Structure-Function Studies of Two Yeast Homing Endonucleases that Evolved to Cleave Identical Targets with Dissimilar Rates and Specificities

The LAGLIDADG family of homing endonucleases (LHEs) bind to and cleave their DNA recognition sequences with high specificity. Much of our understanding for how these proteins evolve their specificities has come from studying LHE homologues. To gain insight into the molecular basis of LHE specificity, we characterized I-WcaI, the homologue of the Saccharomyces cerevisiae I-SceI LHE found in Wickerhamomyces canadensis. Although I-WcaI and I-SceI cleave the same recognition sequence, expression of I-WcaI, but not I-SceI, is toxic in bacteria. Toxicity suppressing mutations frequently occur at I-WcaI residues critical for activity and I-WcaI cleaves many more non-cognate sequences in the Escherichia coli genome than I-SceI, suggesting I-WcaI endonuclease activity is the basis of toxicity. In vitro, I-WcaI is a more active and a less specific endonuclease than I-SceI, again accounting for the observed toxicity in vivo. We determined the X-ray crystal structure of I-WcaI bound to its cognate target site and found that I-WcaI and I-SceI use residues at different positions to make similar base-specific contacts. Furthermore, in some regions of the DNA interface where I-WcaI specificity is lower, the protein makes fewer DNA contacts than I-SceI. Taken together, these findings demonstrate the plastic nature of LHE site recognition and suggest that I-WcaI and I-SceI are situated at different points in their evolutionary pathways towards acquiring target site specificity.

Auditory Attention Detection Dataset The experiment is conducted by Yuanming, Zhang and Ziyan, Yuan, supervised by Jing, Lu. Two zip files are included in this dataset. The script.zip file contains several scripts to divide each trial into several parts. These scripts are useful if you want to split the dataset into training, validation and test set. The other zip file contains the preprocessed EEG and audio signals. (Not the raw EEG!) Each mat file contains EEG and audio signals from one subject. Their are two variables named expinfo and data. The expinfo contains the attended direction information and audio stimulus playback information of each trial of the experiments. The azimuth column in expinfo indicates the directions of the two competing speakers. The negative directions indicated the speaker coming from the left, and positive coming from the right. The data contains several essential fields relative to the EEG and audio signals. The data.eeg field is a cell array with 32 cell elements. Each cell contains the EEG signals in a single trial. And the index of the cell is the number of trial. The data.leftWav and data.rightWav field are original audio stimuli, and the data.leftEnv and data.rightEnv are the envelopes of the data.leftWav and data.rightWav respectively. The data.dim field tells you the physical meaning of each dimension of each signal, and the data.dim.chan.eeg contains the name of each channel of the EEG signal.

Influence of Environmental Knowledge and Green Trust on Green Purchase Behaviour

This study investigates the influence of environmental knowledge and green trust on green purchase intentions and behavior, and the mediating role of purchase intentions. Through questionnaire surveys and statistical analysis, the research findings reveal that environmental knowledge and green trust significantly influence green purchase intentions and actual green purchasing behaviour. The results of this study hold significant meaning for the formulation of environmental education and environmental advocacy strategies, contributing to enhancing consumers' environmental awareness and promoting green purchasing behaviour.

Peptide Truncation Leads to a Twist and an Unusual Increase in Affinity for Casitas B-Lineage Lymphoma Tyrosine Kinase Binding Domain

We describe truncation and SAR studies to identify a pentapeptide that binds Cbl tyrosine kinase binding domain with a higher affinity than the parental peptide. The pentapeptide has an alternative binding mode that allows occupancy of a previously uncharacterized groove. A peptide library was used to map the binding site and define the interface landscape. Our results suggest that the pentapeptide is an ideal starting point for the development of inhibitors against Cbl driven diseases.

Exploiting the P-1 Pocket of BRCT Domains Toward a Structure Guided Inhibitor Design

Breast cancer gene 1 carboxy terminus (BRCT) domains are found in a number of proteins that are important for DNA damage response (DDR). The BRCT domains bind phosphorylated proteins and these protein-protein interactions are essential for DDR and DNA repair. High affinity domain specific inhibitors are needed to facilitate the dissection of the protein-protein interactions in the DDR signaling. The BRCT domains of BRCA1 bind phosphorylated protein through a pSXXF consensus recognition motif. We identified a hydrophobic pocket at the P-1 position of the pSXXF binding site. Here we conducted a structure-guided synthesis of peptide analogs with hydrophobic functional groups at the P-1 position. Evaluation of these led to the identification of a peptide mimic 15 with a inhibitory constant (K(i)) of 40 nM for BRCT(BRCA1). Analysis of the TopBP1 and MDC1 BRCT domains suggests a similar approach is viable to design high affinity inhibitors.

Antecedents of Green Purchasing Intentions among Higher Education Population in Malaysia

This empirical study aims to operationalize the relationship between the direct and indirect influence of factors on green purchase behavior through the mediation of green purchase intention in the context of the Malaysian higher education population. It employed Structural Equation Modeling to assess the strength of relationships between the constructs. The research findings indicate that green purchase intention mediates the relationship between its antecedents and green purchase behavior. This indirect effect is more prominent in the factors of environmental knowledge and green trust. The study helps scholars further understand the facilitating mechanisms of green purchase behavior.

Analysis of Trade Symbiosis between China and Countries along the "Belt and Road"

Under the prevailing trade protectionism and the continuing downturn in the world economy, China has proposed the “Belt and Road” Initiative, so as to build a community of interests, responsibilities and destiny, leading countries to achieve diversified, balanced and sustainable development. The concept of symbiosis coincides with the Silk Road spirit of peace and cooperation, openness and inclusiveness, and mutual benefit and win-win situation, as advocated by the “Belt and Road” Initiative. In this context, this study selects trade data from 2011 to 2020 and measures the symbiosis trade relations between China and different regional countries along the Belt and Road in four dimensions: Complementarity, Competitiveness, Dependence and Integration. The results show that the trade competitiveness between China and the "Belt and Road" countries is weak, the trade complementarity is at a medium level, the trade dependence is generally high, and they have relatively close trade contacts. In brief, trade cooperation needs to be further strengthened. Finally, this study provides some policy recommendations to improve the trade symbiosis relations and perfect trade structure.
