The cover feature picture shows the cellular distribution of chloro-functionalized photo-crosslinking BODIPY. Although nucleophilic substitution of glutathione quenches the fluorescence, the GSH-labeled probe interacted with glutathione S-transferase and recovered fluorescence emission under live-cell conditions. Interestingly, one isomer accumulated in mitochondria without any positively charged mitochondria localizing ligand. More information can be found in the communication by J.-S. Power Spectral Analysis EEG Characteristics of Major Depressive Disorder

본 연구는 주요우울증 환자집단(우울집단)과 정상집단간의 대뇌 활동을 뇌전위분석(electroencephalogram: EEG)을 통해 비교하였고 임상적 진단도구로서 EEG의 활용성을 평가하였다. 이를 위하여 세타, 알파, 베타의 주파수영역 대에서 절대파워, 상대파워를 산출하였으며 우울증에서 나타나는 좌, 우뇌 반구 간 비대칭적인 활동성을 정량화하기 위해 PCT지표와 A지표를 구하여 활동 패턴의 차이를 확인하였다. 그 결과, 우울집단의 상대델타파워는 후두엽을 제외한 모든 영역에서 정상집단보다 높았으며 상대알파파워는 전두엽에서 유의미하게 적었다. 델타주파수는 우울집단이 정상집단에 비해 유의미하게 느렸으며 알파주파수는 전두엽에서 우울집단이 빠른 패턴을 보였다. 한편 우울집단은 우울증에서 특징적인 알파파워 반구 간 비대칭성과 베타파의 반구 간 비대칭성을 나타냈다. 이를 PCT 지표로 산출하였을 때, 알파, 베타 비대칭성 이외에 세타 비대칭성 까지 관찰할 수 있었다. 위와 같은 지표들을 포함한 판별분석 결과 A지표가 포함되었을 때 82.6%, PCT 지표가 포함되었을 때 87%의 비교적 높은 판별율로 각 집단을 구분할 수 있었다. 이런 결과는 EEG 분석이 주요우울증 환자들의 대뇌활동 특징을 확인하고 임상적 진단을 내릴 때 유용한 도구가 될 수 있다는 것을 보여준다.

Diversity-Oriented Approach for Chemical Biology

Synthetic molecules that modulate and probe biological events are critical tools in chemical biology. Utilizing combinatorial and diversity-oriented synthetic strategies, access to large numbers of small molecules is becoming more and more feasible, and research groups in this field can take advantage of the power of chemical diversity. Since the majority of early studies were focused on the discovery of compounds that perturb protein functions, diversity-based approaches are often considered as therapeutic lead discovery tactics. However, the diversity-oriented approach can also be applied to advance distinct aims, such as target protein identification, or the development of imaging probes and sensors. This review provides a personal perspective of the chemical-diversity-based approach and how this principle can be adapted to various chemical biology studies.

Transcriptomic Analysis Provides New Insights into the Tolerance Mechanisms of Green Macroalgae Ulva prolifera to High Temperature and Light Stress

Our research focused on understanding the genetic mechanisms that contribute to the tolerance of Ulva prolifera (Chlorophyta), a marine macroalgae, to the combined stress of high temperature and high light intensity. At the mRNA level, the up-regulated DEGs showed enrichment in pathways related to ribosomes, proteasomes, and peroxisomes. The spliceosome pathway genes were found to be vital for U. prolifera’s ability to adapt to various challenging situations in all the comparison groups. In response to elevated temperature and light intensity stress, there was a significant increase in genes and pathways related to ribosomes, proteasomes, and peroxisomes, whereas autophagy showed an increase in response to stress after 24 h, but not after 48 h. These findings provide novel insights into how U. prolifera adapts to elevated temperature and light stress.

Recent trends in molecular aggregates: An exploration of biomedicine

Abstract Molecular aggregates are receiving tremendous attention, demonstrating immense potential for biomedical applications in vitro and in vivo. For instance, the molecular aggregates of conventional fluorophores influence the electronic excitation states of the aggregates, causing characteristic photophysical property changes. A fundamental understanding of this classical relationship between molecular aggregate structures and photophysics has allowed for innovative biological applications. The chemical characteristics of drug molecules generally trigger the formation of colloidal aggregates, and this is considered detrimental to the drug discovery process. Furthermore, nano‐sized supramolecular aggregates have been used in biomedical imaging and therapy owing to their optimal properties for in vivo utility, including enhanced cell permeability, passive tumor targeting, and convenient surface engineering. Herein, we provide an overview of the recent trends in molecular aggregates for biomedical applications. The changes in photophysical properties of conventional fluorophores and their biological applications are discussed, followed by the effects of conventional drug molecule‐aggregates on drug discovery and therapeutics development. Recent trends in the investigation of biologically important analytes with aggregation‐induced emission are discussed for conventional and unconventional fluorophores. Lastly, we discuss nano‐sized supramolecular aggregates used in imaging and therapeutic purposes, with a focus on in vivo utilization.

Optimal Irradiation Conditions for Radiopacity Tests of Dental Materials Using General-purpose X-ray Imaging Devices

This study verifies the feasibility and optimal conditions for using a general-purpose x-ray imaging device with a relatively large detector and excellent image quality compared to dental x-ray imaging devices according to international standards. The dental material test specimens were produced in the form of disks from four companies with different radiopacity, and aluminum step wedges was manufactured according to ISO 13116. In order to find the optimal irradiation conditions for general X-ray imaging devices, the distance between the target and the tube was fixed at 700 mm, and the exposure time was fixed at 100 ms. And the tube voltage was 50, 60, 70 kV and the tube current was changed to 40, 80, and 160 mA. As a result, it was sufficient to use general-purpose X-ray imaging devices, and the best results were obtained under the optimal conditions of 60 kV and 40 mA.

Focused Fluorescent Probe Library for Metal Cations and Biological Anions

A focused fluorescent probe library for metal cations was developed by combining metal chelators and picolinium/quinolinium moieties as combinatorial blocks connected through a styryl group. Furthermore, metal complexes derived from metal chelators having high binding affinities for metal cations were used to construct a focused probe library for phosphorylated biomolecules. More than 250 fluorescent probes were screened for identifying an ultraselective probe for dTTP.

Global Mapping of Protein–Lipid Interactions by Using Modified Choline‐Containing Phospholipids Metabolically Synthesized in Live Cells

The protein-lipid interaction is an essential metabolic process that mediates cellular signaling and functions. Existing strategies for large-scale mapping studies of the protein-lipid interaction fall short in their incompatibility with metabolic incorporation or inability to remove unwanted interferences from lipidated proteins. By incorporating an alkyne-containing choline head group and a diazirine-modified fatty acid simultaneously into choline-containing phospholipids synthesized from live mammalian cells, protein-phospholipid interactions have been successfully imaged in live cells. Subsequent in situ profiling of the modified Cho phospholipid-crosslinked proteins followed by quantitative proteomics allowed identification of several hundred putative phospholipid-interacting proteins, some of which were further validated.

Discrimination of Avian Influenza Virus Subtypes using Host‐Cell Infection Fingerprinting by a Sulfinate‐based Fluorescence Superoxide Probe

Abstract The current gold‐standard diagnosis method for avian influenza (AI) is an embryonic egg‐based hemagglutination assay followed by immunoblotting or PCR sequencing to confirm subtypes. It requires, however, specialized facilities to handle egg inoculation and incubation, and the subtyping methods relied on costly reagents. Now, the first differential sensing approach to distinguish AI subtypes is demonstrated using series of cell lines and a fluorescent sensor. Susceptibility of AI virus differs depending on genetic backgrounds of host cells. Cells were examined from different organ origins, and the infection patterns against a panel of cells were utilized for AI virus subtyping. To quantify AI infection, a highly cell‐permeable fluorescent superoxide sensor was designed to visualize infection. This differential sensing strategy successfully proved discriminations of AI subtypes and demonstrated as a useful primary screening platform to monitor a large number of samples.
