Abstract Epigenetic alterations of gene function play a central role in the pathogenesis of many tumors and in the process of aging. Abnormal methylation at transcriptional sites of genes results in epigenetic silencing of the genes that protect against tumor formation or that repair DNA. To date, several studies have analyzed methylation status by oligonucleotide arrays, restriction analysis (COBRA), methylation‐specific amplification, and sequence analysis. Thisrequires high concentration of bisulfite‐treated DNA, which mandates use of commercially available expensive kits, and is an often laborious and time‐consuming task. In this article, we report a simplified high‐throughput method, which can serve as a surrogate for screening methylation profiles of various genes and has high sensitivity compared with the other methods described previously. Coordination of Tourism Supply Chain Management

Supply chain coordination in manufacturing sector has been the key focus of many research scholars since last four decades; however, research of tourism supply chain (TSC) management (SCM) has gained little attention. The unique characteristics of tourism sector call for special consideration to satisfy various stakeholders having divergent business objectives across different operating systems. This calls for a holistic view of the entire supply chain and not just playing in silos. The present chapter studies the important features of tourism products/services and identifies issues that are core to managing the supply chain in the tourism sector with the ultimate objective to achieve coordination of the TSC. The chapter gives an overview of current studies in tourism from the perspective of supply chain coordination and provides a coordination framework and stresses the role of information sharing for successful implementation of the coordination mechanism.

Current concept in neural regeneration research: NSCs isolation, characterization and transplantation in various neurodegenerative diseases and stroke: A review

Since last few years, an impressive amount of data has been generated regarding the basic in vitro and in vivo biology of neural stem cells (NSCs) and there is much far hope for the success in cell replacement therapies for several human neurodegenerative diseases and stroke. The discovery of adult neurogenesis (the endogenous production of new neurons) in the mammalian brain more than 40 years ago has resulted in a wealth of knowledge about stem cells biology in neuroscience research. Various studies have done in search of a suitable source for NSCs which could be used in animal models to understand the basic and transplantation biology before treating to human. The difficulties in isolating pure population of NSCs limit the study of neural stem behavior and factors that regulate them. Several studies on human fetal brain and spinal cord derived NSCs in animal models have shown some interesting results for cell replacement therapies in many neurodegenerative diseases and stroke models. Also the methods and conditions used for in vitro culture of these cells provide an important base for their applicability and specificity in a definite target of the disease. Various important developments and modifications have been made in stem cells research which is needed to be more specified and enrolment in clinical studies using advanced approaches. This review explains about the current perspectives and suitable sources for NSCs isolation, characterization, in vitro proliferation and their use in cell replacement therapies for the treatment of various neurodegenerative diseases and strokes.

Exploring Alternative Medicine Options for the Prevention or Treatment of Coronavirus Disease 2019 (Covid-19): A Systematic Scoping Review

Background: Coronavirus disease 2019 (COVID-19) is caused by coronavirus 2 (SARS-CoV-2). Symptoms include fever, cough, shortness of breath, muscle pain, pneumonia, and multi-organ failure. The infection spreads from one person to another via respiratory droplets. Alternative Medicine (AMs) viz., Ayurveda, Homeopathy, Unani, and Traditional Chinese Medicine (TCM), are being promoted for the prevention of COVID-19. The aim of this systematic scoping review was to identify and summarize the scientific evidences promoting the use of AMs for the prevention of COVID- 19. Methods: A comprehensive search of electronic search engines (PubMed and Web of Science) was performed. In addition, freewheeling searches of the government health ministries and government websites was done to retrieve the available information. Records available until 12th March 2020 were considered. Reports proposing the use of AMs for prevention or treatment of COVID-19 across all countries were included. Screening (primary and secondary) of the records and data extraction from the eligible studies were done by a single reviewer followed by a random quality check (10%) by the second reviewer. Results: Overall, 8 records (7 from China and 1 from India) exploring the use of AMs for the prevention or treatment of COVID-19 were identified. Different medicines were explored by different AM systems. Conclusion: Several AMs options are proposed for the prevention or treatment of COVID-19. However, their efficacy and safety still needs scientific validation through rigorous randomized controlled trials. Isolation and Characterization of Stem Cells Sub Population within the Human Fetal Liver

Human fetal liver is the potential source of both hematopoietic and non-hematopoietic stem cells which can be identified using phenotypic markers. Isolation of homogenous populations of hepatic progenitor cells and their sub-populations is an essential prerequisite for investigating specific markers and appropriate cell types for their possible clinical applications. Several studies have demonstrated the presence of a variety of stem cell populations within the fetal liver. The present study was undertaken to identify specialized cell populations, their valuable growth potential and bi-potential differentiation capability derived from human fetal liver using CD133. Liver cirrhosis is characterized by distortion of liver architecture, necrosis of hepatocytes and regenerative nodules formation leading to cirrhosis. Various types of cell sources have been used for the management and treatment of decompensated liver cirrhosis. Knowledge of stem cells has offered a new dimension for regenerative therapy and has been considered as one of the potential adjuvant treatment modality in patients with end stage liver diseases (ESLD). Human fetal hepatic progenitor cells are less immunogenic than adult ones. They are highly propagative and challenging to cryopreservation. In our earlier studies we have demonstrated that fetuses at 10-18 wk of gestation age contain a large number of actively dividing hepatic stem and progenitor cells which possess bi-potent nature having potential to differentiate into bile duct cells and mature hepatocytes. Hepatic stem cell therapy for the treatment of ESLD is in their early stage of the translation. The emerging technology of decellularization and recellularization might offer a significant platform for developing bioengineered personalized livers to come over the scarcity of desired number of donor organs for the treatment of ESLD. Despite these significant advancements long-term tracking of stem cells in human is the most important subject nowadays in order to answer several unsettles issues regarding the route of delivery, the choice of stem cell type(s), the cell number and the time-point of cell delivery for the treatment in a chronic setting. Answering to these questions will further contribute to the development of safer, noninvasive, and repeatable imaging modalities that could discover better cell therapeutic approaches from bench to bed-side. Combinatorial approach of decellularization and nanotechnology could pave a way towards the better understanding in determination of cell fate post-transplantation. The liver is a central metabolic and highly specialized detoxifying organ. Approximate 70%-75% of liver functions are being carried out by the hepatocytes which together with cholangiocytes (5%-10% of hepatic cells) constitute the liver parenchyma. Liver regeneration is a very rapid and well synchronized phenomenon. Liver responds to initial injury compensating the loss of parenchymal mass. If damage perseveres commencement of petite terminal peri-portal oval cells activates which engross mobilization of several vital factors. Liver cirrhosis is characterized by distortion of liver architecture, necrosis of hepatocytes and regenerative nodules formation leading to cirrhosis. The available treatment modalities are not very effective against liver cirrhosis. Stem cells are considered one of the potential adjuvant treatment modalities in liver cirrhosis patients. Fetal hepatic stem cells transplantation in liver cirrhosis has emerged as an alternative to organ transplantation. However long-term stem cells labeling and tracking is needed for cell fate determination after transplantation. Decellularization technology provides a novel tool to develop bioengineered personalized livers to accomplish the shortage of donor livers. Stem cells labeling and tracking in human tissues/organs is the most important subject nowadays in order to answer several unsettled issues regarding the cell delivery routes, stem cell choice, number of cells to be infused and the time-point of cell delivery for the treatment in a chronic setting. Answering to these questions will further contribute to the development of safer, non-invasive, and repeatable imaging modalities that could discover better cell therapeutic approaches from bench to bed-side. Long-term monitoring of transplanted stem cells continuously with high temporal resolution and good biocompatibility will allow us to better understand the precise regeneration mechanisms in different organs. Studying the Importance of <i>VacA</i> Gene of <i>Helicobacter pylori</i> in Identifying the Pathogenicity of Strains by Comparing It with the Disease Status of the Subjects

Introduction: H. pylori is a primary pathogen isolated by Warren and Marshall in 1983. They called it as Campylobacter Pylori and in 1989 Goodwin et al. renamed it as Helicobacter pylori. Helicobacter pylorus is one of the responsible factors for causing gastritis, Peptic ulcer disease and is strongly associated with gastric carcinoma and gastric Malt lymphoma. Materials and Methods: In this study 300 biopsies were collected at Deccan College of Medical Sciences and Allied Hospitals, Hyderabad, India. Of these, 101 patients had peptic ulcer, 95 patients had gastritis and 4 had gastric carcinoma. A total of four gastric biopsy specimens were collected. One was used for culturing H. pylori, one for histological lesions and the remaining two one each from the antrum & corpus was collected in phosphate buffered saline for the DNA analysis. Results: Helicobacter pylori were isolated from many of the biopsies and the identification of Helicobacter pylori was confirmed in 200 biopsies with colony characteristics, Biochemical tests, and 16S rRNA amplification. The presence of VacA marker was detected by using appropriate primers. From the data obtained in our study, in a total of 169/200 isolates s1 region was found in 84.1% and s2 was found in 36/200 i.e. 18% of the isolates. In the middle region, m1awas found in 60/200 which was 30%, m1b was found in 60/200 i.e. 60% and m2 was found in 114/200 i.e. 57.1% of the isolates. Conclusion: Thus, VacA detection might be helpful for determination of which patients are at highest risk for severe clinical outcomes such as duodenal ulcer, gastric ulcer or gastric carcinoma and eventually, to define strategies for the treatment or prevention of H. pylori infection.

Helicobacter pylori and other Helicobacter species DNA in human bile samples from patients with various hepato-biliary diseases

AIM:To investigate the presence of Helicobacter species by nested PCR of 16S rRNA genes followed by the presence of Helicobacter pylori (H pylori )16S rRNA, ureA , cagA genes in bile obtained at endoscopic retrograde cholangio-pancreatography (ERCP) from 60 Indian subjects. METHODS:Sixty bile samples were obtained from patients diagnosed with various hepato-biliary diseases and control subjects at ERCP.PCR analysis was carried out using primers for Helicobacter genus 16S rRNA gene and H pylori (16S rRNA, ureA and cagA ) genes.Gastric H pylori status was also assessed from biopsies obtained at endoscopy from patients with various hepatobiliary diseases and controls.The control group mainly consisted of subjects with gastric disorders.Sequencing analysis was performed to confirm that PCR products with 16S rRNA and cagA primers were derived from H pylori . RESULTSNo Helicobacters were grown in culture from the bile samples.Helicobacter DNA was detected in bile of 96.7% and 6.6% of groups I and II respectively.Ten from group I were positive for 16S rRNA and ureA and 9 were positive for cagA gene.In contrast of the 2 from the control, 1 amplified with 16S rRNA, ureA and cagA primers used.The sequences of the 16S rRNA genes and cagA were 99% similar to Helicobacter pylori .CONCLUSION: Helicobacters are associated with the pathogenesis of various hepato-biliary disorders.

Recent Approaches and Novel Therapeutic Targets in Human Glioma

Malignant gliomas are highly invasive primary brain tumors that are not known to metastasize outside the central nervous system (CNS).The median survival time of patients with glioma is only 6 months to 2 years depending on the variability in patient's condition, type of tumor and variable treatment parameters.In recent times, Gamma knife (GK) and temozolomide (TMZ) have showed a new dimension for the treatment of gliomas, even these modalities have not able to bring a paradigm shift in overall survival and morbidity.Despite the aggressive current therapeutic interventions such as surgery, radiotherapy and chemotherapy, improved therapeutic strategies/targets are greatly needed.Interactions between the tumor and its microenvironment are known to regulate malignancies and there is need to focus more research on these pathways in order to develop more reliable therapeutic strategies for the treatment of gliomas.It has also been shown that drug transporters are highly expressed by small population of most type of tumors, providing for a level of resistance which are relatively quiescent and show higher level of DNA repair, and a lowered ability to enter the apoptosis; can provide another therapeutic targets in most of the cancer types including gliomas.The alteration of miRNA expression profile in glioma has also been found to be associated with neoplastic agents, hence open a new direction for the treatment.Therefore, the present review has been focused on some of these new potential targets for therapeutic interventions in the prognosis and treatment of human glioma.

The epidemiology and prevalence of Ulcerative colitis in the South of India

Ulcerative colitis is considered frequent in majority of European and North American population and exceptional in most of the developing Asian countries. There is a dearth of reported data from South India on the incidence of the disease and its prevalence. Hence the present study was designed to estimate the prevalence of ulcerative colitis in a tertiary care hospital of Hyderabad, South India. The study population consisted of 157 Ulcerative colitis and 204 healthy subjects. All subjects were interviewed by means of a questionnaire for general demographical details and socioeconomic conditions, health related quality of life and history of UC. Patients were categorized based on disease severity; moderate: 95, and severe: 62 and disease manifestation: 73 (46.5%) pancolitis, 60 (38.2%) left-sided colitis and 24 (15.3%) had proctosigmoiditits.
