Safety Assessment and Probiotic Evaluation of Enterococcus Faecium YF5 Isolated from Sourdough

Abstract Enterococcus faecium YF5, a strain previously isolated from sourdough, was assessed for safety and probiotic potential. Its virulence and antibiotic resistant phenotypes (cytolysin and gelatinase production, antibiotic susceptibility) and genes ( cylA , gelE , ace , agg , esp , and vanA ) were surveyed. Results indicated that the tested virulence determinants were nontoxic. In addition, E. faecium YF5 was sensitive to 3 antibiotics such as amoxicillin, vancomycin, and chloramphenicol. Furthermore, results of in vivo animal acute oral toxicity of E. faecium YF5 studies were similar to the control group that indicated no abnormalities. In addition, E. faecium YF5 stably survived in low pH, bile salts, gastric, and intestinal fluids in vitro . Moreover, E. faecium YF5 was found to adhere to human colon cancer cell line HT‐29 at 3.39 (±0.67) × 10 5 CFU/mL. When cocultured with pathogenic organisms ( Enterobacter sakazakii CMCC45402, Escherichia coli CMCC44102, enterohemorrhage Escherichia coli O157: H7 CMCC44828, Salmonella Typhimurium CMCC50071, Shigella flexneri 301, and Shigella sonnei ATCC 29930) and 2 gram‐positive strains ( Listeria monocytogenes CMCC54001 and Staphylococcus aureus CMCC 26003), it inhibited these foodborne pathogens with exception of S. aureus . Therefore, E. faecium YF5 can be regarded as a safe strain and it may be used as a probiotic preparation or for microecologics.

Quantum dots-based system for the detection of bacteria in drinking water

Our paper presents a new development of quantum dot (QD)-based semi-automatic system for the detection of pathogenic bacteria in drinking water. It combined QDs imaging technique (with advantages of higher fluorescence intensity and longer fluorescent signal, multiple color choices, tolerant to reaction environment, et al.) with a fast filtration system, and enabled the semi-automatic detection process. Compared with the traditional culturing-based methods, this system requires less detection time and labor power for quantitative and rapid detection of the pathogenic bacteria. In this study, we spiked Escherichia coli O157:H7 in autoclaved drinking water and used the solution as an example to demonstrate the ability and feasibility of the designed system. The preliminary experimental results indicated that the proposed system is practicable with a very short detection time within 2 h and the detection limit can be as low as single bacterial cell in 1 mL of drinking water.

Previous results showed that overexpression of the WTH3 gene in multidrug resistance (MDR) cells reduced MDR1 gene expression and converted their resistance to sensitivity to various anticancer drugs. The WTH3 gene promoter was found to be differentially regulated in paired MDR vs non-MDR MCF7 cells owing to epigenetic modifications and transcription factor modulations. To understand further the mechanisms that govern WTH3's differential expression, we uncovered a p53-binding site in its promoter, which indicated that WTH3 could be regulated by the p53 gene. This hypothesis was then tested by different strategies. The resulting data revealed that (1) the WTH3 promoter was upregulated by the p53 transgene in diverse host cells; (2) there was a correlation between WTH3 expression levels and p53 gene status in a cell line panel; (3) a WTH3 promoter region was directly targeted by the p53 protein in vitro and in vivo. In addition, overexpression of the WTH3 gene promoted the apoptotic phenotype in host cells. On the basis of these findings, we believe that the negative role played by the WTH3 gene in MDR development is through its proapoptotic potential that is regulated by multiple mechanisms at the transcription level, and one of these mechanisms is linked to the p53 gene.
