Morphology and phylogeny of Thecadinium ovatum (Dinophyceae) from the Pacific coast of Russia

Thecadinium ovatum is one of the four species of Thecadinium sensu lato which has no apical pore, but has a ventral pore on the first apical plate, an unusual 'multi-pimple-plate' in the precingular series, and an antapical plate with several spines. Its original thecal plate formula was 3ʹ 6ʹʹ 6c 5s? 5ʹʹʹ 1ʹʹʹʹ. Morphological examination of cells from the Sea of Japan showed that T. ovatum has four apical plates and that the multi-pimple-plate is an anterior intercalary plate. The revised thecal plate formula of the species is 4ʹ1a 5ʹʹ6c 6s 5ʹʹʹ1ʹʹʹʹ. Molecular genetic data of T. ovatum were obtained for the first time. LSU rRNA gene sequences obtained by a single-cell PCR method from natural samples were used for taxonomic and phylogenetic studies. The sequence for T. ovatum did not show a close phylogenetic relationship to sequences of T. yashimaense, T. kofoidii and T. pseudokofoidii. The phylogenetic analysis of the D1/D2 and D8/D10 regions of the large subunit (LSU) rRNA from single cells indicated that Thecadinium ovatum formed the nearest sister branch to the Protoceratiaceae, including Pentaplacodinium usupianum, Pentaplacodinium saltonense, Protoceratium reticulatum, and Ceratocorys species. This clade was placed within the order Gonyaulacales. Consequently, T. ovatum does not belong to Thecadinium sensu stricto, nor does it show a phylogenetic relationship with other Thecadinium sensu lato species (T. yashimaense and Psammodinium inclinatum). Thus, T. ovatum should be transferred to an independent new genus that may be created after genetic information is available for the other morphologically close species.

Intraspecific heterogeneity, multiple allelism and morphological divergence between morphotypes of Thalassiosira allenii (Bacillariophyta) from the Sea of Japan

Thalassiosira allenii is reported for the first time from marine phytoplankton of the Russian far East. Five strains representing three morphological variants of T. allenii, from three locations in the Sea of Japan, were investigated. The D8–D10 domain of LSU rDNA and rbcL of T. allenii were sequenced for the first time. Three morphological forms of T. allenii showed completely identical rbcL sequences and distinctly different nuclear rDNA sequences. The genetic differences between morphotypes were determined by the number of allelic variants of LSU rDNA, one of which was common to all studied strains. Two different types of the D1–D3 domain and three different types of the D8–D10 divergent domain were detected in the cultured strains of T. allenii. The three morphotypes are described in detail and compared. One allele of D1–D3 and one allele of D8–D10 were identified in the strains of T. allenii from two stations in Ussuri Bay. Two and three alleles of the D8–D10 domain were identified in the strains with an ambiguous morphotype and in T. allenii var. striata, respectively, from one station in Amur Bay. Subdivision of T. allenii into morphotypes based on morphology matched that based on molecular-genetic analyses using D8–D10 LSU rDNA. Caution is recommended in using LSU rDNA sequences and rbcL divergence as the principal criterion for delimiting and metabarcoding T. allenii varieties/morphotypes. The lack of genetic differentiation in rbcL, and the presence of clear intra-specific genetic differentiation among morphotypes in two regions of LSU rDNA suggest a link between morphological differences and the nuclear genome. The D8–D10 domain of LSU rDNA appears more suitable for discriminating morphotypes within T. allenii. The rbcL is an effective marker for recognizing T. allenii at species level, but not at infraspecific level.

Coral Holobionts Possess Distinct Lipid Profiles That May Be Shaped by Symbiodiniaceae Taxonomy

Symbiotic relationships are very important for corals. Abiotic stressors cause the acclimatization of cell membranes in symbionts, which possess different membrane acclimatization strategies. Membrane stability is determined by a unique lipid composition and, thus, the profile of thylakoid lipids can depend on coral symbiont species. We have analyzed and compared thylakoid lipidomes (mono- and digalactosyldiacylglycerols (MGDG and DGDG), sulfoquinovosyldiacylglycerols (SQDG), and phosphatidylglycerols (PG)) of crude extracts from symbiotic reef-building coral Acropora sp., the hydrocoral Millepora platyphylla, and the octocoral Sinularia flexibilis. S. flexibilis crude extracts were characterized by a very high SQDG/PG ratio, a DGDG/MGDG ratio < 1, a lower degree of galactolipid unsaturation, a higher content of SQDG with polyunsaturated fatty acids, and a thinner thylakoid membrane which may be explained by the presence of thermosensitive dinoflagellates Cladocopium C3. In contrast, crude extracts of M. platyphylla and Acropora sp. exhibited the lipidome features of thermotolerant Symbiodiniaceae. M. platyphylla and Acropora sp. colonies contained Cladocopium C3u and Cladocopium C71/C71a symbionts, respectively, and their lipidome profiles showed features that indicate thermotolerance. We suggest that an association with symbionts that exhibit the thermotolerant thylakoid lipidome features, combined with a high Symbiodiniaceae diversity, may facilitate further acclimatization/adaptation of M. platyphylla and Acropora sp. holobionts in the South China Sea.

Scrippsiella trochoidea cysts in recent sediments from Amur Bay, Sea of Japan: distribution and phylogeny

Abstract A study of vertical distribution of dinoflagellate cysts in recent sediments was conducted for the first time in the Far Eastern seas of Russia. Dinoflagellate cysts were studied in sediment cores retrieved from five stations in Amur Bay (Peter the Great Bay, Sea of Japan) in 2010 and 2013. The Scrippsiella trochoidea complex was the dominant type of cyst, the average proportion of which (in the entire sediment cores) reached 39% of the total cyst abundance. The maximum abundance of S. trochoidea complex reached 5513 cysts g -1 dry sediment. Cysts of the S. trochoidea complex were found to remain viable in the sediments after exposure to sub-zero temperatures. The identification of S. trochoidea in the Far Eastern seas of Russia was first confirmed by using biomolecular genetic analyses. The study of vertical distribution showed that cysts of this species dominate the upper sediment layers (0–10 cm). A tendency for abundance to increase from the shore toward the middle part of the bay was observed in the spatial distribution of cysts.

Attheya(Bacillariophyta) from the northwestern Sea of Japan: a description of two subgenera based on molecular and morphological data

Strains of the diatoms Attheya ussurensis, A. decora and A. longicornis, isolated from the northwestern Sea of Japan, were studied to infer their phylogeny and determine their systematic position. Morphological studies were conducted using light and transmission electron microscopy, and molecular analyses were based on nuclear-encoded small-subunit (18S) ribosomal DNA (rDNA) and ribulose-1,5-bisphosphate carboxylase/oxygenase large subunit (rbcL) of the plastid gene. The strains studied, as well as sequences of A. longicornis and A. septentrionalis accessed from GenBank, belong to the class Mediophyceae. Molecular data confirmed that Attheya holds a basal position relative to the genera Biddulphia and Chaetoceros (Chaetocerotales). Thus, Attheya is a member of this lineage and not sister to Bacillariophyceae. Strains of Attheya belong to two clearly distinct lineages, differing in morphology and ecology. One lineage included strains of the type species of the genus, A. decora and the species A. ussurensis (psammophytes); the second lineage included the strains A. longicornis and A. septentrionalis (epiphytes or sea-ice diatoms). Morphologically, the two lineages differed in ratio of horn length to valve diameter, shape and number of plastids, presence/absence of girdle band perforation and presence/absence of rimoportula on the valve. Based on morphological and molecular data, we propose Attheya subgenus Attheya, to include A. decora, A. ussurensis, A.arenicola; and Attheya subgenus Dolichoceratia, to include A. longicornis, A. septentrionalis, A. flexuosa, A. gaussii and A. armata.

Phylogenetic characterization of cryptic species of the marine dinoflagellate, Ostreopsis sp. Shmidt, 1902, from Russian coastal waters, the Sea of Japan

Ostreopsis Schmidt, 1902 is a genus of benthic and epiphytic dinoflagellates known to produce palytoxin (PTX) and its analogues. In 2006, high concentrations (10,970 cells g -1 dry weight) of Ostreopsis spp. were found in waters of Peter the Great Bay, Sea of Japan, near Vladivostok (43˚04ʹ N, 131˚57ʹ E). Since then, high numbers of Ostreopsis spp. have been observed each year associated with the epiphytic assemblages of the bay. The current investigation focuses on molecular-genetic analysis and phylogenetic reconstruction of Ostreopsis sp. from Russian waters based on ribosomal DNA genes (LSU rDNA (D1/D2) and ITS1-5.8S-ITS2 region, SSU rDNA). Single cells from environmental samples and monoclonal cultures were used to isolate DNA, which was compared to sequences of Ostreopsis species from the northwestern Pacific, Indo-Pacific, Malaysia, and Atlantic/Mediterranean regions. Phylogenetic analysis, based on rDNA sequence, revealed three distinctly different Ostreopsis ribotypes: one similar to a toxic Korean strain, the second analogous to a Japanese strain from Okinawa (Ostreopsis sp.2), and the third similar to an extremely toxic Japanese strain (Ostreopsis sp.1). The present study has revealed high genetic homogeneity of SSU rDNA within the Russian strain that allowed us to reliably estimate distinctions among populations from different regions and homogeneity within population.

Morphology and molecular phylogeny of Pseudohaptolina sorokinii sp. nov. (Prymnesiales, Haptophyta) from the Sea of Japan, Russia

:An under-ice brown water discolouration in coastal waters of Vladivostok, the northwestern Sea of Japan, during March 2013 was caused by a bloom of haptophyte algae initially identified as Chrysochromulina sp. Seawater samples collected near the edge of the ice hole contained aggregations of haptophytes with concentrations up to 7.4 × 107 cells·l−1 (99.8% of the total density of phytoplankton). Detailed morphological studies conducted by light microscopy and transmission electron microscopy methods and molecular analyses based on nuclear-encoded small-subunit [18S ribosomal (r)DNA] gene sequences were performed on an environmental isolate (PB2013) to determine its systematic position. The characteristic structure of the covering body scales of our specimens was similar to Chrysochromulina birgeri found earlier from Finland and Japan. However, the scale morphology of our specimens and C. birgeri was not identical, and the sequence of C. birgeri was not known. The molecular signature indicated the systematic position of our specimens as belonging to the genus Pseudohaptolina. The value of rDNA divergence (p) between PB2013 and P. arctica, a single known species of Pseudohaptolina, was 0.011 (1.1%); this value falls within the same order of magnitude used to distinguish species-level differences in other Prymnesiales. On this basis, we proposed the new species Pseudohaptolina sorokinii sp. nov.

Membrane vectorial lipidomic features of coral host cells’ plasma membrane and lipid profiles of their endosymbionts Cladocopium

Abstract The symbiotic relationships between coral animal host and autotrophic dinoflagellates are based on the mutual exchange and tight control of nutritional inputs supporting successful growth. The corals Sinularia heterospiculata and Acropora aspera were cultivated using a flow-through circulation system supplying seawater during cold and warm seasons of the year, then sorted into host cells and symbionts and subjected to phylogenetic, morphological, and advanced lipid analyses. Here we show, that the lipidomes of the dinoflagellates Cladocopium C1/C3 and acroporide-specific Cladocopium hosted by the corals, are determined by lipidomic features of different thermosensitivity and unique betaine- and phospholipid molecular species. Phosphatidylserines and ceramiaminoethylphosphonates are not detected in the symbionts and predominantly localized on the inner leaflet of the S. heterospiculata host plasma membrane. The transmembrane distribution of phosphatidylethanolamines of S. heterospiculata host changes during different seasons of the year, possibly contributing to mutualistic nutritional exchange across this membrane complex to provide the host with a secure adaptive mechanism and ecological benefits.
