Lymphocytes obtained from CLL patients exhibited a delayed and reduced response to PHA when cultured in diffusion chambers. DNA synthesis (8–10 hr) and general time (15–19 hr) of the late‐developing CLL blasts were consistent with normal values ( T s : 8–10 hr; T c : 14–17 hr). However, the G 2 period of CLL blasts seemed more variable, and their mitotic index during the response at 5–6 days was 30–50% of the values determined for normal blasts during their peak response at 2–3 days.

Bone Marrow Lymphocytes: Their Isolation, Colony-Forming Capacity and Graft-Versus-Host Potential I. The Isolation and Characterization of Subpopulations of Bone Marrow Lymphocytes

Bone marrow cells sedimented through a linear sucrose gradient containing fetal calf serum, separated into four major fractions. Three of these were lymphocyte-rich fractions that may be as high as 99 percent pure. About 40 percent of total marrow lymphocytes (originally 25-28 percent) were removed in these three fractions. The technique offers the advantage of short sedimentation intervals (5 minutes) and high reproducibility in the purity and yield of lymphocytes. The technique isolates lymphocytes in a one-step procedure and further subdivides the lymphocytes on the basis of differences in sedimentation and proliferative characteristics. The availability of a technique that may differentiate subpopulations of bone marrow lymphocytes should stimulate future investigations aimed at clarifying the differentiative and functional capacities of these cells.

Tritiated-Thymidine Incorporating Cells in the Peripheral Blood of Normal and Splenectomized Rats.

Aortic blood from normal, sham-splenectomized, and splenectomized rats was incubated with tritiated thymidine (2 μc/ml) for 1 hour at 37°C and the labeling incidences for the buffy coat leukocytes determined autoradiographically. It was found that a small percentage of mononuclear cells in the peripheral blood of normal rats were capable of incorporating tritiated-thymidine. Following splenectomy the numbers of these cells increased more than 4-fold for 23 days post-operation. The labeled cell types are described and possible causes for their increase in numbers after splenectomy are discussed.

Autoradiographic Studies of Human Lymphocytes Cultured in Vivo

Human lymphocytes were cultured in diffusion chambers implanted into the peritoneal cavities of rats. In cultures treated with PHA, the pattern of morphologic transformation and initiation of DNA synthesis paralleled closely similar studies made in vitro. Immunologic stimulation of the human cells by the heterologous host was presumably minimal or absent since control cultures (without PHA) showed no significant degree of blastogenesis when compared with PHA-treated cultures. Minimum DNA synthesis time determined for the PHA-stimulated lymphocytes was 9–10 hr. Values found for the duration of Tc (16–18 hr) and minimum TG2 (2 hr) were shorter than those reported for similar studies in vitro. The in vivo culture method using Millipore chambers appears to offer a more physiologic method for studying lymphocytes.
