Cloning and sequencing of ammonia-oxidizing bacteria amoA gene from environmental samples

To provide basis of establishing gene-engineering bacteria,the ammonia-oxidizing bacteria amoA gene was cloned and sequenced.Samples(marked as 1,2,3,4) from four different environments were cultured in selective enrichment culture for ammonia-oxidizing bacteria.To amplify the whole long gene amoA,PCR primers were designed as AMOB/AMOE,and BamHⅠ and HindⅢ sites were added to the 5′-ends of each primer respectively to make the following steps easily.After amplification of the four samples,agarose gel electrophoresis of amplification product showed that samples 1 and 3 produced the expected product,but samples 2 and 4 did not.Retracted PCR products were ligated into pGEM-T vector to constitute sequencing vector.After being sequenced the sequences were blasted in GenBank through Internet.The results showed that amplified DNA fragments of sample 1 and 3 both have 99.7% homology with the amoA gene of Nitrosomonas sp.GH22.It could be concluded that the amoA gene was obtained from the exacted bacteria.

Assessement of left ventricular diastolic function in patients with early type 2 diabetes mellitus by wave intensity

Objective To assess the left ventricular diastolic function in patients with early type 2 diabetes mellitus(T2DM) and combined with hypertension by using wave intensity(WI) and Doppler tissue imaging(DTI).Methods 21 patients with T2DM and 25 T2DM patients with essential hypertension group (T2DM + EH),all of which had left ventricular ejection fraction (LVEF) more than 50％,were enrolled in this study,and 25 healthy volunteers were taken as control group.Each patient underwent conventional echocardiography,DTI and WI exam.Peak velocities of early (E) and late (A) mitral inflow,peak early (Ea) and late (Aa) diastolic velocities of mitral annulus,E/Ea,and deceleration wave intensity (W2) of right common carotid artery(CCA) in three groups were measured and compared.Results Compared with the control group,W2 in T2DM group increased significantly (P ＜0.01),Ea was reduced (P ＜0.05),and E/Ea was significantly increased (P ＜0.01),Ea was significantly reduced (P ＜0.01) and W2 increased significantly(P ＜0.05) in T2DM + EH group.Compared with the T2DM group,E/Ea in T2DM + EH group was increased (P ＜0.05),while W2 and Ea in T2DM + EH group reduced,but no statistically significant differences were found.Conclusions WI combined with DTI may offer a new rapid method in noninvasive and comprehensive evaluation of left ventricular diastolic dysfunction in patients with T2DM and T2DM combined with EH. Effect of down-regulation of crocetin on expression of LOX-1 in atherosclerosis rats

Objective To explore the effects of crocetin on expression of LOX-1 in atherosclerosis rats.Methods Atherosclerosis rats model was established by feeding the animals with a high lipid diet.The aorta and heart were taken for pathological sections.The expression of LOX-1 in aorta of atherosclerosis rats which were affected by crocetin were analyzed by reverse transcription polymerase chain reaction and western blotting.Results Both high-dosage and low-dosage of crocetin could reduce markedly the expression of LOX-1 gene and protein in atherosclerosis rats.Conclusion Crocetin can down-regulate significantly the expression of LOX-1 in atherosclerosis rats.

Angiotensin II deteriorates advanced atherosclerosis by promoting MerTK cleavage and impairing efferocytosis through the AT₁R/ROS/p38 MAPK/ADAM17 pathway

Vulnerable plaques in advanced atherosclerosis have defective efferocytosis. The role of ANG II in the progression of atherosclerosis is not fully understood. Herein, we investigated the effects and the underlying mechanisms of ANG II on macrophage efferocytosis in advanced atherosclerosis. ANG II decreased the surface expression of Mer tyrosine kinase (MerTK) in macrophages through a disintegrin and metalloproteinase17 (ADAM17)-mediated shedding of the soluble form of MerTK (sMer) in the medium, which led to efferocytosis suppression. ANG II-activated ADAM17 required reactive oxygen species (ROS) and p38 MAPK phosphorylation. Selective angiotensin II type 1 receptor (AT 1 R) blocker losartan suppressed ROS production, and ROS scavenger N-acetyl-l-cysteine (NAC) prevented p38 MAPK phosphorylation. In addition, mutant MERTK Δ483-488 was resistant to ANG II-induced MerTK shedding and efferocytosis suppression. The advanced atherosclerosis model that is characterized by larger necrotic cores, and less collagen content was established by feeding apolipoprotein E knockout (ApoE −/− ) mice with a high-fat diet for 16 wk. NAC and losartan oral administration prevented atherosclerotic lesion progression. Meanwhile, the inefficient efferocytosis represented by decreased macrophage-associated apoptotic cells and decreased MerTK + CD68 + double-positive macrophages in advanced atherosclerosis were prevented by losartan and NAC. Additionally, the serum levels of sMer were increased and positively correlated with the upregulated levels of ANG II in acute coronary syndrome (ACS) patients. In conclusion, ANG II promotes MerTK shedding via AT 1 R/ROS/p38 MAPK/ADAM17 pathway in macrophages, which led to defective efferocytosis and atherosclerosis progression. Defining the molecular mechanisms of defective efferocytosis may provide a promising prognosis and therapy for ACS patients.

Clinical effects of perazine ferulate tablets combined with eucalyptol limonene pinene enteric soft capsules for treatment of children with IgA nephropathy

The clinical effects of piperazine ferulate tablets combined with eucalyptol limonene pinene enteric soft capsules for treatment of children with IgA nephropathy were investigated. Sixty children with IgA nephropathy were included in the study and were randomly divided into the control (n=30) and observation (n=30) groups. The patients in the control group were treated with conservative or hormone therapy while patients in the observation group were treated with piperazine ferulate tablets combined with eucalyptol-limonene-pinene enteric soft capsules. Clinical effects were observed and compared. The total effective rate of the observation group was significantly higher than that of the control group, while the incidence of complications was significantly lower than that of the control group (p<0.05). Serum IgA and fibronectin levels of the observation group were significantly lower than those of the control group, while the level of C3 was significantly higher than that of the control group (p<0.05). In conclusion, piperazine ferulate tablets combined with eucalyptus enteric soft capsule constituted a safe and effective for the treatment of children with IgA nephropathy. The treatment was superior to conservative or hormone therapy, and thus worthy of clinical promotion.

Associations of pre-hospital statin treatment with in-hospital outcomes and severity of coronary artery disease in patients with first acute coronary syndrome-findings from the CCC-ACS project

Background The current burden of dyslipidemia, the pre-hospital application of statins and the association of pre-hospital statins with the severity of coronary artery disease (CAD) and in-hospital outcomes in Chinese patients with first acute coronary syndrome (ACS) are very significant and remain unclear. Methods A total of 41,183 patients who underwent coronary angiography and were diagnosed with ACS for the first time from a nationwide registry study (CCC-ACS) were enrolled. The severity of CAD was assessed using the CAD prognostic index (CADPI). The patients were classified into statin and non-statin groups according to their pre-hospital statin treatment status. Clinical characteristics, CADPI and in-hospital outcomes were compared, and a logistic regression analysis was performed to determine whether pre-hospital statin therapy is associated with in-hospital outcomes and CADPI. A sensitivity analysis was used to further explore the issues above. Results The non-statin group had more in-hospital all-cause deaths (1.2 vs. 0.8%, P = 0.010). However, no association exists between statin pretreatment and in-hospital major adverse cardiovascular events (MACEs) or all-cause deaths in the entire population and subgroups (all P > 0.05). Surprisingly, statin pretreatment was associated with an 8.9% higher risk of severely obstructive CAD (CADPI ≥ 37) (OR, 1.089; 95% CI, 1.010–1.175, P = 0.028), and similar results were observed in subgroups of females, those aged 50 to 75 years, and patients with hypertension. Conclusion Statin pretreatment was not related to MACEs or all-cause death during hospital stay, but it was associated with a higher risk of increased angiographic severity in patients with first ACS.

The Clarification to the Misapprehension on General Education.

It's a trend that many countries over the world try their best to enhance General Education in order to actualize educational reform in higher education. This paper clarifies eight misapprehensions about General Education from its meaning, content and the relationship with Specialized Education in the hope of having correct understandings to General Education and making its effect on higher educational reform. At present, those who engage in research or practice about higher education pay much attention on GE. A great number of Higher Education Institutions (HEIs) carried out GE in order to rectify the abuse of SE, and cultivate successors who are well developed morally, intellectually and physically. In fact, they just followed others blindly, even regarded GE as embellishment for SE. The reason is really lacking correct understanding about the base features of GE. To clarify those misapprehensions about GE is helpful to promote the higher educational reform.

Crystal structure of the nucleoid-associated protein Fis (PA4853) fromPseudomonas aeruginosa

Factor for inversion stimulation (Fis) is a versatile bacterial nucleoid-associated protein that can directly bind and bend DNA to influence DNA topology. It also plays crucial roles in regulating bacterial virulence factors and in optimizing bacterial adaptation to various environments. Fis from Pseudomonas aeruginosa (PA4853, referred to as PaFis) has recently been found to be required for virulence by regulating the expression of type III secretion system (T3SS) genes. PaFis can specifically bind to the promoter region of exsA , which functions as a T3SS master regulator, to regulate its expression and plays an essential role in transcription elongation from exsB to exsA . Here, the crystal structure of PaFis, which is composed of a four-helix bundle and forms a homodimer, is reported. PaFis shows remarkable structural similarities to the well studied Escherichia coli Fis (EcFis), including an N-terminal flexible loop and a C-terminal helix–turn–helix (HTH) motif. However, the critical residues for Hin-catalyzed DNA inversion in the N-terminal loop of EcFis are not conserved in PaFis and further studies are required to investigate its exact role. A gel-electrophoresis mobility-shift assay showed that PaFis can efficiently bind to the promoter region of exsA . Structure-based mutagenesis revealed that several conserved basic residues in the HTH motif play essential roles in DNA binding. These structural and biochemical studies may help in understanding the role of PaFis in the regulation of T3SS expression and in virulence.

Characteristics of breast cancer blood supply before and after chemotherapy with low-dose CT perfusion

Objective To analyze the characteristics of breast cancer blood supply before and after chemotherapy with low-dose CT perfnsion. Methods Fifteen patients with breast cancer underwent CT breast perfusion examination, which was performed before and after chemotherapy within 1 week on Siemens Sensation 4 scanner with 120 kV and 50 mAs, 50 ml of nonionic contrast agent (320 mg I/ml) was injected at a flow rate of 4 ml/s with a power injector, Scan started after 8 seconds delay and data acquisition duration was 50 seconds. The blood flow ( BF), blood volume (BV) and mean transfer time (MTT) of lesion and contralateral normal breast gland were calculated using Basama perfusion 3 software package before and after chemotherapy. At the same time, the tumor size before and after chemotherapy were measured and correlated with the BF values. The t test and non-parametric test were used for the statistics. Results ( 1 ) The mean BF、BV and MTT of breast cancer were (33.20±4. 17) ml · min-1 · 100 ml-1 , (8. 31±2.43) ml · 100 ml-1 and ( 15. 31 ± 4. 31 ) s respectively before chemotherapy, and ( 13.65 ± 6. 04) ml · min-1 100 ml-1, (5.04±2. 33) ml · 100 ml-1 and (25. 97±9. 07) s respectively after chemotherapy and there were statistically significant (P =0. 000). The mean BF、BV and MTT of normal breast were (4. 31 ± 2.23) ml · min-1 · 100 ml-1, (1.38±0.75) ml · 100 ml-1 and (19.25±3.94) s respectively before chemotherapy, and (4.03±2.35) ml · min-1 · 100 ml-1、(1.44±0.84) ml · 100 ml-1、(22.56 ± 7.71 ) s respectively after chemotherapy and there were not statistically significant (P >0. 05). (2)The BF of breast cancer was higher than the normal breast before chemotherapy ( P < 0. 01 ). (3) There was a positive correlation between the BF values and tumor size before and after chemotherapy ( r = 0. 902, P = 0. 000). Conclusion The BF value has a positive correlation with tumor size after chemotherapy, CT perfusion is more sensitive for the evaluation of chemotherapy response than morphologic assessment.
