[Morphological characteristics and immunophenotype of dendritic cells generated from monocytes of human peripheral blood].

Morphological features and immunophenotype of mature dendritic cells (DC) generated from the monocytes of healthy donor blood after culture with GM-CSF and IL-4 with the addition of TNF, were studied using light and electron microscopy, as well as flow-cytometry. It was shown that DC were characterized by a number of morphological features such as: large size, eccentrically located nucleus, highly developed system of extensions, large vacuolated cytoplasm, prominent and activated Golgi complex and ribosomal apparatus. Mature DC are characterized by active surface expression of MHC and co-stimulatory molecules (MHC I, MHC II, CD40, CD80, CD86).

[Influence of dendrite cells, generated with the use of immunomodulators of microbial origin, on the proliferative and cytotoxic activity of lymphocytes].

The influence of ripe dendrite cells (DC) on the proliferative activity of mononuclear leukocytes and the cytotoxic activity of lymphocytes of syngenic mice was studied. As inducers of DC ripening, the combination of antigenic components incorporated into the vaccine lmmunovac Bh-4, (Klebsiella pneumoniae, Proteus vulgaris, Escherichia coli, Staphylococcus aureus), as well as K. pneumoniae LPS and TNF-alpha, were used. This study demonstrated that DC activated with these preparations enhanced the proliferative activity of mononuclear leukocytes, the activity of Bh-4 being higher than that of K. pneumoniae LPS. The increase of proliferative activity was accompanied by a rise in the cytotovicity of mouse lymphocytes with respect to the NK-sensitive tumor line YAC-1 or Ehrlich tumor cells. The incubation of the lymphocytes with ripe DC (with the use of the preparations under study as ripening inducers), loaded with tumor antigens, made it possible to obtain cytotoxic lymphocytes having high cytotoxic activity with respect, mainly, to those tumor lines from which lysates for the treatment of DC had been produced. The activation of DC with bacterial immunomodulators led to an increase in the antigen-presenting function of these cells, to their higher capacity for regulating the differentiation of mononuclear leukocytes and for activating the cytotoxicity of natural killers.

[Generation of dendrite cells and the use of immunomodulators of bacterial origin as cell differentiation inducers].

The generation of ripe dendrite cells (DC) of marrow origin was obtained with the use of the vaccine Immunovac-BN-4, an immunomodulator of microbial origin, as well as Klebsiella pneumoniae LPS and TNF-alpha, as ripening inducers. These inducers equally led to the ripening of DC. The generation of ripe DC was characterized by morphological, phenotypical and functional changes. The immunophenotype of cells altered from CD34+, CD38-, CD40-, CD80-, CD86-, MHC I-, MHC II-, F4/80- to CD34-, CD38+, CD40+, CD80+, MHC I+, MHC II+, F4/ 80(low). In parallel with the ripening of DC their phagocytic activity decreased. In culture medium with ripe DC the levels of such cytokines as IL-1b, IL-6, IL-12, IFN-gamma, TNF-alpha significantly increased and the production of IL-4 decreased. The content of IL-2 and IL-10 remained unchanged.

Analysis of the changes of stromal precursor cell numbers in the thymus and the spleen of animals of different age groups

The objective of this study was to analyze the species differences in the numbers of stromal precursor cell (CFU-f), their cloning efficiency (CFE-0 and their dynamics in different organs during aging, using the mathematical gradient decrease method. Age changes of CFU-f numbers and of their CFE-f were studied in the thymus and the spleen of mice and guinea pigs. The study was performed using CFU-f cloning in monolayer cultures. CFU-f numbers and CFE-f were found to decrease with aging both in the thymus and the spleen of mice and guinea pigs. However these changes were different in each species and were variable in different organs of the animals of the same species, which, probably was associated with the physiological characteristics and aging peculiarities of the animals of different species and with the functional role of organs studied. The process of reduction was more significant in the thymus of guinea pigs and mice - the numbers of CFU-f were decreased 75- and 12-fold, respectively. Since it is known that the population of CFU-f in the thymus and the spleen includes inducible osteogenic precursor cells, the data obtained indicate the possibility of a reduction in numbers of this category ofstromal precursors, that could be one of the reasons of osteoporosis of aging. The application of a mathematical analysis using the gradient decrease method allows to predict the time-course of age changes and to evaluate the dynamics of CFU-f numbers and of CFE-f in association with organism aging.

[Comparative characteristic of the methods of immunocytochemistry and immunomagnetic separation for detection of cytokeratin-positive cells in bone marrow and lymph nodes].

Comparative study of cytokeratin-positive (CK+) cells, isolated from bone marrow and lymph node micrometastases in patients with lung and esophageal cancer, was performed using the methods of immunocytochemistry and immunomagnetic separation. It was found that immunocytochemical analysis permitted the detection of CK+ cells in the smears of bone marrow of oncologic patients. All the samples of bone marrow with immunocytochemically demonstrated CK+ cells, contained also the cellular complexes marked by magnetic spheres. The method of positive immunomagnetic separation has some advantages for the intraoperative diagnosis. However, when either method was applied, among CK+ cells in bone marrow and lymph nodes some false-positive, questionable cellular forms and hemopoietic cells expressing epithelial markers were found. Theses results suggest that for verification of micrometastases in these organs, an additional study using the standard staining of the slides, is required.

[Age changes in the numbers of stromal precursor cells in the animal bone marrow].

Age changes of stromal precursor cell (CFU-f) numbers and their cloning efficiency (CFE-f) in monolayer cultures were evaluated using the mathematical gradient decrease method in the bone marrow of mice, guinea pigs and rats. It was found that aging was accompanied by a decline of CFE-f and CFU-f numbers in all animal species studied. However, these changes were variable in different species, that could possibly be explained by species-related physiological characteristics and aging peculiarities. Since the population of bone marrow CFU-f is known to include committed osteogenic precursor cells, these findings indicate the possibility of age-related decline in the numbers of this cell type, which could be one of the reasons of osteoporosis of aging. The application of a mathematical gradient decrease method allows to predict the course of age changes and to evaluate the dynamics of CFU-f numbers and of CFE-f in association with organism aging.

[Effect of nanodispersed manganese oxide (III, IV) on morphological properties of various tissues and body systems].

The study of morphological features of tissue organs of experimental animals after a single intragastric administration of an aqueous suspension nanodis-dispersed manganese (III, IV) at doses of 2000, 3500 and 5000 mg/kg showed complex of morphological changes in the form of circulatory disorders up to hemostasis and bleeding which lead to changes in the structure of organs, enhanced apoptosis, activation of macrophage system by proliferation and macrophage phagocytosis of degradation products of cells, hypertrophy of lymphoid tissue of immune system; development of histiocytic infiltrates in parenchymal organs and central nervous system. With the introduction of microsized analogues in the same doses of the morphological changes in the circulatory system, macrophage system and immune organsfound predominantly in a dose of 5000 mg/kg. Changes were less significant and were presented as activation of macrophages in the liver in the form of activation of Kupffer cells, proliferation of lymphoid tissue in the spleen, lymph histiocytic infiltrates in the liver, kidney, medium size.

[Morphological and functional characteristics of lymphokine-activated killer cells generated from the mononuclear cells of human peripheral blood].

The aim of this research was to study the morphological, functional and immunophenotypical characteristics of lymphokine-activated killer cells (LAKC) generated from the mononuclear cells (MNC) of healthy donors' peripheral blood at different time intervals after the cultivation with interleukin-2 (IL-2). LAKC had the appearance of large lymphoid cells of prolymphocyte and immunoblast type with highly pyroninophilic cytoplasm and electrone-microscopic features indicative of synthetic activity. LAKC were shown to intensely express activation antigens and adhesion molecules on their surface and to posess high cytotoxic potential in respect to tumor cells. Time-course of LAKC surface antigen expression corresponded to the changes of a proportion of activated cellular forms, generated from MNC of healthy donors' peripheral blood by incubation with IL-2. On the basis of these experimental findings, the usage of 3-5-day culture of LAKC could be recommended for the immunotherapy of malignant tumors.

[Influence of various bacterial ligand application methods on cytokine expression].

Study the production of cytokines in mice during vaccination with polycomponent Immunovac-VP-4 vaccine containing TLR ligands with various administration methods.Immunovac-VP-4 was administered to mice by subcutaneous, intranasal or per oral methods. The preparation was administered nasally at a single dose of 500 microg in the volume of 30 microl. Per oral single dose was 2000 microg in the volume of 0.5 ml. 200 microg of the preparation was administered subcutaneously. Cytokines in blood sera were determined by EIA 8 hours after the administration of the vaccine.In mice 8 hours afterthe single administration of Immunovac-VP-4 the levels of IL-1beta, IL-6, IL- 12, IL-5 increased significantly. However their concentration differed depending on the method of administration. The most active expression of cytokines was observed during subcutaneous administration. The indexes of cytokine expression were significantly higher (p < 0.05) than during non-parenteral administration methods.Mucosal application methods along with parenteral were established to be able to activate effector mechanisms of immune repose with its consequent polarization by Th1/Th2 pathways. These mechanisms lay the groundwork for development of antigen-specific immune responses against antigens/pathogens.

[Characterization of lymphokine-activated killer cells obtained from the natural killers--T cells of patients with hepatic tumor lesion].

To analyze the functions of lymphokine-activated killer cells (LAKC) and to study their phenotype are of paramount importance in the assessment of their role in antitumor immunity. The natural killers--hepatic T cells--have been shown to be used to prepare LAKS that have a higher cytotoxic activity than LAKS that are generalized from peripheral mononuclear cells.
