Design of Bio-Graphene-Based Multifunctional Nanocomposites Exhibits Intracellular Drug Delivery in Cervical Cancer Treatment

The advent of bio-nanotechnology has revolutionized nanodrug delivery by improving drug efficacy and safety. Nevertheless, acceptable carriers for therapeutic molecules are one of the most difficult challenges in drug delivery. Graphene material-based (GMB) and polymer-based drug-loaded nanocarriers have both demonstrated clinical advantages in delivering drugs of interest in vitro/in vivo. Cisplatin (CDDP) is an inorganic chemotherapeutic drug that is commonly used to treat a variety of cancers. However, its clinical use is associated with drug resistance and few side effects, which reduces its antitumor effects. Therefore, we developed a CDDP-loaded chitosan-functionalized graphene oxide nanocomposite (CDDP@CS-GO NC)-based nanodrug delivery system (NDDS). Flow cytometry and confocal imaging show that the CDDP@CS-GO NCs lead to significantly increased intracellular drug accumulation in tumor cells. Cancer cells take up the nanocomposite via endocytosis and can generate intracellular reactive oxygen species (ROS) to increase mitochondrial membrane potential loss (Δψm) and enable cytochrome-c release, followed by the dysregulation of Bcl-2 into the cytosol and activation of caspase-3 to induce cancer cell apoptosis. In vitro experiments demonstrated the excellent cancer therapeutic effect with few side effects of the carriers. CDDP@CS-GO NCs are expected to play an important role in responsive NDDSs for cancer therapy.

Studies On the Kinetics of Oil Oxidation Using Benzoyl Peroxide and Its Synergistic Effect in Fish Oil Tanning

The oxidation of oil is an unavoidable process during oil tanning which aids in the forming of aldehydes that subsequently interact and react with amino groups in collagen. Oxidizing agents are commonly used to catalyze a faster reaction, thereby reducing days required for oil tanning. Fish oils are widely and regularly used for oil tanning. The oxidation of fish oil depends on its content of unsaturated fatty acids. In the current research, we focused on the kinetics of fish oil oxidation using Benzoyl Peroxide [BPO]. To assess the catalytic behavior of BPO, the kinetic study of the oil oxidation was carried out by determining the peroxide and p-Anisidine values. The maximum peroxide and p-Anisidine values were obtained on the 4th day. This is in accordance with our earlier studies.

Colon cancer (CRC) is a serious health problem throughout the world. Development of novel drugs without side effects for this cancer is crucial. Luteolin (LUT), a bioflavonoid, has many beneficial effects such as antioxidant, anti-inflammatory and anti-proliferative potential. was a potent chemical carcinogen used for the induction of colon cancer. Colon carcinogenesis was initiated by intraperitoneal injection of azoxymethane (AOM) to mice at the dose of 15 mg/body kg weight in Balb/C mice for 3 weeks. Mice were treated with LUT at the dose of 1.2 mg/body kg weight orally. Mitochondrial enzymes such as isocitrate dehydrogenase (ICDH), ${\alpha}$-keto dehydrogenase (${\alpha}$-KDH), succinate dehydrogenase (SDH) and the activities of respiratory chain enzymes NADH dehydrogenase and cytochrome c oxidase were found to be elevated in AOM-treated animals. Treatment with LUT decreased the activities of all the parameters significantly. Hence, LUT might be a potent anticancer agent against colorectal cancer.

An Update on the Effect of Natural Products on Oncogenic Transcription Factors in Triple Negative Breast Cancer

Breast cancer is the most aggressive and rapidly growing cancer worldwide. It is classified into several subtypes among which triple negative breast cancer (TNBC) is the most aggressive. Oncogene regulation leads to increased signal activity in the cells and contributes to the tumorigenic phenotype. Such signals generate a large number of proteins that provoke cell growth and inhibit cell death. Transcription factors, such as miR-27a, NF-κB, and FOXM1, are proteins that are conserved across species, abundantly found in the nucleus, bind to DNA, and enforce specific target genes. Oncogenic transcription factors arising from mutation accompanying aberrant gene expression transfer signals to the nucleus and interrupts transcription patterns and stimulation of oncogenic transcription factors consistent with cell cycle regulation, apoptosis, proliferation, and cell differentiation. This review gives a detailed framework about the treatment opportunities and highlights the importance of few transcription factors which may act as an important biomarker specifically for TNBC.
