To detect Trichomonas vaginalis (Tv) by direct immunofluorescence assay (DFA) and discuss its clinical significance.The parasites at a concentration of 3 x 10(5) cells/L were fixed by acetone on slides which were then blocked by 1% BSA (bovine serum albumin) or 10% BSA or 10% NCS (newborn calf serum) respectively, incubated with different dilution of polyclonal goat anti-Tv IgG (1:20-1:2,560) for different incubation time (15, 30, 45, 60, 90, 120 min). 120 clinical vaginal specimens were examined by direct immunofluorescence assay, the wet mount method and the in vitro cultivation.Blocked by 1% or 10% BSA, incubated at 37 degrees C for 45 min with a titer 1:160 of polyclonal antibody were the optimal conditions for direct immunofluorescence assay. Its sensitivity and specificity were 87.9% and 98.6% respectively in comparison with the in vitro cultivation method.Direct immunofluorescence assay is a useful alternative to the wet mount method which shows a lower sensitivity.
To evaluate the contraception efficacy, mode of bleeding, side effects and other positive effects of drospirenone-ethinylestradiol (Yasmin) in healthy Chinese women.This was a multicenter, randomized, control study of 768 healthy Chinese women who consulted about contraception. The subjects were randomized into Yasmin group (30 microg ethinylestradiol plus 3 mg drospirenone, 573 cases) or desogestrel group (30 microg ethinylestradiol plus 150 microg desogestrel, 195 cases) with the ratio of 3:1. Each individual was treated for 13 cycles. Further visits were required at cycle 4, cycle 7, cycle 10 and cycle 13 of treatment Weight, height, body mass index were evaluated at each visit. The menstrual distress questionnaire (MDQ) was given to the women at baseline, visit 3 (cycle 7) and visit 5 (after cycle 13).The values of basal features were similar between two groups (P > 0.05). The Pearl index (method failure) of Yasmin was 0. 208/hundred women year which was lower than that of desogestrel (0. 601/hundred women year). The mode of bleeding was similar between two groups after trial without showing any significant difference. According to MDQ subscale, the improvement of water retention and increasing appetite during inter-menstrual period and water retention and general well-being during menstrual period in the Yasmin group (-0.297, -0.057, 0.033, 0.150 respectively) was more obvious than that in the desogestrel group (-0.108, 0.023, 0.231, -0.023 respectively) with a significant difference (P < 0.05). Some other values which improved in both two groups, especially the improvement of breast tenderness and pain and skin abnormality in Yasmin group (18.0%, 89/494; 12.6%, 62/494) was more distinct than that in desogestrel group (11.3%, 19/168; 5.4%, 9/168). The mean weight increased in desogestrel group (0.57 kg) while it decreased in Yasmin group (-0.28 kg) with a significant difference (P < 0.01).Both Yasmin and desogestrel have good efficacy on contraception and similar modes of menstrual bleeding. Yasmin is better than desogestrel in terms of weight control and premenstrual syndrome of oral contraceptive.
To study the effect of aqueous abstract from eucommia ulmoides oliv on the activity of superoxide dismutase (SOD) and alpha-actin expression in the penile tissues of rats with diabetes mellitus (DM) in vitro.A diabetes model was established by administration of alloxun twice to Sprague Dawley (SD) rats. Ten diabetic and 10 normal rats were randomly selected and the penile strips of each rat were divided into four equal shares and cultured in two groups, a eucommia ulmoides oliv coculture group (Group A, further dicided into 1 microg/ml, 10 microg/ml and 100 microg/ml subgroups) and a control group (Group B). Seven days later, the activity of SOD in the culture medium was detected by spectrophotometry, and the levels of micro-actin expression in the penile tissues were examined with the immunohistochemical method.Compared with Group B, the activity of SOD in the culture medium in athe 10 and 100 microg/ml subgroups was notably elevated (P < 0.01), and the numbers of immunoreactive positive cells of alpha-actin in the penile tissues remarkably increased (P < 0.01).The activity of SOD and alpha-actin expression in the penile tissues of diabetic rats in vitro can be increased by eucommia ulmioides oliv.
We have found that Rhynchosia volubilis Lour will suppress the motility of human sperm in virto aggressively at other experiment.In further information of its bacteriostatic action,the bacteriostatic test was done in experimentation with several bacteria including their resistant strains,which is common to find from the genital tract of both genders.It was found that Rhynchosia volubilis Lour is very effective agents against Staphylococcus aureus,Neisseria gonorrhoeae,Escherichia coli,and it has the same effect on their resistant strain.The MIC against Staphylococcus aureus is 31.25 mg/mL,Escherichia coli is 250 mg /mL,Neisseria gonorrhoeae is 31.25 mg/mL.
To assess the association between a 5T polymorphism in intron 8 of cystic fibrosis transmembrane conductance regulator (CFTR) gene and congenital bilateral absence of vas deferens (CBAVD) in Han Chinese males.Genomic DNA from 33 individuals with CBAVD and 99 azoospermic males with CBAVD were recruited. The 5T polymorphism was detected with PCR, TA cloned and sequenced.CFTR gene mutations were identified in 17 (51.5%) of patients with CBAVD. In 3 patients (17.6%), the mutations were identified on both alleles. Nine CFTR gene mutations (9.1%) were detected in 99 azoospermic patients, for whom none had mutations on both alleles.This study has confirmed molecular heterogeneity of CFTR mutations in CBAVD. For CBAVD patients without 5T mutations, other changes may be found in the same gene.
Objective:To study changes of the sex hormone levels after MCMV infection on male mice,to reveal the possible mechanisms of MCMV-induced reproductive handicap.Methods:Following establishment of murine testicular MCMV infection model,experimental and control groups were set.In different periods of MCMV infection(within one seminiferous cycle),the sex hormone levels in murine sera and testicular tissue was detected by using radioimmunoassay to study the regulatory effects of hypothalamus-pituitary-testis axis on the reproductive function in different periods after MCMV infection as viewed from reproductive biology.Results:In the testicular homogenate,testosterone(T) concentration in the experimental group was obviously decreased on D4 to D9 PI(P0.05),transiently restored on D14 to D21 PI,and on D38 PI,it was decreased as compared with that in the control group(P0.05).Serum luteinizing hormone(LH) in the experimental group was significantly increased on D4,D9 and D14 PI(P0.05),but there was no significant difference on the other time points between the experimental groups and control group.Serum T and follicle stimulating hormone(FSH) showed no significant difference among the experimental groups.Conclusion:MCMV acute infection could result in the changes of the sex hormone levels in male mice by influencing the regulatory of hypothalamus-pituitary-testis axis to affect the reproductive function of the mice.
