Enteroaggregative Escherichia coli (EAEC) is emerging as a significant diarrheal pathogen in multiple population groups. Like any other enteropathogen, EAEC may encounter life-threatening levels of inorganic acids (H+) during their natural route of infection. In this study, we showed that EAEC orchestrates acid tolerance by modulating the levels of acid-induced outer membrane proteins (OMP). EAEC (T8) was grown in vitro by mimicking in vivo pH conditions of both in stomach and intestine. The lowest pH where EAEC showed growth was 4.0 in Luria Bertani (LB) media, and surprisingly during log phase two de novo OMP of sizes 41 kDa and 48 kDa were exclusively observed at this pH. Further, acid-induced proteins at pH 4.0 were identified by 2D gel electrophoresis and Matrix Assisted Laser Desorption Ionization-Time of Flight (MALDI-TOF). The role of the most notable acid/pH response regulator rpoS in ASP expression was determined by gene knockout in the clinical strain EAEC (T8). Mutation of rpoS gene suppressed growth and downregulated the expression of ASP in EAEC (T8) at pH 4.0 confirming probable role of rpoS in EAEC.

Tuberculosis remains one of the most dreaded infectious diseases notwithstanding the availability of a number of anti-tuberculosis drugs. The recent rise of multidrug-resistant tuberculosis and its association with HIV infection poses a challenging health concern. Therefore, there exists a pressing requirement to identify novel drug targets and develop new anti-tuberculosis drugs that will be effective against multidrug-resistant-tuberculosis. Shikimate kinase is a novel and attractive drug target as it is vital for the survival of Mycobacterium tuberculosis but is absent in mammals. Hence, inhibitors designed against shikimate kinase will be specific to the pathogen and be least harmful to the host. Till date, no drug candidates are available against this target. The crystal structure of Mycobacterium tuberculosis shikimate kinase complexed with shikimate has been used to identify a dipeptide inhibitor using in silico structure-based design approach. The designed peptidic inhibitor has a predicted binding affinity of 5.5 nm which is 8000 times better than substrate shikimate and 10 times greater than the best suggested inhibitor. It is potent in both the known open and closed LID conformations of target protein. As small peptides are known to be non-toxic, this inhibitor could be a lead compound for development of specific anti-tuberculosis drugs.

A 14-month-old infant had a classical Wilms' tumor of the left kidney and subsequently developed a tumor showing predominantly mature mesenchymal elements and lack of cellular atypia in the right kidney. His father had classical Wilms' tumor of the left kidney. The various possibilities regarding the precise nature and mechanism of the development of the tumor in the right kidney are discussed. The similarities and the differences between this tumor and congenital mesoblastic nephroma are noted. The possible relation of chemotherapy to mesenchymal differentiation and maturation in Wilms' tumor is considered. The possibility of an unusual, histologically benign, mesenchymal variant in the spectrum of infantile nephroblastic neoplasia is also suggested.
