Chitosan: A Potential Biopolymer in Drug Delivery and Biomedical Applications

Chitosan, a biocompatible and biodegradable polysaccharide derived from chitin, has surfaced as a material of promise for drug delivery and biomedical applications. Different chitin and chitosan extraction techniques can produce materials with unique properties, which can be further modified to enhance their bioactivities. Chitosan-based drug delivery systems have been developed for various routes of administration, including oral, ophthalmic, transdermal, nasal, and vaginal, allowing for targeted and sustained release of drugs. Additionally, chitosan has been used in numerous biomedical applications, such as bone regeneration, cartilage tissue regeneration, cardiac tissue regeneration, corneal regeneration, periodontal tissue regeneration, and wound healing. Moreover, chitosan has also been utilized in gene delivery, bioimaging, vaccination, and cosmeceutical applications. Modified chitosan derivatives have been developed to improve their biocompatibility and enhance their properties, resulting in innovative materials with promising potentials in various biomedical applications. This article summarizes the recent findings on chitosan and its application in drug delivery and biomedical science.

Development and In-vitro Evaluation of Dexamethasone Enriched Nanoemulsion for Ophthalmic Indication

Background: Dexamethasone (DEX) is a glucocorticosteroid used in the treatment of steroid-responsive inflammatory conditions of the eye. The currently marketed formulations pose several issues, like poor drug residence time, resulting in frequent administration of the formulation, making them less effective. Objective: The present study aims to provide comprehensive data encompassing the designing, op-timization, development, and characterization of DEX nanoemulsion (DEX NE) for treating in-flammatory conditions of the anterior segment of the eye by employing the Quality by Design (QbD) approach. Methods: A Plackett-Burman Design (PBD) was employed to screen seven independent variables, such as oil concentration, surfactant concentration, polymer concentration, homogenization speed and time, microfluidization pressure and cycles, and their influence on critical quality attributes (CQAs), such as globule size, zeta potential, and viscosity, was evaluated. Furthermore, the Box-Behnken design (BBD) was employed for optimization, and design space was generated to obtain the optimized DEX NE. Results: The experimental results after DEX NE characterization reveal a globule size of 181 ± 90 nm with a zeta potential of -21.03 ± 1.68 mV and a viscosity of 19.99 cp. Furthermore, the drug re-lease study of simulated tear fluid demonstrated prolonged and steady release for up to 48 hr. Cyto-toxicity assay of DEX NE exhibited good cell viability. Conclusion: All these findings pave the way for a better understanding of developing a robust, safe, and non-toxic formulation for ocular drug delivery.

Decoding Artificial Intelligence in Neuroscience: Applications Beyond Diagnosis

Background: Artificial Intelligence has witnessed exponential expansion in health care applications. The article pronounces the dynamic excellence AI is achieving in the healthcare discipline of neuroscience. Objective: The paper highlights basic concepts of AI and acmes the interdisciplinary collaboration of Computational neuroscience, Cognitive science, and AI. Also, the article draws out important findings related to AI in neuroscience amongst its diverse application in the various disciplines. An ephemeral overview of applications of AI-based constructs in neurological disorders namely Neuroinflammation, Schizophrenia, Parkinson’s disease, Epilepsy, Autism spectrum disorder, Alzheimer’s disease, Brain tumor and Anesthesiology has been demonstrated in the present work. Methods: The method includes the collection of data from different search engines like google Scholar, PubMed, ScienceDirect, SciFinder, etc. to get coverage of relevant literature for accumulating appropriate information regarding AI, neuroscience, and their linkages. Results: These considerations are made to expand the existing literature on the progressing role of AI in the management of neurological disorders. Conclusion: The exponential expansion in the development of AI-based systems might aid in addressing the prevailing limitations in the domain of neurological disorders and neuroscience.

Peptide Functionalised Nanocarriers for Bone Specific Delivery of PTH (1-34) in Osteoporosis

: Osteoporosis represents a major public health burden especially considering the aging population worldwide. Treatment modalities for osteoporosis are classified into two categories based on the effect on bone remodelling: anabolic drugs and antiresorptive drugs. Anabolic drugs are preferred as it stimulates new bone formation. Currently, PTH (1-34) is the only peptide-based drug approved as an anabolic agent for the treatment of osteoporosis by both USFDA as well as EMA. However, its non-specific delivery results in prolonged kidney exposure, causing hypercalcemia. Nanotechnology-based drug delivery systems functionalized by conjugating it with homing moieties, such as peptides, offer an advantage of targeted delivery with reduced off-target effects. Here, we propose an innovative and targeted nanovesicle approach to efficiently deliver PTH (1-34) to the bone surface using peptides as a homing moiety. The proposed innovative delivery approach will augment the specific interaction between the drug and bone surface without producing side effects. This will reduce the off-target effects of PTH (1-34), and at the same time, it will also improve the outcome of anabolic therapy. Therefore, we postulate that the proposed innovative drug delivery approach for PTH (1-34) will establish as a promising therapy for osteoporotic patients, specifically in postmenopausal women who are at greater risk of bone fracture.

Dexamethasone Loaded Electrospun Nanocomposite Ocular Insert: In-vitro Drug Release and Mechanical Assessment

Background: Dexamethasone is currently available as suspension for the treatment of anterior ocular inflammatory diseases that are given through eye drops. Upon topical delivery by eye-drops, less than 5% of the drug gets absorbed when applied topically as eye drops. The major portion of administered drug either comes out from the eye or gets subjected to nasolacrimal drainage resulting in poor bioavailability. Objective: The present study is aimed at developing polymeric ocular insert containing dexamethasone as a drug by employing the use of electrospinning technique. Methods: Dexamethasone (0.1% w/v) loaded electrospun sheet was also made using 10% w/v EC: HPMC: PEO (60:20:20) in solvent system of ethanol: water (90:10) at optimized electrospinning parameters of 12kV with a flow rate of 0.8mL/hr and distance of 20 cm between tip and collector. Results: The prepared nanocomposite insert was characterized for DSC and FTIR-ATR spectroscopy revealing no physical-chemical interaction between drug with polymers. The thickness of the electrospun sheet was found to be 270±0.02 μm and % drug content was found to be 0.43±0.01% w/w. The release profile showed that around 95% drug was released in 48 hrs. This release profile showed the prepared drug-loaded electrospun ocular insert was best suitable for once-a-day delivery. Assessment of mechanical properties like young’s modulus, tensile strength, and % elongation showed that the prepared insert can be handled easily without any breakage or damage. Conclusion: Upon delivery in conjunctival sac, this is best suitable for once-a-day delivery. The developed electrospun insert, consisting of a unique polymer composite of biodegradable polymers, avails the benefits of nanofibers imparting prolonged release and this novel formulation overcomes the limitations of conventional therapies. This reduces the dosing frequency and improves patient compliance.

Quantification of Anti-Osteoporotic Anabolic Peptide in Stealth Lipid Nanovesicles Through Validated RP-HPLC Method

Teriparatide is a recombinant fragment of human parathyroid hormone, a potent osteoanabolic agent used for osteoporosis.The present study endeavored to develop a simple, rapid, and reliable reverse phase-high performance liquid chromatography (RP-HPLC) method for the determination of teriparatide in pegylated lipid nanovesicles for rapid formulation development/optimization.A rapid RP-HPLC-based analytical method was developed for the quantification of teriparatide in pegylated lipid nanovesicles. The method was optimized on a Waters XBridge C18 (4.6 × 150 mm, 10 μm) column with a mobile phase consisting of 0.1% formic acid in water and acetonitrile both in a linear gradient program. In the method, a short run time of 9 min was achieved at a flow rate of 1.0 mL/min with an injection volume of 50 µL at a detection wavelength of 210 nm. The developed method was validated according to the ICH Q2 (R2) guideline. The method was applied for the quantification of teriparatide in prepared pegylated lipid nanovesicles. Teriparatide encapsulated pegylated lipid nanovesicles were prepared by the ethanol injection method. Further, these vesicles were characterized for % entrapment efficiency (%EE), particle size, zeta potential, and morphology by Cryo-SEM.The teriparatide was eluting at 4.8 min in the run. Further, for the method validation, the linear relationship between concentration and response was established over the concentration range of 50-250 µg/mL with the R2 > 0.999. The method sensitivity was shown with LOD and LOQ with the value of 100 ng/mL and 500 ng/mL, respectively. The method was found to be accurate and precise with the recovery ranging in 100 ± 2% and RSD <2%, respectively. Minor deliberate changes proved the robustness of the developed method.These results indicated that the developed and validated method is accurate, precise, rapid, reliable, and fit for the quantification of teriparatide in different formulations.The RP-HPLC method was developed and validated for the quantification of teriparatide from novel pegylated lipid nanovesicles.
