Live vaccines induce both cellular and humoral immunity and are cheap and easy to use. The induction of immunity is provided through the reproduction of the vaccine strain in the host body without the development of the disease, since the bacterium to which it is necessary to induce the immunity is characterized by reduced virulence (attenuation). The first generation of attenuated strains was chosen from a variety of spontaneous or physically, chemically and biologically induced mutants after virulence assessment. The rapid development of molecular genetics makes it possible to significantly reduce the time of pathogen attenuation via obtaining knockout mutants with genes selected by a researcher or by inserting “avirulence genes” into the genome. But, given that the methodological aspects of the design of avirulent strains are basically clarified, the absence of officially established criteria for assessing the hazard in regulatory documents hinders the determination of the degree of attenuation. In this regard, there is a need for changes in the procedure for accounting and storage of bacterial cultures, as well as regulation of the process of transferring plague pathogen avirulent strains from the 1 st into the 3 rd pathogenicity group for subsequent use in the vaccine preparations development. Thereat, the requirements to methodological aspects of the safe generation of attenuated Yersinia pestis strains and the criteria for testing the virulence loss should be maintained at high levels

Analysis of Implementation of the Requirements for Provision of Biological Safety at a Potentially Hazardous Facility

Discussed are the problems of implementation of requirements for the provision of biological safety at a potentially hazardous biological facility by the example of Federal Budgetary Institution of Science “State Scientific Center of Applied Microbiology and Biotechnology” of the Rospotrebnadzor. Identified are the necessary and sufficient organizational-andpreventive and control measures and procedures on management of works with microorganisms of the I-IV groups of pathogenicity (hazard) in isolated labs of the Center. Issues associated with exploitation of biological safety engineering systems and assessment of their protective efficiency, problems of medical support of the activities and response to the emergency situations of biological character with the involvement of doctors from a specialized medical wing of the Federal Medical-Biological Agency of the Russian Federation, serving the facility on a constant basis, are analyzed. Determined have been approaches to and principles of establishing the requirements for the system of biological safety at a potentially hazardous biological facility taking into account all the range of biological risks when working with microorganisms. Complex approach and realization of appropriate necessary measures in the sphere of biological safety provision, creating an environment for the reduction of biological risks up to acceptable level while working with pathogenic microorganisms, can be an effective solution of the problem on the whole.

Water cleaning of the fire box screens of the PK-14-2 boiler with combustion of shredded peat

Combustion of shredded peat in the PK-14-2 boiler of the Yaroslav HPP-1 is accompanied by intensive slagging of the furnace screens, with the formation of connected tough deposits, which leads to slagging of the panel steam superheater and the convective steam superheater, limitations on the boiler load and to a decrease in its technico-economic indicators. The AVOD-1BL system for cleaning the fire box using long-range water spraying equipment is an effective means of removing connected and friable deposits forming with combustion of shredded peat. Considering the high concentration of calcium oxide in the ash of shredded peat, the results of studies of slagging and cleaning of fire box screens of the PK-14-2 boiler of the Yaroslav HPP-1 might be of interest in planning boiler units for Berezov coal, and also in operation of boilers operating on Kansko-Achin basin coals.

The system of monitoring combined extract and input ventilation during organization of works with pathogenic biological agents (PBA) is necessary for ensuring protection of the personnel and environment from harmful emissions. Stable ensuring optimum physical and hygienic air environment parameters (microclimate) using ventilation in PBA laboratories is an additional part of management of possible risks in the laboratories. An effective mode of safe aeration of microbiological laboratory is possible only by combination of hygienic requirements and parameters of thermal balance of the air environment taking into account thermo insulating characteristics of overalls, means of individual protection, including respiratory organs, specificity and intensity of the performed works with PBA. Use of biological safety boxes creates additional protection against harmful emissions in the laboratories and environment.
