Poultry waste is an abundant renewable source for the recovery of several value-added metabolites with potential industrial applications. This study describes the production of protease on poultry waste, with the subsequent use of the same poultry waste for the extraction of antioxidants. An extracellular protease-producing strain was isolated from Cuddalore coast, India, and identified as Streptomyces sp. MAB18. Its protease was purified 17.13-fold with 21.62% yield with a specific activity of 2398.36 U/mg and the molecular weight was estimated as 43 kDa. The enzyme was optimally active at pH 8–10 and temperature 50–60°C and it was most stable up to pH 12 and 6–12% of NaCl concentration. The enzyme activity was reduced when treated with Hg 2+ , Pb 2+ , and SDS and stimulated by Fe 2+ , Mg 2+ , Triton X-100, DMSO (dimethyl sulfoxide), sodium sulphite, and β -mercaptoethanol. Furthermore, the antioxidant activities of protease were evaluated using in vitro antioxidant assays, such as DPPH radical-scavenging activity, O 2 scavenging activity, NO scavenging activity, Fe 2+ chelating activity, and reducing power. The enzyme showed important antioxidant potential with an IC 50 value of78±0.28 mg/mL. Results of the present study indicate that the poultry waste-derived protease may be useful as supplementary protein and antioxidant in the animal feed formulations.

Ecological Factors Shaping Marine Actinobacterial Niches in Diverse Coastal Habitats of the Andamans Sea, India

Marine microbial communities are structured by the spatial temporal, variability of physico-chemical, and biotic parameters. These parameters form a useful tool for the ecological assessment and monitoring of coastal ecosystems. The present investigation aims to study the influence of the physico-chemical parameters on the distributional variations of culturable actinobacterial population density, at six different locations on the Havelock Island coast of the Andamans, India. The study covered the important parameters viz. temperature in air (29–23°C), surface water (26–24°C), sediments (27–23°C), salinity in water (18–35 psu), sediments (17–34 psu), pH in water (6–8) and sediments (7.6–8.7), dissolved oxygen (DO) (4.60–2.10 ml l−1), electrical conductivity (EC) (2–5.5 dS m−1), nitrogen (N) (0.55–8 mg g−1), potassium (K) (2.63–10.88 mg g−1), phosphorus (P) (4.50–3.13 mg g−1), total organic carbon (TOC) (60.88–11.88 mg g−1), sand (65–95%), silt (3.5–9.4%), and clay (1.2–30%). Culturable actinobacterial population density in the sediments ranged from 8 × 103 to 21 × 103 CFU g−1, registering the higher value at station 4 (mangrove) and the lower value at station 3 (beach). Cluster and principal component analysis (PCA) results have revealed that the mangrove environment favors a higher density of actinobacteria than the coral, seagrass, and beach habitats of the Island.

Production, Biochemical Characterization and Detergents Application of Keratinase from the Marine Actinobacterium Actinoalloteichus sp. MA‐32

Abstract The present study is the first report on poultry feathers as a novel, inexpensive substrate for the production of a thermo‐ and detergent stable keratinase from a marine actinobacterium belonging to the genus Actinoalloteichus . Medium composition and culture conditions for the keratinase production by Actinoalloteichus sp. MA‐32 were optimized using two statistical methods: Plackett–Burman design was applied to find the key ingredients and conditions for the best yield of enzyme production and central composite design used to optimize the concentration of the five significant variables: whole chicken feather, soy flour, MgSO 4 ·7H 2 O, KH 2 PO 4 and NaCl. The medium optimization resulted in a 19.30‐fold increase with a 31.99 % yield with a specific activity of 3842.57 U mg −1 and the molecular weight was estimated as 66 kDa. The enzyme was optimally active at pH 8–10 and temperature 50–60 °C and it was most stable up to pH 12 and 10–14 % of NaCl concentration. The enzyme activity was reduced when treated with Hg 2+ , Pb 2+ , Tween‐80, 1,10‐phenanthroline and EDTA and stimulated by Fe 2+ , Mg 2+ , Cu 2+ , Ca 2+ , Ni 2+ , Mn 2+ , SDS, ethoxylated (9.5EO) octylphenol, DMSO, sodium sulfite and β ‐mercaptoethanol. The keratinase exhibited a significant stability and compatibility with most of the tested commercial laundry detergents, demonstrating its feasibility for inclusion in laundry detergent formulation. These results suggest that this extracellular keratinase may be a useful alternative and eco‐friendly route for handling the abundant amount of waste feathers or for applications in detergent formulation and other industrial processes.

Prospects and challenges of environmental DNA (eDNA) metabarcoding in mangrove restoration in Southeast Asia

Species detection using environmental DNA (eDNA) is a biomonitoring tool that can be widely applied to mangrove restoration and management. Compared to traditional surveys that are taxa-specific and time-consuming, eDNA metabarcoding offers a rapid, non-invasive and cost-efficient method for monitoring mangrove biodiversity and characterising the spatio-temporal distribution of multiple taxa simultaneously. General guidelines for eDNA metabarcoding are well-established for aquatic systems, but habitat-specific guidelines are still lacking. Mangrove habitats, as priority ecosystems for restoration in Southeast Asia, present unique prospects and challenges in these regards. Environmental DNA metabarcoding can be used to (1) track functional recovery in ecological restoration, (2) prioritise conservation areas, (3) provide early warning for threats, (4) monitor threatened taxa, (5) monitor response to climate change, and (6) support community-based restoration. However, these potential applications have yet been realized in Southeast Asia due to (1) technical challenges, (2) lack of standardised methods, (3) spatio-temporal difficulties in defining community, (4) data limitations, and (5) lack of funding, infrastructure and technical capacity. Successful implementation of eDNA metabarcoding in mangrove restoration activities would encourage the development of data-driven coastal management and equitable conservation programs. Eventually, this would promote Southeast Asia’s shared regional interests in food security, coastal defence and biodiversity conservation.
