Three new sulphur glycosides from the seeds of Descurainia sophia

Three new sulphur glycosides, raphanuside B–D (1–3), together with a known sulphur glycoside, raphanuside (4) were isolated from the decoction of the seeds of Descurainia sophia (L.) Webb ex Prantl, and the compound 4 was reported for the first time from this plant. Their structures were identified by means of UV, IR, 1D, 2D NMR (HSQC, HMBC and NOESY) and HR–ESI–MS spectroscopic data.

Overexpression of bacterial γ‐glutamylcysteine synthetase mediates changes in cadmium influx, allocation and detoxification in poplar

Summary Overexpression of bacterial γ‐glutamylcysteine synthetase in the cytosol of P opulus tremula × P . alba produces higher glutathione ( GSH ) concentrations in leaves, thereby indicating the potential for cadmium ( C d) phytoremediation. However, the net C d 2+ influx in association with H + / C a 2+ , C d tolerance, and the underlying molecular and physiological mechanisms are uncharacterized in these poplars. We assessed net C d 2+ influx, C d tolerance and the transcriptional regulation of several genes involved in C d 2+ transport and detoxification in wild‐type and transgenic poplars. Poplars exhibited highest net C d 2+ influxes into roots at p H 5.5 and 0.1 mM C a 2+ . Transgenics had higher C d 2+ uptake rates and elevated transcript levels of several genes involved in C d 2+ transport and detoxification compared with wild‐type poplars. Transgenics exhibited greater C d accumulation in the aerial parts than wild‐type plants in response to C d 2+ exposure. Moreover, transgenic poplars had lower concentrations of O 2 ˙ − and H 2 O 2 ; higher concentrations of total thiols, GSH and oxidized GSH in roots and/or leaves; and stimulated foliar GSH reductase activity compared with wild‐type plants. These results indicate that transgenics are more tolerant of 100 μM C d 2+ than wild‐type plants, probably due to the GSH ‐mediated induction of the transcription of genes involved in C d 2+ transport and detoxification.

Metabolomics reveals the response of hydroprimed maize to mitigate the impact of soil salinization

Soil salinization is a major environmental stressor hindering global crop production. Hydropriming has emerged as a promising approach to reduce salt stress and enhance crop yields on salinized land. However, a better mechanisitic understanding is required to improve salt stress tolerance. We used a biochemical and metabolomics approach to study the effect of salt stress of hydroprimed maize to identify the types and variation of differentially accumulated metabolites. Here we show that hydropriming significantly increased catalase (CAT) activity, soluble sugar and proline content, decreased superoxide dismutase (SOD) activity and peroxide (H2O2) content. Conversely, hydropriming had no significant effect on POD activity, soluble protein and MDA content under salt stress. The Metabolite analysis indicated that salt stress significantly increased the content of 1278 metabolites and decreased the content of 1044 metabolites. Ethisterone (progesterone) was the most important metabolite produced in the roots of unprimed samples in response to salt s tress. Pathway enrichment analysis indicated that flavone and flavonol biosynthesis, which relate to scavenging reactive oxygen species (ROS), was the most significant metabolic pathway related to salt stress. Hydropriming significantly increased the content of 873 metabolites and significantly decreased the content of 1313 metabolites. 5-Methyltetrahydrofolate, a methyl donor for methionine, was the most important metabolite produced in the roots of hydroprimed samples in response to salt stress. Plant growth regulator, such as melatonin, gibberellin A8, estrone, abscisic acid and brassinolide involved in both treatment. Our results not only verify the roles of key metabolites in resisting salt stress, but also further evidence that flavone and flavonol biosynthesis and plant growth regulator relate to salt tolerance.

Kandelia candel Thioredoxin f Confers Osmotic Stress Tolerance in Transgenic Tobacco

Water deficit caused by osmotic stress and drought limits crop yield and tree growth worldwide. Screening and identifying candidate genes from stress-resistant species are a genetic engineering strategy to increase drought resistance. In this study, an increased concentration of mannitol resulted in elevated expression of thioredoxin f (KcTrxf) in the nonsecretor mangrove species Kandelia candel. By means of amino acid sequence and phylogenetic analysis, the mangrove Trx was classified as an f-type thioredoxin. Subcellular localization showed that KcTrxf localizes to chloroplasts. Enzymatic activity characterization revealed that KcTrxf recombinant protein possesses the disulfide reductase function. KcTrxf overexpression contributes to osmotic and drought tolerance in tobacco in terms of fresh weight, root length, malondialdehyde (MDA) content, and hydrogen peroxide (H2O2) production. KcTrxf was shown to reduce the stomatal aperture by enhancing K+ efflux in guard cells, which increased the water-retaining capacity in leaves under drought conditions. Notably, the abscisic acid (ABA) sensitivity was increased in KcTrxf-transgenic tobacco, which benefits plants exposed to drought by reducing water loss by promoting stomatal closure. KcTrxf-transgenic plants limited drought-induced H2O2 in leaves, which could reduce lipid peroxidation and retain the membrane integrity. Additionally, glutathione (GSH) contributing to reactive oxygen species (ROS) scavenging and transgenic plants are more efficient at regenerating GSH from oxidized glutathione (GSSG) under conditions of drought stress. Notably, KcTrxf-transgenic plants had increased glucose and fructose contents under drought stress conditions, presumably resulting from KcTrxf-promoted starch degradation under water stress. We conclude that KcTrxf contributes to drought tolerance by increasing the water status, by enhancing osmotic adjustment, and by maintaining ROS homeostasis in transgene plants.

Contrasting Leaf Trait Responses of Conifer and Broadleaved Seedlings to Altered Resource Availability Are Linked to Resource Strategies

(1) Understanding tree seedling responses to water, nutrient, and light availability is crucial to precisely predict potential shifts in composition and structure of forest communities under future climatic conditions. (2) We exposed seedlings of widespread Central European tree species with contrasting leaf habit, deciduous broadleaves (DB) and evergreen conifers (EC), to factorial combinations of manipulated precipitation (100% and 50% of ambient), shade (40% and 60% of full sunlight), and nutrient availability (low and high NPK), and measured specific leaf area, C/N ratio, soluble sugars, starch and non-structural carbohydrate concentration, and δ13C of the leaves. (3) We found contrasting effects of water and nutrient availability on foliar traits of the two species groups: EC exhibited higher tolerance to low resource availability but also less plasticity in foliar traits, which is congruent with a “slow” resource strategy. In contrast, foliage of DB reacted particularly to altered nutrient availability, corresponding to a “fast” resource strategy with high foliar plasticity and rapid adjustments to resource fluctuations, commonly adopted by species with high growth rates. (4) We conclude that DB will respond to environmental change with foliar acclimation, while EC will either tolerate, to some extent, or shift their distribution range in response to environmental change.

Two new flavonoid glucosides from the fruits of Sinopodophyllum hexandrum

Two new flavonoid glucosides, sinoflavonoidgs A (1) and B (2), along with three known analogues 3–5, were isolated from the fruits of Sinopodophyllum hexandrum. Their structures were established on the basis of extensive spectroscopic (UV, IR, HR-ESI-MS, 1H-NMR, 13C-NMR, HSQC, HMBC) and chromatographic (HPLC) analysis. The isolation of compounds 1–2 represents the first report of ring B-glucosided flavonoids from the genus Sinopodophyllum. The cytotoxic activities of all isolated compounds were evaluated in comparison with etoposide against four cell lines (MCF-7, HepG2, HeLa, KB). The antioxidant activities of all isolated compounds were examined by DPPH free radical-scavenging assay. The preliminary structure-activity relationships showed that the glycosilation of 3-methoxyquercetin at C-3' resulted in a greater decrease of cytotoxic and antioxidant activity.

Evaluating forest harvesting to reduce its hydrologic impact with a spatial decision support system

Timber harvesting changes the condition of forest ecosystems, which are a major influence on the characteristics of headwater streams. Such characteristics include the quantity and timing of base flow and storm flow, concentrations of sediment and dissolved nutrients, water temperature, and the stability of the stream channels. This paper explores previous studies dealing with the relationship between timber harvesting and its hydrologic effects, especially long term water yield increase. The watershed disturbance threshold theory is raised and investigated in detail. The development and evaluation of a spatial decision support system, the Harvest Schedule Review System (HSRS), is then described. The HSRS will aid in the minimization of hydrological impacts of forest harvesting, along with its related, negative environmental influences. It provides a spatially and temporally explicit tool for users to analyze the hydrologic impact of forest harvest schedules.
