Photo-Fenton process of oxidative destruction of rice husk alkaline hydrolysates lignin

Abstract The present work studies the destruction of a silica-free solution of alkaline hydrolysate of rice husk (pH 7.25, COD = 22.8O g·L−1) using the photo-Fenton process in the UV layer at a wavelength of 365 nm. Oxidation of lignin was carried out at a constant mass ratio of COD:H2O2 = 1:2 in the solutions with different dilution times (1:10, 1:50) and with varying iron(II) concentrations in the range from 10 to 100 mg·L−1. It was established that the oxidative degradation of alkaline lignin effectively proceeds in the UV layer when the solution is diluted 50 times, at an iron(II) concentration from 50 to 70 mg·L−1. Color, the content of polyphenols and chemical oxygen demand in the silica-free solution with a dilution ratio of 1:50, after oxidation, is reduced by several tens of times. The products of lignin degradation were identified using gas chromatography.

Coral Holobionts Possess Distinct Lipid Profiles That May Be Shaped by Symbiodiniaceae Taxonomy

Symbiotic relationships are very important for corals. Abiotic stressors cause the acclimatization of cell membranes in symbionts, which possess different membrane acclimatization strategies. Membrane stability is determined by a unique lipid composition and, thus, the profile of thylakoid lipids can depend on coral symbiont species. We have analyzed and compared thylakoid lipidomes (mono- and digalactosyldiacylglycerols (MGDG and DGDG), sulfoquinovosyldiacylglycerols (SQDG), and phosphatidylglycerols (PG)) of crude extracts from symbiotic reef-building coral Acropora sp., the hydrocoral Millepora platyphylla, and the octocoral Sinularia flexibilis. S. flexibilis crude extracts were characterized by a very high SQDG/PG ratio, a DGDG/MGDG ratio < 1, a lower degree of galactolipid unsaturation, a higher content of SQDG with polyunsaturated fatty acids, and a thinner thylakoid membrane which may be explained by the presence of thermosensitive dinoflagellates Cladocopium C3. In contrast, crude extracts of M. platyphylla and Acropora sp. exhibited the lipidome features of thermotolerant Symbiodiniaceae. M. platyphylla and Acropora sp. colonies contained Cladocopium C3u and Cladocopium C71/C71a symbionts, respectively, and their lipidome profiles showed features that indicate thermotolerance. We suggest that an association with symbionts that exhibit the thermotolerant thylakoid lipidome features, combined with a high Symbiodiniaceae diversity, may facilitate further acclimatization/adaptation of M. platyphylla and Acropora sp. holobionts in the South China Sea.

Features of interaction between selective lipid metabolites and immune system signaling molecules in patients with asthma

Introduction. Lipid metabolism is a key component in many pathophysiological processes, and its disruption can play a significant role in the development of chronic inflammation in asthma. Aim. To determine the nature of the interaction between fatty acids (FAs) and their derivatives with cytokine parameters of the immune system and their contribution to systemic inflammation in patients with asthma. Materials and methods. The spectrum of FAs in the plasma of patients with asthma was analyzed using gas chromatography-mass spectrometry. Levels of endogenous fatty acid ethanolamides (NAEs) were measured using high-performance liquid chromatography with mass spectrometry. Cytokine levels were determined by enzyme-linked immunosorbent assay (ELISA). The degree of interaction between the parameters was assessed using systemic analysis based on the integral coupling index (D). Results. It was established that the immune system response was most strongly associated with the relative content of n-6 polyunsaturated fatty acids. Modification of the FA composition was most significantly linked with interleukins (IL) 17A, 10, 4, and 6. Endogenous NAEs—arachidonoylethanolamide (AEA, 20:4n6) and docosahexaenoylethanolamide (DHEA, 22:6n3)—showed significant involvement in cytokine regulation in mild asthma. NAE 20:4n6 had the strongest association with IL-17A, interferon-γ, tumor necrosis factor (TNF)-α, and IL-2; NAE 22:6n3 was associated with IL-17A, IL-6, and TNF-α. Conclusion. The study established the contribution of disturbances in trigger parameters of lipid metabolism to systemic inflammation. Modification of FA composition and disruption of the synthesis of their mediators lead to dysregulation of the cytokine network of the immune system, which may contribute to the development and chronicity of systemic inflammatory reactions in patients with asthma.

Neuroinflammation and adult hippocampal neurogenesis in neuropathic pain and alkyl�glycerol ethers treatment in aged mice

Neuropathic pain is a condition characterized by unpleasant sensory and emotional experiences associated with a number of diseases or injuries affecting the sensory system through various mechanisms. In this study, we focused on the impact of chronic neuropathic pain on the microglial state and hippocampal neurogenesis in aged mice. In addition, we examined the effects of alkyl glycerol ethers (AGE) treatment on behavioral parameters, hippocampal neuronal and microglial plasticity in aged C57BL/6 mice with neuropathic pain. For the induction of neuropathic pain, we used the model of chronic constriction injury (CCI) of the sciatic nerve. We observed painful behavior in animals subjected to CCI, expressed as a decrease in locomotor activity and the development of cold allodynia. A violation of working and long‑term memory was also observed. AGE administration reduced the severity of cold allodynia and prevented memory impairment. In addition to behavioral changes, neuropathic pain was accompanied by microglial activation, changes in the hippocampal production of pro‑ and anti‑inflammatory cytokines, as well as a decrease in neurogenesis. The administration of AGE prevented the neuropathic pain‑derived effects, including M1 microglial activation and neurogenesis disruption. However, in vitro experiments demonstrated the pro‑inflammatory activation of microglial cells, emphasizing the complexity of the mechanisms underlying the pharmacological effects of AGE. On the whole, the findings of this study demonstrate that AGE treatment prevented behavioral effects of neuropathic pain in mice, and AGE may thus have potential for use in the prevention or treatment of neuropathic pain cognitive and emotional effects. However, as the mechanisms underlying this type of pain are complex, further studies are required to determine the detailed pharmacological effects of AGE.

The Phospholipid Molecular Species Profile of Apostichopus japonicus Tissues Modifies through Exposure to n-3 Polyunsaturated Fatty Acid-Deficient Diet

The sea cucumber Apostichopus japonicus, being a target species of commercial fisheries and aquaculture, is also used as a source of biologically active compounds with high pharmacological potential. By the methods of high-performance liquid chromatography with high resolution mass spectrometry, we analyzed the major structural phospholipids (PL)-glycerophosphoethanolamines (PE), glycerophosphocholines (PC), glycerophosphoserines (PS), and glycerophosphoinositols (PI)-in tissues of wild and cultured sea cucumbers. The intestines of the wild and cultured animals differed from the other tissues by an elevated content of molecular species of PE, PC, and PS with 22:6n-3 fatty acid. The respiratory trees of the studied animals contained a high level of odd-chain PI and PI with 20:4n-6. The exposure to n-3 PUFA-deficient diet resulted in substantial changes in the molecular species profile of PL of the wild and cultured animals. The cultured sea cucumbers showed a significant decrease in the 20:5n-3 content in all four studied PL classes. A replacement of 20:5n-3 by 20:4n-6 occurred in PE, PC, and PI. The decrease in the level of molecular species of PS with 20:5n-3 was compensated by an increase in the level of monounsaturated long-chain PS. The diet of cultured sea cucumbers is a crucial factor for enhancing the nutritional properties of the product obtained from them.

1-O-alkylglycerols (AKG) are a class of natural ether lipids derived from 1-O-alkyl-2,3-diacyl-sn-glycerols by deacylation. In this study, 1-O-alkylglycerol (AKG) composition was investigated in the hepatopancreas lipids of the crab Paralithodes camtschaticus and the liver lipids of the squid Berryteuthis magister and the skate Bathyraja parmifera. One of the principal AKG in marine organisms was 1-O-hexadecyl-sn-glycerol (AKG 16:0). To assess AKG influence on melanoma, we evaluated the cytotoxicity and antiproliferative actions of natural AKG 16:0 and synthetic 1-O-octyl-sn-glycerol (AKG 8:0) on three human melanoma cell lines SK-Mel-5, SK-Mel-28, and RPMI-7951. Natural AKG 16:0 in concentration up to 20 µM was not toxic to all cell lines. AKG 8:0 showed no toxicity to cells SK-Mel-5 and SK-Mel-28 in concentrations up to 20 µM but had moderate cytotoxicity to RPMI-7951 cells with an IC50 of 13 µM. Both investigated substances inhibited the proliferation, formation, and growth of cell colonies of RPMI-7951. PRACTICAL APPLICATIONS: AKG exhibit a variety of biological activities, including anticancer effects. In this study, the liver lipids of the skate B. parmifera and the hepatopancreas lipids of crab P. camtschaticus were shown to be sources of AKG. Our data showed that AKG can be used to prevent the formation of new colonies of malignant cells in combination therapy against melanoma. The results will be useful for future studies involving marine ether lipids and the examination of their anticancer properties against malignant cells.
