This phase 3, randomized, open-label, active-controlled, multicenter study investigated the efficacy of triptorelin pamoate prolonged-release (PR) 3-month in Chinese patients with endometriosis by demonstrating the noninferiority of the 3-month formulation to the standard of care, triptorelin acetate PR 1-month. The trial was conducted in 24 clinical centers in China, and included 300 Chinese women (18–45 years) with endometriosis and regular menstrual cycles who required treatment with a gonadotropin-releasing hormone agonist for 6 months. One group of patients (n = 150) was treated with triptorelin pamoate PR 3-month (15 mg per injection, once every 12 weeks), and the other (n = 150) with triptorelin acetate PR 1-month (3.75 mg per injection, once every 4 weeks). The primary outcome measure was the proportion of patients with estradiol (E2) concentrations suppressed to castration levels (≤ 184 pmol/L, or 50 pg/mL) after 12 weeks of treatment. Triptorelin pamoate PR 3-month was noninferior to triptorelin acetate PR 1-month for the treatment of endometriosis: over 98% of patients in both groups were chemically castrated at week 12. Both formulations were also equally efficacious in reducing endometriosis-associated pelvic pain, and reducing serum concentrations of E2, luteinizing hormone, and follicle-stimulating hormone over time. No new safety concerns were identified. Triptorelin pamoate PR 3-month is a valid alternative to triptorelin acetate PR 1-month for the treatment of Chinese women with endometriosis, with fewer injections and a potentially lower burden of care. Pre-trained language models have achieved promising performance on general benchmarks, but underperform when migrated to a specific domain. Recent works perform pre-training from scratch or continual pre-training on domain corpora. However, in many specific domains, the limited corpus can hardly support obtaining precise representations. To address this issue, we propose a novel Transformer-based language model named VarMAE for domain-adaptive language understanding. Under the masked autoencoding objective, we design a context uncertainty learning module to encode the token's context into a smooth latent distribution. The module can produce diverse and well-formed contextual representations. Experiments on science- and finance-domain NLU tasks demonstrate that VarMAE can be efficiently adapted to new domains with limited resources.
Aiming to investigate the health risk impact of PM2.5 pollution on a heavily populated province of China. The exposure response function was used to assess the health risk of PM2.5 pollution. Results shows that the total number of premature deaths and diseases related to PM2.5 pollution in Shandong might reach 159.8 thousand people based on the new WHO (2021) standards. The health effects of PM2.5 pollution were more severe in men than in women. Five of the 16 cities in Shandong had higher health risks caused by PM2.5 pollution, including LinYi, HeZe, JiNing, JiNan, and WeiFang. PM2.5 pollution resulted in nearly 7.4 billions dollars in healthy economic cost, which accounted for 0.57% of GDP in Shandong in 2021. HeZe, LiaoCheng, ZaoZhuang, and LinYi were the cities where the health economic loss was more than 1% of the local GDP, accounted for 1.30, 1.26, 1.08, and 1.04%. Although the more rigorous assessment criteria, the baseline concentration was lowered by 30 μg/m3 compared to our previous study, there was no significant increase in health risks and economic losses. China's air quality improvement strategy may already be having a positive effect.
Abstract Tea is one of the most popular beverages in the world, especially in Asian societies. Green, oolong and black tea are three main types of tea products. Catechin is the principal polyphenol compound in all tea products including four major subgroup compounds, epigallocatechin gallate (EGCG), epicatehin (EC), epigallocatechin (EGC) and epicatechin gallate (ECG). Green tea contains highest amount of catechin compared to oolong and black tea since fermentation process can significantly reduce the amount of catechin in tea product, which polyphenol oxidase can convert catechins to theaflavins and thearubigins during fermentation process. Therefore, green tea catechin is becoming more and more attractive to nutritionists since it can provide several health benefits to human body. Cholesterol lowering effect is one of the health benefits been studied and proposed over decade. There are well documented evidences that suggested green tea catechin, in particular EGCG has the potential to lower blood cholesterol concentrations. Since the pool bioavailability and absorption ability of catechin, researchers believed that green tea catechin may significantly inhibit lipids absorption in intestine. Mechanisms are including inhibition of pancreatic lipase activity, lipids hydrolysis, and emulsification in intestine and precipitation of micellar cholesterol. In vitro studies, animal studies as well as most of human RCT, consistent results been observed that dietary intake of green tea beverages or extracts could significantly lower circulating cholesterol concentration, in particular lower LDL-C and total cholesterol level. However, in 2010, European Food Safety Authority (EFSA) denied the health claim of cholesterol lowering benefits of dietary intake of green tea or green tea catechins. In this presentation, current scientific evidences and EFSA judgment will be reviewed and discussed.
The treatment of large bone defects remains challenging due to the lack of spatiotemporal management of the immune microenvironment, inflammation response and bone remodeling. To address these issues, we designed and developed a nanoparticle/hydrogel hybrid system that can achieve the combined and sequential delivery of an anti-inflammatory factor (IL-10) and osteogenic drug (icariin, ICA). A photopolymerizable composite hydrogel was prepared by combining gelatin methacryloyl (GelMA) and heparin-based acrylated hyaluronic acid (HA) hydrogels containing IL-10, and poly(dl-lactide-co-glycolide) (PLGA)-HA nanoparticles loaded with ICA were incorporated into the composite hydrogels. The nanoparticle/hydrogel hybrid system demonstrates an array of features including mechanical strength, injectability and photo-crosslinking. The rapid release of IL-10 from the hydrogel effectively exerts immunomodulatory activity, whereas the long-term sustained release of icariin from the PLGA-HA nanoparticles significantly triggers the osteogenic differentiation of bone marrow-derived mesenchymal stem cells (BMSCs). Notably, the combined delivery of IL-10 and ICA from the hybrid system exhibit a synergistic effect for bone remodeling in a critical cranial defect rat model. Our findings indicate the importance of the immunomodulatory microenvironment and osteogenic differentiation for high-quality skull remodeling, and thus the dual-factor releasing nanoparticle/hydrogel hybrid system could be a promising candidate for repairing bone defects.
