The effect of Allium sativum (Garlic) extract on infectious bronchitis virus in specific pathogen free embryonic egg.

Garlic is a plant has been used as a flavor, and anti-microbial and anti-diarrheal agent. Infectious bronchitis virus (IBV) is a coronavirus. The available vaccines against IBV cannot cover new variants. This study evaluated the inhibitory effects of garlic extract on IBV.The constituents of garlic extract were detected by gas chromatography. This study was done in four groups of embryonic SPF eggs; first group was used for virus titration; second group received the mixture of different virus titration and constant amount of garlic extract; third group received 10(-3) titration of virus and after 8 hr received garlic extract and the last group received different dilutions of garlic extract.Based on our results, in the second group, IBV vaccine strain (4/91) at all titration and M41 in 10(-2) and 10(-3) titration and in the third group both variants of virus the embryonic Index (EI) was significantly increased.The garlic extract had inhibitory effects on IBV in the chickens embryo.

Isolation of infectious laryngotracheitis virus in broiler chicken in Iran: First report.

In February 2019, a severe respiratory distress with co-infection of infectious laryngotracheitis (ILT) and Newcastle disease accompanied with Salmonella Enteritidis occurred in a broiler flock in the western region of Iran. Clinical signs included paralysis, torticollis, nasal discharge, conjunctivitis, gasping and respiratory rale with high mortality. At necropsy, caseous diphtheritic membrane adherent to the larynx and trachea was observed. Microscopically, syncytial cells formation with dense eosinophilic intranuclear inclusion bodies were main histopathological findings in tracheal tissues. Conventional polymerase chain reaction (PCR) for ICP4 gene amplification as a definitive diagnosis was utilized for the detection of ILT virus nucleic acid in suspected tracheal samples inoculated on to the chorioallantioc membrane of 11-day-old specific pathogen free (SPF) chicken eggs. Tracheal tissues taken from these SPF birds were positive by nested ILT PCR. In conclusion, because of no vaccination policy against ILT in broilers, the most probable scenario is that virus-laden dust or other fomites can be vectors and virus persistence and disease outbreak can be a sequel of wild virus introduction to the farm.

Isolated Bacteria from the Uteri of Camels with Different Reproductive Backgrounds: A Study on Sampling Methodology, Prevalence, and Clinical Significance

The objectives of this study were to comparatively identify the common bacterial isolates from the uteri of camels coming from different reproductive backgrounds after standardizing the sampling method and to investigate the association of clinically measurable parameters with uterine colonization by these isolates. The uterine samples from 856 dromedary camels yielded a total of 17 different bacterial species with a higher proportion of sub-fertile camel uteri being colonized by bacteria (66.6%) as compared to nulliparous, recently calved, and those with unknown reproductive history combined (44.2%; p < 0.05). Camels with body condition scoring < 3 and those with a consistently echogenic appearance of the uterine lumen by sonography were more likely to be positive on uterine culture, while the presence of pus in uterine discharge was not associated with the odds of bacterial isolation (p > 0.05). While certain strains were more likely to be obtained from the uteri of the sub-fertile group (p < 0.05), embryo transfer to camels with a positive uterine culture in the absence of other gross reproductive pathologies did not necessarily affect the overall pregnancy rate compared to recipients with a negative uterine culture (p > 0.05). In conclusion, a relatively high bacterial load can be identified from the uteri of both sub-fertile and normal dromedary camels, with a higher frequency among the former. The uterine ultrasonography and evaluation of the body condition score can help in identifying the camels in which uterus is contaminated by bacteria.

The Cytoprotective Effects of Morus Alba Leaves in Cultured Fetus Fibroblast Cells against Hyperglycemia

: Background : Diabetes is a chronic disease characterized by high blood glucose levels. Medicinal plants have useful source of hypoglycemic compounds and play important role in preventing the progress of this disease. Morus alba leaves is natural therapeutic agent shown hypoglycemic effects. In this study, the protective effects of alcoholic extract Morus alba leaves on fetus fibroblast cells under hyperglycemic condition were assessed.Materials and Methods : In this experimental study the cells were treated with six different concentrations of extracts. The nontoxic concentration of plant extract was measured with MTT assay in fetus fibroblast cells. Then, the fetus fibroblast cells were incubated with high dose of glucose and plant extract at concentrations of 1000 ppm. DNA synthesis, as marker of cell proliferation was determined.Results : There were significant changes in cell proliferation in treated group with plant extract. Morus alba leaves could cause cell attachment and proliferation. The attachment of cells to culture plates has been shown to be affected by plant extract.Conclusion : The data obtained from this study approved that Morus alba leaves has cytoprotective effects against hyperglycemia.

The Inhibitory Effect of cLF-chimera, a Recombinant Antimicrobial Peptide, on Avian Influenza Virus Subtype H9N2

Abstract Avian influenza subtype H9N2 is the most prevalent influenza virus in poultry worldwide. It imposes economic losses on the poultry industry and has zoonotic potential. Currently, there are two main groups of anti-influenza drugs in use; Adamantanes and Neuraminidase inhibitors. In recent years, there has been increasing resistance to existing anti-influenza drugs. Antimicrobial peptides are a group of potential drug candidates with broad-spectrum activity. cLF-chimera is an antimicrobial peptide synthesized from camel milk lactoferrin. This study's objective is to evaluate the inhibitory effects of cLF-chimera on avian influenza, subtype H9N2. For this purpose, one hundred and seventy 11-day-old embryonated eggs were randomly distributed in 17 groups. Different virus and peptide concentrations were injected into the eggs. The eggs were incubated for four days with daily candling for viability assessment. On the 4th day of incubation, each group's alive or dead embryos were sorted and evaluated for gross anomalies. Next, the chick embryos were fixed with 10% neutral buffered formalin for one week for histopathological studies. The MTT assay was also performed for the peptide and virus concentrations. Overall, the embryo viability results and macroscopic and histopathologic findings showed that the peptide has inhibitory effects against the virus. These findings agree with the MTT assay. Moreover, the peptide has proven effects against pathogenic bacteria that can be advantageous compared to common anti-influenza medications.

Diagnosis of Chlamydia abortus by Isolation in Cell Culture and Real Time PCR in Aborted Sheep and Goats

Background: Ovine enzootic abortion (OEA) caused by Chlamyidia abortus is one of the most important abortive disease in small ruminants. Diagnosis of Ovine enzootic abortion depends on the isolation and detection of the agent or its nucleic acid. The aim of the present study was to detect Chlamydia abortus using both isolation method and real-time PCR in Brucella free flocks in Iran. Methods: Twenty-eight vaginal and conjunctival swab samples which were Chlamydia abortus seropositive, were selected from ewes and does with recent abortion. Then the samples were tested by real-time PCR and positive molecular samples were inoculated into McCoy cells. Results: Using real-time PCR, 18 samples (64.3%) were positive and 7 (25%) of them were isolated in cell culture. Conclusion: The present results indicate that Chlamydia can play a relatively significant role in the abortion in does and ewes in Iran. Although the isolation of Chlamydia abortus have 100% specificity, because of low sensitivity, time consuming, cost and high probability of contamination, it is not suitable for routine laboratory diagnosis. Therefore, applying real-time PCR which have high sensitivity and specificity is recommended.

Paracrine Neuroprotective Effects of Neural Stem Cells on Glutamate-Induced Cortical Neuronal Cell Excitotoxicity

Glutamate is a major excitatory neurotransmitter in mammalian central nervous system. Excessive glutamate releasing overactivates its receptors and changes calcium homeostasis that in turn leads to a cascade of intracellular events causing neuronal degeneration. In current study, we used neural stem cells conditioned medium (NSCs-CM) to investigate its neuroprotective effects on glutamate-treated primary cortical neurons.Embryonic rat primary cortical cultures were exposed to different concentrations of glutamate for 1 hour and then they incubated with NSCs-CM. Subsequently, the amount of cell survival in different glutamate excitotoxic groups were measured after 24 h of incubation by trypan blue exclusion assay and MTT assay. Hoechst and propidium iodide were used for determining apoptotic and necrotic cell death pathways proportion and then the effect of NSCs-CM was investigated on this proportion.NSCs conditioned medium increased viability rate of the primary cortical neurons after glutamate-induced excitotoxicity. Also we found that NSCs-CM provides its neuroprotective effects mainly by decreasing apoptotic cell death rate rather than necrotic cell death rate.The current study shows that adult neural stem cells could exert paracrine neuroprotective effects on cortical neurons following a glutamate neurotoxic insult.

The Survey of Microbiological Contamination of Pitcher Cheese in West Azarbayjan Province, Iran

Milk acts as a suitable peripheral culture for growth and propagation of different kinds of micro organisms. During the process of producing cheese, some micro organisms such as Escherichia coli, Coliform, Staphylococcus, Mold and Yeast may cause its contamination. In respect to the fact that pitcher cheese is produced in traditional way in different regions in West Azarbayjan, the aim of this research is examining the rate of contamination of pitcher cheese in West Azarbayjan. About 42 samples of pitcher cheese were gathered under strill condition from different parts of West Azarbayjan. In order to study microbes contamination, the samples were examined by standard microbiologic ways in laboratory from the 42 samples of pitcher cheese, four samples were contaminated by Staphylococcus aureus coagulase positive, 16 samples were contaminated by E. Coli, 21 samples by Coliform, 17 samples by mold and yeast. The producing and delivering should be controlled because of the rate of contamination in pitcher cheese and this kind of cheese should be produced in half industrial way by controlling and making special facilities for pitcher cheese producers.
