Development and validation of Cellular Senescence-Related Gene Signature for predicting the survival and immunotherapeutic responses in Skin Cutaneous Melanoma

Abstract Background Accumulating evidence has suggested the impact of cellular senescence on tumorigenesis, development, and immune modulation in cancers. However, the prognostic value of cellular senescence-related genes (SRGs) and their association with immunotherapy response remain unexplored in skin cutaneous melanoma (SKCM) patients. Methods In this study, we explored the expression profiles of 279 SRGs in 469 SKCM patients included from TCGA database. The univariate and least absolute shrinkage and selection operator (LASSO) were conducted to construct a cellular senescence-related signature (SRS), and Kaplan–Meier survival curves as well as ROC curve were used to validate the predictive capability. The GSE65904 dataset was further used to validate the predictive ability of prognostic signature. Moreover, we explored the correlations of the SRS with tumor-infiltrating immune cells and response to immunotherapy. The expression levels of prognosis related SRGs were validated based on immunohistochemistry. In addition, consensus clustering analysis was performed to stratify SKCM patients into different clusters and compared them in OS. Results We developed a prognostic prediction SRS for patients with SKCM and verified patients in low-risk group were associated with better prognosis. Moreover, the correlation analysis showed that the SRS could predict the infiltration of immune cells and immune status of the immune microenvironment in SKCM, and patients with low-risk score might benefit from immunotherapy. In addition, all the SKCM patients in this study were classified into three clusters based on the mRNA expression profiles of 113 SRGs, which revealed that cluster 1 suffered poorer outcomes relative to clusters 2 and 3. Conclusions The SRS developed in this study could be used as a prediction tool in survival assessment and immunotherapy for SKCM patients.

BRD7 expression and c-Myc activation forms a double-negative feedback loop that controls the cell proliferation and tumor growth of nasopharyngeal carcinoma by targeting oncogenic miR-141

miR-141 is up-regulated and plays crucial roles in nasopharyngeal carcinoma (NPC). However, the molecular mechanism underlying the dysregulation of miR-141 is still obscure. Thus, the ChIP-PCR was performed to identify the c-Myc-binding sites in miR-141 and BRD7. qRT-PCR, western blot and immunohistochemistry assays were used to detect the expression of miR-141 and its up/down stream molecules. The rescue experiments on the c-Myc/miR-141 axis were performed in vitro and in vivo. Our results showed that the levels of mature miR-141, pre-miR-141 and pri-miR-141 were downregulated in c-Myc knockdown NPC cells. Meanwhile, c-Myc transactivates the expression of miR-141 by binding its promoter region. Moreover, BRD7 was identified as a co-factor of c-Myc to negatively regulate the activation of c-Myc/miR-141 axis, as well as a direct target of c-Myc. Moreover, restoration of miR-141 in c-Myc knockdown NPC cells notably rescued the effect of c-Myc on cell proliferation and tumor growth, as well as the blocking of PTEN/AKT pathway. Additionally, the expression of c-Myc was positively correlated with that of miR-141 and the clinical stages of NPC patients and negatively associated with the expression of BRD7. Our findings demonstrated that BRD7 expression and c-Myc activation forms a negative feedback loop to control the cell proliferation and tumor growth by targeting miR-141. These observations provide new mechanistic insights into the dysregulation of miR-141 expression and a promising therapeutic option for NPC.

Deciphering the vital roles and mechanism of m5C modification on RNA in cancers.

5-methylcytosine (m5C modification) plays an essential role in tumors, which affects different types of RNA, the expression of downstream target genes, and downstream pathways, thus participating in the tumor process. However, the effect of m5C modification on RNA in tumors and the exact mechanism have not been systematically reviewed. Therefore, we reviewed the status and sites of m5C modification, as well as the expression pattern and biological functions of m5C regulators in tumors, and further summarized the effects and regulation mechanism of m5C modification on messenger RNA (mRNA), ribosomal RNA (rRNA), transfer RNA (tRNA), long non-coding RNA (lncRNA) and other RNA in tumors. Finally, we summed up the interaction network, potential application, and value in clinical diagnosis and treatment of tumors. Taken together, this review benefits revealing the mechanism of m5C modification in tumor progression and provides new strategies for tumor diagnosis and treatment.

Effect of a 60-day weight reduction intervention prior to IVF/ICSI on perinatal outcomes in overweight or obese infertile women

Abstract Purpose: The aim of this study was to determine whether a 60-day weight reduction intervention prior to in vitro fertilization/intracytoplasmic sperm injection(IVF/ICSI) and a higher weight reduction ratio effectively improved perinatal outcomes for infertile overweight or obese women. Methods: This was a retrospective cohort study conducted at a university-affiliated fertility center.Two thousand three hundred and eighty-one overweight or obese infertile women who underwent or did not undergo a 60-day weight reduction intervention prior to IVF/ICSI between February 27, 2017 and November 11, 2020 were included in our study. All of these women achieved clinical pregnancies and delivered a single child after assisted pregnancy therapy.Primary outcomes included neonatal birth weight and the incidence of premature delivery, low birth weight, and pregnancy complications. Results: Birth weight was significantly higher in the intervention group (3519.6 ± 484.8 g) than the control group (3406.8 ± 554.2 g; P < 0.001). There was no significant difference in the incidence of pregnancy complications between the two groups. As the weight loss ratio increased, the incidence of low birth weight, premature delivery, stillbirth, hypertensive disorders of pregnancy, and premature membrane rupture showed a downward trend. Linear regression analysis found that the weight-loss intervention prior to IVF/ICSI and lower HOMA-IR at ovulation induction wereassociated with increased birth weight. Conclusions: A 60-day weight reduction intervention prior to IVF/ICSI may increase neonatalbirth weight, reduce blood glucose concentration, and improve insulin resistance in infertile overweight or obese women. Several perinatal outcomes may be improved by an increased weight reduction ratio.

Estimation of Transmission Input&Output Shaft Torque and Drive Wheel Speed for Compound Power Split Powertrain Based on Unknown Input Observer

Precise estimation of transmission input&output shaft torque and wheel speed is significant to the development of torque distribution and coordinated control method for the power split hybrid powertrain system. This paper studies an estimation method of transmission input and output torque as well as drive wheel speed using an unknown input observer in different operating modes for a compound power split hybrid powertrain system. Firstly, the configuration and dynamic characteristic of power split transmission is analyzed. Then, state equations in different operating modes are established, and a Takagi-Sugeno (T-S) fuzzy model is utilized to deal with the nonlinearities of load resistance torque. Furthermore, an unknown input reduced-order observer is designed to estimate the transmission input&output shaft torque and wheel speed. The asymptotical stability of the developed observer is verified based on Lyapunov theory. Finally, the performance of the unknown input observer is assessed by comparing with the Luenberger observer based on Matlab/Simulink simulation platform and test bench. Simulation and experiment results show that, the proposed observer can estimate the transmission input&output shaft torque and wheel speed more effectively in different operating modes.

Introducing High Density of Very Active Sites and Stepwise Postmodification for Tailoring the Porosity of Highly Demanding Cr³⁺-Based Metal–Organic Frameworks

Chromium(III)-based metal–organic frameworks (Cr-MOFs) are highly robust and porous and have been very attractive in a wide range of investigations. However, the harsh direct synthetic conditions not only impede the synthesis of new Cr-MOFs but also restrict the introduction of functional groups into them. Postsynthetic modification has somewhat alleviated such difficulties; nevertheless, it still suffered from procedures that are tedious and conditions that are not mild, which often result in low concentration of the functional groups introduced. To overcome these shortcomings, here, in this paper, we supplied a new route and prepared a benzyl alcohol functionalized Cr-SXU-2 from the judiciously designed benzyl alcohol functionalized Fe-SXU-2 through solvent-assisted metal metathesis strategy. The functionalized Cr-SXU-2 shows well-preserved crystallinity, porosity, and high chemical stability. The benzyl alcohol group can be converted into a very active benzyl bromide group in an almost quantitative yield and thus for the first time produce the benzyl bromide functionalized MOF, Cr-SXU-2-Br, in which the −Br group can be exchanged by a nucleophilic group. As a proof of concept, −N3 was introduced and transformed into other active sites via "click reaction" to further tailor the interior of Cr-SXU-2. All these functionalized Cr-MOFs showed improved adsorption performance in contrast to the nonfunctionalized one. This step-by-step postmodification process not only diversifies the functionalization of robust MOFs but also opens a new route to employ many different functional groups in the demanding highly stable Cr-MOF platforms.
