Sonoporation of Cells by a Parallel Stable Cavitation Microbubble Array

Sonoporation is a targeted drug delivery technique that employs cavitation microbubbles to generate transient pores in the cell membrane, allowing foreign substances to enter cells by passing through the pores. Due to the broad size distribution of microbubbles, cavitation events appear to be a random process, making it difficult to achieve controllable and efficient sonoporation. In this work a technique is reported using a microfluidic device that enables in parallel modulation of membrane permeability by an oscillating microbubble array. Multirectangular channels of uniform size are created at the sidewall to generate an array of monodispersed microbubbles, which oscillate with almost the same amplitude and resonant frequency, ensuring homogeneous sonoporation with high efficacy. Stable harmonic and high harmonic signals emitted by individual oscillating microbubbles are detected by a laser Doppler vibrometer, which indicates stable cavitation occurred. Under the influence of the acoustic radiation forces induced by the oscillating microbubble, single cells can be trapped at an oscillating microbubble surface. The sonoporation of single cells is directly influenced by the individual oscillating microbubble. The parallel sonoporation of multiple cells is achieved with an efficiency of 96.6 ± 1.74% at an acoustic pressure as low as 41.7 kPa.

A microfluidic device based on a pair of slant-finger interdigital transducers (SFITs) is developed to achieve a selective and flexible manipulation of microbubbles (MBs) by surface acoustic waves (SAWs). The resonance frequency of SAWs generated by the SFITs depends on the location of its parallel pathway; the particles at different locations of the SAWs' pathway can be controlled selectively by choosing the frequency of the excitation signal applied on the SFITs. By adjusting the input signal continuously, MBs can be transported along the acoustic aperture precisely. The displacement of MBs has a linear relationship with the frequency shift. The resolution of transportation is 15.19 ± 2.65 μm when the shift of input signal frequency is at a step of 10 kHz. In addition, the MBs can be controlled in a two-dimensional plane by combining variations of the frequency and the relative phase of the excitation signal applied on the SFITs simultaneously. This technology may open up the possibility of selectively and flexibly manipulating MBs using a simple one-dimensional device.

On-chip targeted single cell sonoporation with microbubble destruction excited by surface acoustic waves

We demonstrate that a surface acoustic wave at tens of megahertz frequency is capable of inducing microbubble cluster destruction at a desired location to achieve a single cell's reparable sonoporation. By controlling the position of the microbubble cluster relative to the targeted cell precisely, the effective size of the collapsing microbubbles is measured to be less than 0.68 times the diameter of microbubble cluster. Furthermore, the sonoporation efficiency and the cell viability are 82.4% ± 6.5% and 90% ± 8.7%, respectively, when the targeted cell is within the effective microbubble destruction region.

Aim: Amongst all kinds of cardiovascular diseases (CVDs), atherosclerotic (AS) vulnerable plaque rupture is of the highest morbidity and mortality. By now, improvements on the differentiation of vulnerable atherosclerotic (AS) plaques are still needed for early diagnosis and intervention of AS. As a key step in the advanced stage of AS plaque formation, inflammatory fibrous cap appears to be a valuable target. Evidence showed that circulating neutrophils possess inflammatory endothelial targeting property by the intrinsic cell adhesion molecules on neutrophils. Inspired by this mechanism and in order to realize accurate localization and evaluation of vulnerability of atherosclerotic plaques, we designed a novel bionic microbubble by assembling neutrophils-microbubble complexes, named "BionicBells".

An efficient background timing skew calibration technique for time-interleaving analog-to-digital converters

An efficient background timing skew calibration algorithm is proposed in this article, which detects the sampling time mismatches in time-interleaving analog-to digital converter (TIADC) by estimating the skew-related errors with a reference channel and aligns the sampling edge of each sub-ADC to that of the reference channel by analog variable-delay lines in the negative feedback loop. Compared with conventional background calibration methods based on complex algorithms or serious input restrictions, the proposed technique detects timing skews by only negligible hardware consisting of simple digital blocks and is applicable for a wide range of input including completely random signals. The detailed theoretical analysis and sufficient simulated results revealed that this algorithm is not sensitive to some non-ideal components in actual circuits like mismatches between channels or jitters in clock circuits, which verifies the practicability and robustness of this method.

Tunable Manipulation of Microparticles by CMUT

We achieve trapping and aligning of microparticles by using a miniature capacitive micromachined ultrasonic transducer (CMUT) combined with a phononic crystal plate (PCP). The interaction of an incident wave generated by CMUT with the PCP at resonant frequency can result in periodic and gradient field near the surface of PCP. Then the microparticles on the surface of PCP suffer the acoustic radiation force and can be arranged regularly. Our observations show that the sound field generated by the CMUT device is feasible for particles manipulation.
