagnosing reasons and were correlated with pathologic findings,Results:All cases were showed solitary pulmonary masses or nodules under the pleura,among them 13 cases were displayed superficial lobulation,8 cases were showed typical pleural retraction,6 cases with thin spiculated contour,4 cases with serrated margin,7 cases with air alveologram,1 cases with decentration cavity,10 cases were intensified distinctly and early,4 cases enhanced uniformly,13 cases with swelling lymph nodules in the hilus of lung or mediastinum.Conclusion:Not identifying accurately the CT characteristics of solitary pulmonary masses or nodules or excessively emphasizing the specificity of some signs instead of anslyzing all around are main cause of the misdiagnosis.For the cases without typical CT characteristics,entirely meticulous analysis and puncture are necessary.

Graphene‐Enhanced Nanomaterials for Wall Painting Protection

Abstract Wall paintings as the highly valuable treasure have been facing severe damaging problems over the years. Ca(OH) 2 as a promising material to protect them has been studied for more than a decade. However, the synthesis methods are still costly, complex, and usually, involve an organic solvent. In addition, problems such as large particle size (>150 nm), slow and weak carbonation, and low consolidation strength for wall paintings remain unsolved. This study demonstrates an unprecedented concept of wall painting protection enabled by graphene‐enhanced nanomaterials through strategically synthesizing Ca(OH) 2 /graphene quantum dot (GQD) nanohybrids using a facile and economic aqueous method. The nanohybrids are uniform and small (center distribution at about 80 nm) and show strong adhesivity to wall painting pigments. In addition, the anti‐UV absorption ability of the nanohybrids is also a significant advantage. More importantly, the Ca(OH) 2 is fully carbonated into a stable CaCO 3 crystal phase “calcite” because of the GQD, which is crucial in wall painting consolidation. As a result, the graphene‐enhanced nanohybrids show superior conservation efficiency compared to as‐synthesized Ca(OH) 2 and commercial Ca(OH) 2 materials. The result opens a new direction for protecting cultural heritage using the newly emerging 2D nanomaterials.

Discovery of Zidovudine as a cardiomyocyte protectant for doxorubicin-induced toxicity through high-throughput phenotypic drug screening

Doxorubicin (DOX) constitutes a cornerstone in cancer chemotherapy, yet its administration is associated with dose-dependent toxicity to the heart, known as doxorubicin-induced cardiotoxicity (DOX-IC). Presently, dexrazoxane stands as the sole approved drug for mitigating DOX-IC; however, its clinical application is restricted due to concerns over severe adversary effects. It is therefore urgent to discover alternative drug candidates to ameliorate DOX-IC. Here, we report the discovery of Zidovudine (ZIDO), a clinically available anti-retroviral medication, as a potent candidate to protect against DOX-IC both in vitro and in vivo by high-throughput phenotypic screening of a library of 1804 Food and Drug Administration (FDA)-approved drugs. Transcriptomic analysis reveals that ZIDO treatment significantly alleviates DOX-induced dysregulation of genes associated with cardiac function and proteotoxic stress. This study sets a paradigm towards discovering novel cardiac protective drugs through repurposing and establishes ZIDO as an agent holding promise for the treatment of DOX-IC patients.

Impact of preoperative Karnofsky Performance Scale (KPS) and American Society of Anesthesiologists (ASA) scores on perioperative complications in patients with recurrent glioma undergoing repeated operation

The objective of this study was to document the impact of the preoperative Karnofsky Performance Scale (KPS) and American Society of Anesthesiologists (ASA) scores on perioperative complications in patients with recurrent glioma who underwent tumor resection via craniotomy. A total of 96 patients were retrospectively reviewed. Based on KPS and ASA scores, patients were categorized into high KPS (> 70) or low KPS (≤ 70) and high ASA (3~4) or low ASA (1~2) groups. Differences in intraoperative risk factors and perioperative complications among the groups were analyzed. Multivariate analysis was performed to identify risk factors for perioperative complications. The most frequent perioperative complications were cerebrospinal fluid leakage (31.8%) and intracranial infection (27.0%); 30-day mortality was 5.2%. The incidence rates of severe complications, central nervous system complications, and total complications were comparable in the low and high KPS groups and in the low and high ASA groups (all p > 0.05). Multivariate analysis showed that low KPS and high ASA scores were not the independent risk factors for perioperative complications. Low KPS and high ASA scores are not associated with increased postoperative complications in patients with recurrent glioma who undergo tumor resection via craniotomy.
