The transient depression of hippocampal CA1 LTP induced by chronic intermittent ethanol exposure is associated with an inhibition of the MAP kinase pathway

Abstract Using electrophysiological and biochemical approaches, we investigated the effects of chronic, intermittent ethanol (CIE) treatment on activation of the mitogen activated protein kinase (MAPK), also known as extracellular signal regulated protein kinase 1 and 2. In hippocampal slices taken from control rats, brief high‐frequency stimulation to Schaffer collateral fibers induced a large post‐tetanic potentiation (PTP) in the CA1 region that decayed to stable long‐term potentiation (LTP) of field extracellular postsynaptic potentials. Western blot analyses showed that phosphorylation of MAPK was increased during PTP and returned to baseline levels during LTP. In slices from the rats removed immediately from CIE treatment, PTP and MAPK activation during the PTP was significantly less than that observed in control slices and LTP was absent. In slices from rats subjected to 1 day withdrawal from CIE treatment, both the reduction in MAPK phosphorylation during PTP and the impairment of PTP and LTP were still evident. Recovery of PTP and partial recovery of LTP was observed in slices obtained from 5‐day withdrawn rats. However, MAPK activation during PTP was still attenuated significantly. Interestingly, MAPK activation was enhanced significantly during LTP in 5‐day withdrawn rats as well as the sensitivity to MAPK inhibitor PD 098059. In addition to these changes in HFS‐induced MAPK activation, we also observed a significant reduction in the basal phosphorylation of MAPK in slices removed from rats immediately after CIE treatment. These results implicate the MAPK signal transduction pathway as a potential cellular target of ethanol. Alterations in MAPKs could play an important role in the alcohol‐induced changes in synaptic plasticity associated with the effects of alcohol abuse on learning and memory processes.

Dantrolene does not block calcium pulse‐induced calcium release from a putative calcium channel in sarcoplasmic reticulum from malignant hyperthermia and normal pig muscle

Calcium pulse additions to isolated SR membranes can cause a reversible efflux of calcium. The threshold level of calcium loading at which calcium efflux occurs is lower for SR membranes isolated from malignant hyperthermia susceptible (MHS) swine. Dantrolene, a unique muscle relaxant, had no effect on threshold calcium load, amounts and rates of calcium release from SR isolated from control and MHS skeletal muscle. It is concluded that the putative calcium channel through which this calcium pulse‐induced calcium release mechanism occurs is not affected by dantrolene under these experimental conditions.

Response of Rabbit Aortic Strips Under Pulsed-Tension to Catecholamines.

A technique for studying the pharmacology of vascular smooth muscle has been described. This technique utilizes pulsed tension rather than the static tension customarily employed and has permitted the following observations: Contraction responses to the catecholamines have shown accelerated onset and recovery with increased uniformity of response. A differential sensitivity to norepinephrine has been shown to exist in segments taken from different parts of the same aorta. A progressive reduction in sensitivity to norepinephrine was observed from the arch to the origin of the superior mesenteric artery.

Patterns of Spontaneous Hemostasis in the Blood Vessels of the Rabbit's Ear

Abstract1. A technique has been developed for the continuous recording of the amount of blood flowing from single vessels following their transection.2. the blood is washed away by a stream of saline flowing at constant rate. the mixture of blood and saline crosses a beam of blue light directed to a phototube. the output of the associated circuit is fed into an ink writer which records the changes in the concentration of erythrocytes suspended in the saline.3. Experiments performed with this technique on the small vessels of the rabbit's ear have revealed two basic patterns of hemostasis.4. the “arterial pattern” is characterized by a sudden outburst of blood when the vessel is cut, followed within a few seconds by a marked decrease in the rate of blood outflow. Therefore, the record begins with a sharp initial peak. Rhythmic fluctuations in the rate of blood outflow which might be due to vasomotion, are often seen in this type of patterns.5. the “venous pattern” lacks the initial peak, looking roughly trapezoidal with a hump or plateau.6. A mixed “peak and hump” pattern is also often found; its significance is discussed.7. in a small group of experiments, the influence of mechanical pressure, infiltration of the perivascular tissue with local anesthetics, and the injection of heparine and antiplatelet serum on the patterns of hemostasis were studied.

Dantrolene Dose Response in Malignant Hyperthermia-susceptible (MHS) Swine

The authors examined the thesis that a dose of dantrolene producing 95 per cent of maximal muscle relaxation (ED95) would effectively prevent or treat malignant hyperthermia (MH). In one group of four pigs sensitive to malignant hyperthermia (MHS) a dose response to intravenous dantrolene was determined by quantitation of toe twitch tension. From these data, the ED95 relaxant dose (3.5 mg/kg) was derived. In a second group of four MHS pigs, the ED95 dantrolene dose was administered prior to MH challenge with succinylcholine, 2 mg/kg, and halothane, 1.5 per cent. MH was prevented in each animal, while measurements of arterial pressure, arterial blood-gas, pH and lactate values, rectal temperature, and heart rate were done. Later, MH rapidly developed in the same four animals when they were again challenged. When treated with the ED95 dose, each animal survived. Finally, each pig was challenged without dantrolene prophylaxis or therapeusis, and all succumbed from MH. Previous studies have shown the efficacious use of dantrolene in prevention or treatment of porcine MH, but doses used have varied, without rationale. The present study shows that in MHS pigs the ED95 muscle relaxant dose of dantrolene (3.5 mg/kg) successfully prevents and treats MH.
