A highly pathogenic bacterium(No.BC-2) was isolated from the infected hemorrhage tilapia.Combination of the colonial morphology observation,the biochemistry test and 16S rDNA sequencing,the highly pathogenic bacterium was determined as Bacillus cereus,which could cause tilapia dead.Then infective test was carried out to normal tilapias with different concentrations of the pathogen,and the qualitative probability data analysis results showed that the logarithmic concentration of LD50 was 7.534 8 with the confidence interval 95% when the infective concentration was 3.4×107 cfu·mL-1.After observing the pathological structure of different tissue and organ sections from infected tilapia,we found gill and intestine were the most evident lesion parts.
To obtain new marine microbe enzyme,marine Penicillium oxalicum was selected from conches and their adhesive matters and cultured by using solid state fermentation.The chitosanase produced with marine Penicillium oxalicum was separated and purified by PEG(relative molecular mass is 1 000,4 000,6 000,10 000,20 000) and AB-8 macroporous resin adsorption,respectively,and the related properties of which were researched.The experimental result shows that this reaction system has better adsorption effect under the optimal adsorption condition of enzyme extracted with the rough macroporous resin,i.e.pH=5,1 g resin and diluted enzyme liquid of 15 mL.Using volume fraction 50% alcohol as desorption agent has better desorption effect.The enzyme activity is increased by 4 times when rude enzyme is purified by both PEG6 000 and macroporous rein adsorption.There are two kinds of chitosanase.The suitable reaction conditions are that the temperature is 30 ℃ or 60 ℃,pH=6 or pH=9 for the reaction system.Adding Fe2+ ion can enhance enzyme activity.Further isolation purification of chitosanase is needed in the future so as to determine the structure and performance of the two components.
The purpose of this study is to analyze the influence of Chinese Shandong opera webcast on audience acceptance behavior in terms of use and artistic characteristics, and to verify the expansion mode of adding the performance attribute and cultural attribute of artistic products to the theory of technological acceptance behavior. this paper analyzes the structural relationship between the variables in the public choice of Shandong opera under the new communication mode of In this study, in order to test the relationship between Shandong opera webcast and audience acceptance behavior, on the basis of combing and summarizing the characteristics of webcast, the personal characteristics of anchor are added as external variables. In this study, two online questionnaires were conducted, the first questionnaire was conducted to test the influence of various variables on receptive behavior, and the second questionnaire was conducted on the basis of revising the results of the first survey. to investigate whether public perception affected by external factors is associated with receptive behavior. The results show that the basic elements of technology acceptance behavior theory, perceived ease of use and perceived usefulness, have a positive (+) influence on the public acceptance behavior of live broadcast of Shandong opera network. Among the external variables, the use characteristics of webcast platform (interaction, convenience, security) and the personal characteristics of Shandong opera anchor (professionalism, attractiveness, popularity) have a positive (+) influence on perceived ease of use and perceived usefulness, which reflects the unique combination of technology and art of Shandong opera webcast. In addition, under the influence of the public's determination of the existing viewing behavior, the possibility of actual viewing and sharing is greatly increased in the future. The scope of this study covers the practical application fields such as webcast operation, Shandong opera performance, public acceptance of art products, etc., enriches the relevant research, and enhances the public's understanding and attention to the traditional art webcast. An effective plan is put forward to expand the audience.
The symbiotic microorganisms fungus S9 was isolated using different cultural medium from marine organisms in Chongwu Sea.It′s physiological characters and relation between fermentation conditions and growth period were stuied.The experimental results show the optimum condition for the fermentation of the obtained marine fungal: concentration of sucrose is 5 g·L-1,concentration of peptone is 1.5 g·L-1,pH=6.5,fermentation time is 10 d.Fungal secondary metabolites have antagonistic activity on fungal growth.
