Mitochondrial genomes and phylogenomic analysis of Ulva lactuca Linnaeus (Ulvophyceae, Chlorophyta)

Two mitogenomes of Ulva lactuca Linnaeus, one from Florida, USA and another nearly complete mtDNA from Chile, had previously been sequenced. Here, the complete mitogenome of U. lactuca from Shantou, China was sequenced and compared with them. The circular-mapping mitogenome of U. lactuca was 62,021 bp in size and 407 bases longer than that from the USA. The average sequence identity in gene sequence regions was 99.8% among these three U. lactuca mtDNAs. Differences in genome size were mainly caused by duplication mutations and insertion/deletion mutations of short DNA sequences. Two intergenic regions (nad6-trnS and trnF-trnG) contained long tandem repeats which displayed a high level of variation in repeat sequences and copy numbers, indicating fast mutation rates in these regions. The phylogenomic analysis revealed that these three samples of U. lactuca formed a strongly supported clade. The mitogenome can be used to understand the phylogeography and population genetics in Ulva on a global scale.

Toxicological effect of microcystin on mice liver

Lake Dianchi is a typical eutrophication lake and we make a toxic experiment by the MC test liquid from the heavy algae bloom. The toxicological experiment on ICR-mice shows that the MC test liquid has extraordinary harm to the mice liver, which become hyperemia and swelling. Through the electron microscope observation, we can see the rough endoplasmic reticulum of the liver cell is broken, and the mitochondria cristae is partly dissolved and the chromatin in cell nucleolus is reduced.The experiment shows that the 0.06mg/kg MC-LR injected into the abdominal cavity of ICR-mice causes the submicrostruture changes in the liver cell and the ICR-mice to death.

Preliminary study on macrobenthos in waters around Yangshan Deepwater Port

The benthic survey was carried out in waters around the area of the First Phase Project of Yangshan Deepwater Port(30°30′～30°54′N、121°45′～122°30′E)during the months of February,May and August in winter,spring and summer respectively from 2003 to 2005.According to the macrobenthos survey method setted in the Specification for Marine Monitoring,the macrobenthic species composition,biomass,inhabit density,community diversity and structure changes were discussed based on the results of investigation.The results showed that the benthic species numbers in 2003,2004 and 2005 were 21,17 and 16,the biomass were 4.26 g/m2,0.62 g/m2 and 2.18 g/m2,the average inhabit densities were 31.10 ind/m2,12.50 ind/m2 and 11.00 ind/m2,the community diversity indices were 0.81,0.36 and 0.16 respectively.The results also indicated that benthic species numbers,biomass,inhabit densities and diversity indexes have all been reducing year by year and it was affected by the construction of Yangshan Deepwater Port.

Preliminary study on inter-annual variations of macrobenthos in Yangshan sea area

According to the results of macrobenthos survey,the inter-annual variation of macrobenthic species composition,biomass,density,community diversity and structure are discussed with parameters including Y,C,d,H′,J,by using the method of cluster analysis.The result showed that,in 2003,2004 and 2005,the numbers of benthic species were 21,17 and 16,the biomass were 4.26g/m2,0.62g/m2 and 2.18 g/m2,and the average inhabit densities were 31.10 ind/m2,12.50 ind/m2 and 11.00 ind/m2,respectively.The result indicated that benthic species numbers,biomass,inhabit densities,and diversity indexes were all reducing year by year,while the purity index increased accordingly.Cluster analysis revealed that the results of three surveys in February,May and August,2003 were in one cluster,while the results of 6 surveys in 2004~2005 were in one cluster.This may indicate that the ecological changes between 2003 and 2004~2005 were obvious,and the ecological changes of macrobenthos were affected by the construction of Yangshan deepwater port.

Environmental degradation and loss of species diversity in Dianchi Lake

Dianchi Lake,a large shallow lake in Yunnan-Guizhou Plateau,southwest of China,lies in(102°42′E) and 25°04′N.It consists of Grass Lake and Outer Lake,which is under the jurisdiction of Kunming City.It is 1885 m above sea level,with a length of 40 km and an average width of 10.5 km.Its surface area is 298.4 km~2,with an average depth of 4.1 m and water capacity of 1.29 billion m~3(1886.5 m above sea level).This article bears on the study of environmental degradation and loss of species diversity in Dianchi Lake.Dianchi Lake once enjoyed the prestige of Flower Sea.However,having been heavily polluted by industrial and agricultural waste water in the recent three decades,the water environment in Dianchi Lake was degenerated,and the hydrophyte was sharply destroyed and furthermore the loss of species diversity was typical.The dominant blue-green algae have increased whereas green algae have decreased rapidly.Zooplankton species diversity has enhanced and the species structure of large benthos has become simple.Species diversity of fish decreased from 1.81 in 1957 to 0.11 in 1997.From the 1960s to the 2000s,the species diversity index in Grass Lake declined from 2.70(Shannon-Weiner Index) to 0.29 and that in Dianchi Lake declined from 1.08 to 0.67 during the1970s-the 2000s.This article expounds the dynamic mechanism between the environmental degeneration and the loss of species diversity from the 1960s to the 2000s in Dianchi Lake.The dynamics shows that environmental change in Dianchi Lake can provide some theoretical bases for ecological prevention and warning,environmental protection and restoration in lakes.

Ecological characters of Ctenophora in East China Sea

Based on the investigations in four seasons from 1997 to 2000,this paper studied the seasonal quantitative variation and geographical distribution characters of Ctenophora in East China Sea(23°30′～33°00′N、118°30′～128°00′E).The results showed that the abundance of Ctenophora peaked in autumn(6.3 ind·(100 m~3)~(-1)),followed by in spring(1.6 ind·(100 m~3)~(-1)),winter(1.4 ind·(100 m~3)~(-1)),and summer(0.6 ind·(100 m~3)~(-1)).A total of six species were observed,among which,Pleurobrachia globosa was the most important species,Hormiphora palnata and Beroe cucumis were the common species,Cestum sp.was the subordinate species,while Ocyropsis crystalline and B.avata were the rare species.The Ctenophora in East China Sea was a kind of warm water species,because its quantitative variation had a close relation with the motion of warm water current,and the key factor affecting Ctenophora abundance was warm water current but not the seasonal change of temperature.In all species of Ctenophora,P.globosa had stronger adaptability to its environment,followed by Cestum sp.and B.cucumis,and H.palnata,Ocyropsis crystalline and B.avata.

Sequence analysis of ITS regions of Ulva prolifera in green tides in Yellow Sea of China in 2009 and 2010

Thirteen drifting samples were collected at 5 sites in Jiangsu and Shandong in 2009~2010.The internal transcribed spacers(ITS region) including 5.8S rDNA were sequenced.Based on ITS sequences with the neighbor joining(NJ) and maximum parsimony(MP) method,the phylogenetic trees were constructed respectively.The morphological comparison showed that the bloom-forming alga in 2009 was similar to that in 2010,The similar morphological features included the obvious main-branch,which was full of air,algae somatic cell in the longitudinal rows,quadrilateral or polygonal in shape,(13~21) μm ×(15~28)μm in size,and chloroplast with one pyrenoids.Based on the comparative analysis of the full length of ITS sequences from the 13 samples and data from GenBank,it showed that the ITS including 5.8S rDNA sequences of the 13 samples were identical and also the same as the data about Ulva prolifera from the Yellow Sea in the GenBank,which fell into the same clade including the information about U.linza,U.prolifera and U.procera.The molecular and morphological results consistently showed that the bloom-forming alga in the Yellow Sea in 2009 and 2010 was the same species.Combined the results of previous studies,it showd that U.prolifera was the bloom-forming alga in the Yellow Sea for 4 years(2007~2010).

Study on intertidal macrobenthos in North Branch of Changjiang Estuary

In July of 2003(high water period) and Jan.of 2004(low water period),three intertidal transets,which are Yonglongsha(A) section,Fuminlongchang(B) section and Dongwangsha(C) section,were set up in the north edge of Island Chongming.In high water period,there are 17 species and the species of crustacea are the most.In low water period,the species of polychaetous annelids are the most in seven species.In high water period,Helice japonica is the dominant species,on the other hand,in low water period,Xenophthalmus pinnotheroides is the main.As the biomass reduction of Sesarma haematocheir、Helice japonica and Periophthalmus cantonensis in low water period,the biomass in high water period is 20～65 times to the biomass in low.Biomass,species number and species diversity are increased as the reduction of the distance to Changjiang Estuary.The community succession between the high and low water period was distinct.

Complete mitochondrial genome of the brown algaSargassum fusiforme(Sargassaceae, Phaeophyceae): genome architecture and taxonomic consideration

Sargassum fusiforme (Harvey) Setchell (=Hizikia fusiformis (Harvey) Okamura) is one of the most important economic seaweeds for mariculture in China. In this study, we present the complete mitochondrial genome of S. fusiforme. The genome is 34,696 bp in length with circular organization, encoding the standard set of three ribosomal RNA genes (rRNA), 25 transfer RNA genes (tRNA), 35 protein-coding genes, and two conserved open reading frames (ORFs). Its total AT content is 62.47%, lower than other brown algae except Pylaiella littoralis. The mitogenome carries 1571 bp of intergenic region constituting 4.53% of the genome, and 13 pairs of overlapping genes with the overlap size from 1 to 90 bp. The phylogenetic analyses based on 35 protein-coding genes reveal that S. fusiforme has a closer evolutionary relationship with Sargassum muticum than Sargassum horneri, indicating Hizikia are not distinct evolutionary entity and should be reduced to synonymy with Sargassum.
